In diesem Protokoll wurde eine neuartige Technik für den Aufbau von NK/T-Zell-Linien aus dem Vollblut von CAEBV Patienten entwickelt. Im Vergleich mit den bisherigen Verfahren, ist der Hauptvorteil dieser Methode seiner Einfachheit und seiner Forderung nach nur ein kleines Volumen des Blutes, während es eine hohe Erfolgsquote und gute Bedingungen der Zellviabilität aufweist. Epstein - Barr-Virus (EBV) ist allgegenwärtig und befällt nicht nur B-Zellen, sondern auch T und natürlichen killer (NK)-Zellen, wodurch eine Reihe von EBV-assoziierten NK/T Lymphoproliferative Erkrankungen (LPD) und Lymphom/Leukämie, wie z. B. EBV-assoziierten hemophagocytic Lymphohistiocytosis, Hydroa Vacciniforme-wie Lymphom, extranodal NK/T-Zell-Lymphom, nasale Art und aggressive NK Zell-Leukämie1,2,3. Unter diesen ist schwerer chronisch aktive EBV (SCAEBV) Erkrankung, die einfallenden vor allem in Ostasien, und das gilt jetzt als ein LPD verursacht durch klonale Expansion von EBV-infizierte T oder NK Zellen4,5,6, 7, aber ohne die scheinbare Immunschwäche präsentieren in infektiöse Mononukleose (IM)-wie Symptomen wie Fieber, Hepatosplenomegalie, Lymphadenopathie und Leberfunktionsstörungen, anhaltend oder wiederkehrend sowie hohen EBV-DNA in die peripheren Blut8,9. Patienten mit CAEBV haben eine schlechte Prognose10,11, und seine Pathogenese und die Rolle der EBV ist unklar. Daher Zellinien von EBV-assoziierten NK/T Lymphoproliferative Erkrankungen abgeleitet und Lymphome sind sehr hilfreich, wie Zellmodelle für Klärung des Mechanismus der EBV NK oder T-Zell-Proliferation und seine Beziehung mit hoher Inzidenz von Leukämie induziert oder Lymphom.
Bislang wurden mehrere Zellinien mit verschiedenen Techniken12,13,14,15,16eingerichtet. Eine menschliche NK-Zell-Linie, NK-YS, entstand von NK-Zellen-Lymphom/Leukämie, durch Kultivierung zusammen mit einer Maus Stromazellen Zelllinie als Zubringer und in Anwesenheit von RhIL-2 in einer Konzentration von 20U pro mL15. KAI3 war ein weiterer NK-Zell-Linie gegründet von Patienten mit einer schweren Moskito-Allergie oder SCAEBV mit autologen Lymphoblastoid Zelllinie (LCL), B-Zellen durch EBV, als Feeder-Zellen und Zugabe von RhIL-2 bei 100U/mL16verwandelt. SNK6 und SNT8 wurden von Tumorgewebe des nasalen NK/T Zelle Lymphom Patienten abgeleitet, durch Zugabe von Hochdosis-RhIL-2 (700U/mL)12. Mit ähnlicher Technik war die SNK-1 Zelle von CAEBV Patienten, kultiviert von PBMC durch T-Zellen entfernen und Hinzufügen von 700U/mL RhIL-213,17. SNT13 und SNT15 wurden durch Entfernen von CD4 + und CD8 + Zellen18gegründet. Bisher wurden alle anderen T-Zellen und NK-Zell-Linien von EBV-NK/T LPD Patienten mit dieser Methode19entwickelt.
Die Nachteile der oben erwähnten vorhandenen Methoden gehören die Beschäftigung von Feeder-Zellen, das Erfordernis einer hohen Dosis von IL-2, die Auslastung von mehr als 10 mL Vollblut oder die Reinigung von NK/T-Zellen, die sehr anspruchsvoll in der Klinik die Notwendigkeit der Überprüfung, welche Arten von Zellen, dass EBV latent infiziert vor Beginn der Kultur. Als CAEBV vor allem bei Kindern in Asien tritt, ist 10 mL Blut nicht einfach, in allen Regionen zu erwerben. In dieser Studie haben wir eine neue einfache Methode mit einer hohen Erfolgsquote, NK und/oder T-Zell-Linien zu etablieren, durch Kultivierung PBMC von CAEBV Patienten mit niedrig dosierten rhIL2 und ein Volumen von 2 mL Vollblut ohne Feeder-Zellen entwickelt. Die Ergebnisse dieser Methode haben ihre hohe Effizienz und Zeitersparnis bewiesen.

Eine Reihe von Methoden wurden beschrieben, NK/T-Zell-Linien von Patienten mit Lymphom oder Lymphoproliferative Syndrome zu etablieren. Diese Methoden angewendet Feeder-Zellen, gereinigten NK oder T-Zellen mit mehr als 10 mL Blut oder eine hohe Dosis von IL-2. Diese Studie stellt eine neue Methode, eine leistungsstarke und einfache Strategie zu etablieren, dass NK und T-Zell-Linien durch Kultivierung der peripheren Mononukleäre Zellen (PBMC) Blut mit dem Zusatz von rekombinanten menschlichen Il-2 (RhIL-2) und weniger als 2 mL Vollblut verwendet. Die Zellen können in zwei Wochen schnell vermehren und für mehr als 3 Monate gehalten werden. Mit dieser Methode wurden 7 NK oder T-Zell-Linien mit einer hohen Erfolgsquote eingerichtet. Diese Methode ist einfach, zuverlässig und für Schaffung Zelllinien aus mehr Fälle von CAEBV oder NK/T-Zell-Lymphom.

Nach 3 Tagen während der Einrichtung von Zelllinien Kultur, beginnen die polymorphen Zellen erscheinen (Abbildung 2). Nach 7 Tagen wachsen Zellen schnell, da sowohl die Anzahl als auch die Lebensfähigkeit der Zellen mit einer hohen Rate (Abbildung 3) erhöht werden.
