I denne protokol, er blevet udviklet en ny teknik til oprettelse af NK/T cellelinjer fra fuldblod af CAEBV patienter. Sammenlignet med de eksisterende metoder, er den største fordel ved denne metode dets enkelhed og sit krav om kun en lille mængde blod, mens det udviser en høj succesrate og gode betingelser for cellernes levedygtighed. NK/T cellelinjer kan desuden etableres ved dyrkning PBMC uden fastsættelse af celletyper EBV latent inficerede på forhånd som bestemmelse ville forbruge mere blod og tid før dyrknin...

Epstein - Barr virus (EBV) er allestedsnærværende og inficerer ikke kun B-celler, men også T og natural killer (NK) celler, der forårsager en række EBV-associerede NK/T Lymfoproliferative sygdomme (LPD) og lymfom/leukæmi, såsom EBV-associerede hemophagocytic lymphohistiocytosis, hydroa vacciniforme-lignende lymfom, extranodal T-NK-celle lymfom, nasal type og aggressive NK celle leukæmi1,2,3. Blandt disse er svær kronisk aktiv EBV (SCAEBV) sygdom, som er hændelse hovedsagelig i Østasien, og der nu betragtes som værende en LPD forårsaget af klonal ekspansion af EBV-inficerede T eller NK celler4,5,6, 7, men uden den tilsyneladende immundefekt præsentere i infektiøs mononukleose (IM)-som symptomer, herunder feber, hepatosplenomegaly, lymfadenopati og leverdysfunktion vedvarende eller regelmæssige vareleverancer, samt høj EBV-DNA belastning i den perifert blod8,9. Patienter med CAEBV har en dårlig prognose10,11, og dens patogenese og EBV rolle er uklart. Derfor, cellelinjer afledt af EBV-associerede NK/T Lymfoproliferative sygdomme og lymfomer er meget nyttige som cell modeller for afklaring mekanisme af EBV induceret NK eller T-celle spredning og sine forbindelser med høj forekomst af leukæmi eller lymfom.
Til dato har adskillige cellelinjer etableret med forskellige teknikker12,13,14,15,16. En menneskelig NK cellelinie, NK-YS, blev etableret fra NK celle lymfom/leukæmi, co dyrkning med en mus stromale cellelinie som feeder, og rhIL-2 i en koncentration på 20U pr. mL15. KAI3 var en anden NK cellelinie etableret fra patienter med en svær myg allergi eller SCAEBV med autolog lymphoblastoid cellelinje (LCL), B-celler forvandlet af EBV, som feeder cellerne og tilsætning af rhIL-2 på 100U/mL16. SNK6 og SNT8 var afledt af tumor væv af nasal NK/T celle lymfom patienter ved at tilføje højdosis rhIL-2 (700U/mL)12. Med lignende teknik var cellen SNK-1 fra CAEBV patienter, kulturperler fra PBMC ved fjernelse af T-celler og tilføje 700U/mL af rhIL-213,17. SNT13 og SNT15 blev etableret ved at fjerne CD4 + og CD8 + celler18. Hidtil, blev andre T-celle og NK cellelinjer fra EBV-NK/T LPD patienter alle udviklet med denne metode19.
Ulemperne ved de eksisterende metoder nævnt ovenfor omfatter beskæftigelse af feeder celler, kravet om en høj-dosis IL-2, udnyttelse af så meget som 10 mL fuldblod eller rensning af NK/T-celler, som er meget udfordrende i klinikken grund nødvendigheden af at kontrollere, hvilke typer af celle at EBV latent inficerer før begyndelsen til kultur. Som CAEBV primært opstår hos børn i Asien, er 10 mL blod ikke let at erhverve i alle regioner. Resultaterne af denne metode har bevist sin høje effektivitet og tidsbesparende.

En række metoder har været beskrevet at etablere NK/T cellelinjer fra patienter med lymfom eller Lymfoproliferative syndrom. Disse metoder ansat feeder celler, renset NK eller T celler med så meget som 10 mL blod eller en høj-dosis af IL-2. Denne undersøgelse præsenterer en ny metode med en kraftfuld og enkel strategi at etablere NK og T-celle linier ved dyrkning af perifert blod mononukleære celler (PBMC) med tilsætning af rekombinant humant IL-2 (rhIL-2), og bruger så lidt som 2 mL fuldblod. Cellerne kan formere sig hurtigt i to uger og opretholdes i mere end 3 måneder. Med denne metode, er der etableret 7 NK eller T-celle linier med en høj succesrate. Denne metode er simpel, pålidelig og gælder for oprettelse af cellelinjer fra flere tilfælde af CAEBV eller NK/T celle lymfom.

Efter 3 dage kultur under etableringen af cellelinjer, begynder polymorfe cellerne at dukke (figur 2). Efter 7 dage vokse celler hurtigt, da både antal og levedygtighed af celler er øget med en høj hastighed (figur 3).
