Extracellulära blåsor (EVs) är lipid lipidens vesikler utsöndras av celler och deras storlek varierar mellan 30 och 300 nm. EVs rapporterades först 1978, med belägg från blåsor av patienter med Hodgkins sjukdom1. Dessa blåsor rapporterades vara 40 till 120 nanometer i storlek. 1980 rapporterades det att EVs är inblandade i koagulationssystemet och trombos, bildandet av en blodpropp inuti ett blodkärl i cancer patienter2. Efter 30 år, har EVs rapporterats vara en viktig faktor att främja tumör invasion, immun fly och angiogenes3. Intresset för exosomes som en biomarkör för tidig diagnos ökar snabbt, och rening och karakterisering av exosomes måste vara i sällskap med molekylära avbildningstekniker. Exosomes är nano-storlek extracellulära vesikler utsöndras av kroppsvätskor och är kända som representerar egenskaperna hos celler som utsöndrar dem. Innehåll och morfologi av de utsöndrade blåsor återspeglar cell beteende eller fysiologiska status, till exempel celltillväxt, migration, klyvning och döden. Den exosomes' roll kan bero mycket på storlek och storleken på exosomes varierar från 30 till 300 nm. Den mest använda metoden för exosome imaging är negativ färgning, medan andra resultat baseras på Cryo-transmissionselektronmikroskopi, Scanning Electron Microscopy och Atomic Force Microscopy. Den typiska exosome morfologi bedömas genom negativ färgning är en cup-form, men ytterligare detaljer är ännu inte klart. En exosome välkarakteriserad genom strukturella studier är nödvändiga särskilt i medicinska och farmaceutiska områdena. Funktion beroende av morfologi bör därför kontrolleras av elektronmikroskopi tekniker såsom märkning ett specifikt protein i de detaljerade strukturen exosome. För att observera detaljerade struktur, jämfördes ultratunna sektionerad bilder och negativa färgade bilder av exosomes. I detta protokoll föreslår vi transmissionselektronmikroskopi för bildtagning av exosomes inklusive negativ färgning, hela mount immuno-färgning, block förberedelse, tunna avsnitt och immuno-guld märkning.

För närvarande indelas exosomes i storlek och form kategorier av transmissionselektronmikroskopi. Figur 2 visar negativt färgade exosome och immun-märkt exosome i hela mount status.
