Hier, wij laten zien dat humanin peptide-gemedieerde ERK1/2 activering treedt op in twee verschillende soorten cellen, en de subcellular localisatie van geactiveerde ERK1/2 verschillen naargelang de omstandigheden (bijvoorbeeld, dosis van peptide, tijdstip en cel kunnen type). Het is aangetoond dat humanin signalen door middel van twee verschillende receptoren22,23, wat de verschillen verklaart wellicht in de signalering tussen de twee cellijnen, evenals...

Nieuwe studies tonen aan dat mitochondriale afkomstige peptiden (MDP's) spelen een belangrijke rol in cytoprotection en metabolisme1,2,3. Inzicht in het signaaltransductie traject in aanwezigheid van MDP's geeft ons inzicht in het mechanisme waarmee MDP's verschillende functies moduleren. De eerste geïdentificeerde MDP, humanin, heeft aangetoond dat het verhogen van extracellulaire signaal-gereglementeerde kinase 1/2 (ERK1/2) fosforylering door haar receptor binden4,5. De downstream effect van ERK1/2 activering is echter nog steeds underexplored.
De ERK1/2 cascade fungeert als een essentiële bemiddelaar in een verscheidenheid van cellulaire processen zoals proliferatie, cel migratie, cellulaire metabolisme, overleving en apoptosis6,7,8. Om te orkestreren van al zijn deze verschillende cellulaire processen, de activiteit en subcellular localisatie van ERK1/2 strak geregeld phosphatases en steiger eiwitten9,10. Naast posttranslationele modificatie regelt de dynamische pendelen van ERK1/2 ook haar signalering functie, activiteit en specificiteit11,12. ERK1/2 is vooral gelokaliseerd in het cytosol13. Een set van verankering en steiger eiwitten helpen behouden ERK1/2 in cytoskeletal elementen, op de oppervlakte van organellen of diffuus in het cytoplasma13. Bij stimulatie, ERK1/2 is phosphorylated en wordt losgekoppeld van de verankerende eiwitten, waardoor de translocatie van ERK1/2 om andere subcellular compartimenten, met inbegrip van de celkern, mitochondriën, Golgi, en lysosomen14, 15 , 16.
Hoewel humanin is bekend dat de ERK1/2 traject signalering activeren, wordt de activering van ERK1/2 alleen waargenomen in de cel Totaal lysates. Zoals eerder beschreven aangezien ERK1/2 subcellular localisatie een cruciale rol in de downstream effecten, de analyse van zowel de subcellular localisatie en de totale hoeveelheid speelt is gefosforyleerd ERK1/2 nodig om een grondig inzicht van humanin-geïnduceerde ERK1/2 activering en het activeren van downstream doelstellingen.
Om te begrijpen de organellen van de doelstelling van geactiveerde ERK1/2, werd subcellular fractionering, gevolgd door het westelijke bevlekken voor gefosforyleerd ERK1/2 uitgevoerd. Deze methode kan gemakkelijk worden uitgevoerd, zoals het maakt gebruik van Standaard laboratoriumapparatuur en reagentia. De geïsoleerde subcellular compartimenten zijn van hoge zuiverheid, waardoor de resultaten ronduit worden geïnterpreteerd. Immunokleuring van ERK1/2 kan vergelijkbare resultaten geven. Bepaalde subcellular compartimenten zijn echter relatief moeilijk te visualiseren en vereisen speciale fixatie en permeabilization methoden. ERK1/2 niveaus variëren in subcellular compartimenten, en deze variatie kan valse positieve en valse negatieve signalen wanneer we kijken naar hele cel lysates. Daarom geeft een immunoblot met behulp van geïsoleerde subcellular compartimenten ons een beter begrip van ERK1/2 lokalisatie.
De veelzijdigheid van de methode maakt het mogelijk wijzigingen van het protocol bij het onderzoeken van de effecten van andere stimulerende middelen met inbegrip van andere MDP's of translocatie van andere signalering moleculen zoals STAT3. Onlangs worden ontdekte kleine humanin-achtige peptiden (SHLPs) gecodeerd uit 16S rRNA regio waar humanin is gecodeerd, en ze hebben vergelijkbare, maar verschillende eigenschappen vergeleken met HN17. Bijvoorbeeld, SHLP2 en SHLP3 activeren ERK1/2 na 8u hoewel humanin ERK1/2 binnen 5 min. onderzoeken subcellular localisatie van ERK1/2 reactie op verschillende peptiden activeert ons een beter begrip van de biologie van deze peptiden geeft. Opkomende bewijsmateriaal bleek dat de subcellular localisatie van signalering van moleculen een cruciale rol in de downstream effecten speelt. Bijvoorbeeld, STAT3 traditioneel bekend is vooral gelokaliseerd in het cytosol in rustende cellen, en vervolgens het translocates in de kern te activeren van genexpressie in reactie op de cytokines18. STAT3 ook translocates naar de mitochondriën en regelt de TCA cyclus en de ATP productie19. Met betrekking tot autophagy verordening moduleert verschillende subcellular localisatie van STAT3 autophagy in verschillende manieren20. Bijvoorbeeld, nucleaire STAT3 transcriptionally regelt autophagy-gerelateerde genen en fungeert als een autophagy modulator. Autophagy remt cytoplasmatische STAT3 constitutively door interactie met autophagy signalering van moleculen. Mitochondriale STAT3 remt en voorkomt mitophagy door het onderdrukken van oxidatieve stress geïnduceerde autophagy. Daarom is deze methode van de isolatie subcellular compartiment is cruciaal voor het begrip van de rol van andere signalering moleculen, alsmede ERK1/2.

Mitochondriale afkomstige peptiden (MDP's) zijn een nieuwe klasse van peptiden die zijn gecodeerd door kleine open lezing frames binnen andere bekende genen van het mitochondriaal genoom. MDP's hebben een breed scala aan biologische effecten zoals neuronen beschermt tegen apoptosis, verbetering van de metabole markeringen en bescherming van cellen tegen chemotherapie. Humanin was de eerste MDP om ontdekt te worden en is de meest bestudeerde peptide onder het MDP-familie. De membraan-receptoren en de stroomafwaartse signaalroutes van humanin zorgvuldig gekenmerkt. Extra MDP's zoals MOTS-c en SHLP1-6 hebben meer recentelijk ontdekt en de signalering mechanismen nog moeten worden opgehelderd. Hier beschrijven we een cel cultuur gebaseerde methode om te bepalen van de functie van deze peptiden. In het bijzonder, toestaan cel fractionering technieken in combinatie met het westelijke bevlekken voor de kwantitatieve bepaling van activering en translocatie van belangrijke signalering molecuul. Hoewel er andere methoden van cel fractionering, is de hier beschreven een gemakkelijke en eenvoudige methode. Deze methoden kunnen verder het ophelderen van het werkingsmechanisme van deze peptides en andere therapeutische agenten worden gebruikt.
