Så vidt vi ved, er det første gang, at en detaljeret procedure med henblik på at studere effekten af naturligt uran på knogle resorbing celler er beskrevet. Denne tilgang vil være nyttigt at opnå en bedre forståelse af uran indvirkning på knogle fysiologi og kan give en interessant ny værktøj til screening af uran chelatorer. Derudover mener vi, at protokollen beskrevet her kunne anvendes til at undersøge virkningen af andre tungmetaller på osteoclatogenesis.

Uran er en naturligt forekommende radioaktivt element findes i jord, luft og vand; som sådan, er dyr og mennesker udsat for naturligt uran i deres kost. Ud over naturlige kilder stammer uran fra menneskeskabte aktiviteter, som øger sin overflod i miljøet. Uran udgør både kemiske og radiologiske farer. Men fordi naturligt uran, (som er en isotopisk blanding indeholdende 99.27% 238U, 0,72% 235U og 0,006% 234U) har en lav specifik aktivitet (25.103 Bq.g-1), dens virkninger på sundhed tilskrives dens kemiske toksicitet.
Uanset hvad dens indfaldsvej (indånding, indtagelse eller dermal eksponering), de fleste af uran ind i kroppen er elimineret med afføring og kun en lille del når systemisk cirkulationen. Ca. 67% af uran i blodet filtreres igen af nyrer og forlader kroppen i urinen inden for 24 h1. Resten er for det meste deponeres i nyrer og knogler, de to vigtigste målorganer uran toksicitet2,3,4. Fordi skelettet er blevet identificeret som den primære lokalitet af uran langsigtede fastholdelse2,3,4,5,6, flere undersøgelser er blevet gennemført for at udforske den effekt af uran på knogle fysiologi7.
Bone er en mineraliseret væv, der er løbende ombygget i hele dens levetid. Knogle remodellering er en kompleks proces, der afhænger af specialiserede celletyper og består hovedsageligt af to faser: resorption af de allerede eksisterende gamle matrix af osteoklasterne efterfulgt af de novo knogle konstruktion af osteoblaster. Osteoklaster er store multinuclear celler som følge af en sammenlægning af forløber celler hæmatopoietisk oprindelsesregler, migrere til resorption steder hvor de tillægger knogle8. Deres tilknytning opstår samtidigt med en omfattende omorganisering af deres cytoskeleton9. Denne omlægning er nødvendig for oprettelsen af en isoleret rum mellem cellen og knoglen overfladen hvor osteoklast udskiller protoner, fører til opløsning af hydroxyapatit og proteaser involveret i nedbrydningen af den organiske matrix. De resulterende nedbrydningsprodukter er endocytosed, transporteres gennem celle til området membran over knoglen overfladen og udskilles, en proces kaldet transcytosis10,11.
Resultater fra i vivo og in vitro- undersøgelser viser, at uran hæmmer knogledannelse og ændrer antallet og aktivitet af osteoblaster7,12. Derimod har effekter af uran på knogleresorption og osteoklaster været dårligt udforskede. Flere i vivo undersøgelser har rapporteret en forøgelse af knogleresorption efter administration af uranyl nitrat til mus eller rotter13,14. Derudover foreslog en epidemiologisk undersøgelse, at stigningen i uran indtag via drikkevand tendens til at være forbundet med en stigning i serum niveau af en knogle resorption markør i mænd15. Taget sammen, disse resultater førte til den konklusion, at uran, som ophobes i knogler kunne fremme knogleresorption. De cellulære mekanismer involveret i denne potentielle effekt af uran er dog stadig et åbent spørgsmål. Derfor besluttede vi at undersøge virkningen af uran på opførsel af resorbing knogleceller.
Her, vi beskrive den protokol, vi har etableret at karakterisere og kvantificere effekterne af naturligt uran på pre osteoklaster levedygtighed og osteoklaster differentiering og resorptive aktivitet. Eksperimenter beskrevet heri har gjort med den RAW 264.7 murine transformerede makrofag cellelinie, som let kan differentiere i osteoklaster når kulturperler i overværelse af cytokin RANKL for 4 eller 5 dage, og som er klassisk bruges til at undersøge osteoklast differentiering og funktion16. Udviklet fremgangsmåder er pålidelige, giver meget reproducerbare resultater og er fuldt ud gældende for primære osteoklaster. Alle disse grunde mener vi, at denne metode er nyttig for at få en bedre forståelse af de molekylære mekanismer involveret i uran toksicitet i knoglen. Vi mener desuden, at denne tilgang kunne tilpasses som en screeningsværktøj til at identificere nye uran chelatdannere.

Uran har vist sig at blande sig med knogle fysiologi og det er velkendt, at denne metal ophobes i knogler. Dog er lidt kendt om effekten af naturligt uran på opførsel af knogleceller. Især er virkningen af uran på osteoklaster, de celler, der er ansvarlig for resorption af knoglematrix, ikke dokumenteret. For at undersøge dette spørgsmål, har vi etableret en ny protokol bruger uranyl acetat som en kilde til naturligt uran og RAW 264.7 murine cellelinie som en model af osteoklast prækursorer. Heri, detaljerede vi alle assays til at teste uran cytotoksicitet på osteoklast prækursorer og til at vurdere dens indvirkning på osteoclastogenesis og den resorbing funktion af modne osteoklaster. Betingelserne, vi har udviklet, navnlig forberedelsen af uranyl-holdige dyrkningsmedier og for såning af RAW 264.7 celler tillade for at opnå pålidelig og yderst reproduktive resultater. Derudover har vi optimeret brug af softwareværktøjer til at lette analysen af forskellige parametre som størrelsen af osteoklasterne eller procentdelen af resorberet matrix.

Tartrat-resistente sur fosfatase farvning blev brugt til at visualisere osteoklaster som store lilla celler med 3 eller flere kerner. Repræsentative billeder af osteoklasterne fremstillet af RAW 264.7 celler dyrkes i overværelse af RANKL og uranyl ioner er vist i figur 1. Ændringer i antallet og størrelsen af osteoklasterne svar på uran er synligt i sammensatte billeder af hele brønde og forstørrede billeder.
