Så vidt vi vet, er dette første gang at en detaljert prosedyre å studere effekten av uran på bein resorbing celler er beskrevet. Denne tilnærmingen vil være nyttig å oppnå en bedre forståelse av uran innvirkning på bein fysiologi og oppgir et interessant nytt verktøy for screening av uran chelater. Videre tror vi at protokollen beskrevet her kan brukes for å studere virkningen av andre tungmetaller på osteoclatogenesis.

Uran er et naturlig forekommende radioaktivt element finnes i jord, luft og vann. som sådan, er dyr og mennesker utsatt for uran i sine dietter. I tillegg til naturlige kilder kommer uran fra menneskelige aktiviteter, noe som øker sin overflod i miljøet. Uran utgjør både kjemiske og radiologiske farer. Men fordi uran (som er en isotopanrikning blanding med 99.27% 238U, 0.72% 235U og 0.006% 234U) har en lav bestemt aktivitet (25.103 Bq.g-1), er dens virkninger på helse knyttet til sin kjemiske giftighet.
Hva oppføring ruten (innånding, inntak eller dermal eksponering), mest uran inn i kroppen er eliminert med avføring og bare en liten del når systemisk sirkulasjon. Omtrent 67% av uran i blodet filtreres igjen av nyrene og forlater kroppen i urin innen 24 h1. Resten er stort sett satt i nyrer og bein, to hovedmål organer uran toksisitet2,3,4. Fordi skjelettet har blitt identifisert som det primære området uran langsiktig oppbevaring2,3,4,5,6, flere studier har vært gjennomført for å utforske den effekten av uran på bein fysiologi7.
Bone er en mineralholdig vev som er kontinuerlig remodeled gjennom hele levetiden. Ubalanse er en komplisert prosess som er avhengig av spesialiserte celletyper og består hovedsakelig av to faser: benresorpsjon av eksisterende gamle matrix av osteoklast etterfulgt av de novo bein konstruksjon av osteoblasts. Osteoklast er store multinuclear celler fra fusjon av precursor celler blodkreft opprinnelsesland overføre benresorpsjon områder hvor de legger til bein8. Deres vedlegg oppstår samtidig med en omfattende omorganisering av deres cytoskjelett9. Denne omorganiseringen er nødvendig for etablering av en isolert rom mellom cellen og bein overflaten som osteoclast utskiller protoner, førte til oppløsningen av hydroxyapatite og proteaser involvert i nedbrytning av det organisk matrise. De resulterende nedbrytningsprodukter er endocytosed, transporteres gjennom cellen membran området motsatt bein overflaten og utskilles, en prosess som kalles transcytosis10,11.
Resultater fra vivo og i vitro studier viser at uran hemmer bein dannelse og endrer nummeret og aktiviteten til osteoblasts7,12. Derimot har effekten av uran på benresorpsjon og osteoklast blitt dårlig utforsket. Flere i vivo studier har rapportert en forbedring av benresorpsjon etter administrasjon av uranyl nitrat mus eller rotter13,14. Videre foreslo en epidemiologiske undersøkelse at økningen i uran inntak via drikkevann tendens til å være forbundet med en økning i serum nivået av en bein benresorpsjon markør i menn15. Sammen disse funnene førte til konklusjonen at uran, som akkumuleres i bein kan fremme benresorpsjon. Imidlertid fortsatt de cellulære mekanismene involvert i potensielle effekten av uran et åpent spørsmål. Av denne grunn besluttet vi å undersøke virkningen av uran på oppførselen til resorbing beinet cellene.
Her beskriver vi protokollen har vi etablert å beskrive og kvantifisere effekten av uran på pre osteoklast levedyktighet og osteoklast differensiering og resorptive aktivitet. Eksperimentene beskrevet her har blitt gjort med rå 264.7 murint transformert macrophage cellen linje, som lett kan skille ut osteoklast når kultivert i nærvær av cytokin RANKL for 4 eller 5 dager, og som brukes til å studere klassisk osteoclast differensiering og funksjonen16. Prosedyrene utviklet er pålitelig, gir svært reproduserbar resultater og er fullt gjelder primære osteoklast. For alle disse grunner tror vi at denne metoden er nyttig for å få en bedre forståelse av molekylære mekanismer involvert i uran toksisitet i bein. Dessuten, vi tror at denne tilnærmingen kan være som en screening verktøyet for å identifisere nye uran chelaterande agenter.

Uran har blitt vist å påvirke bein fysiologi og det er godt etablert at dette metallet akkumuleres i bein. Men er lite kjent om effekten av uran på oppførselen til beinet cellene. Spesielt er virkningen av uran på osteoklast celler ansvarlig for benresorpsjon av bein matrix, ikke dokumentert. For å undersøke dette problemet, har vi etablert en ny protokoll med uranyl acetate som en kilde til uran og murine rå 264.7 celle linje som modell av osteoclast prekursorer. Her, detaljerte vi alle analyser nødvendig å teste uran cytotoksisitet på osteoclast prekursorer og vurdere virkningen på osteoclastogenesis og resorbing funksjonen av eldre osteoklast.
