Virus de Epstein - Barr (EBV) es ubicuo y se infecta no sólo las células de B, pero también T y células naturales del asesinas (NK), que causa un número de enfermedades linfoproliferativas NK/T EBV-asociado (LPD) y linfoma, leucemia, como hemophagocytic EBV-asociado lymphohistiocytosis, Hidroa vacciniforme-como el linfoma, el linfoma de células NK/T extranodal, tipo nasal y NK agresiva de la célula leucemia1,2,3. Entre éstos es severa active EBV (SCAEBV) enfermedad crónica, que es incidente principalmente en Asia oriental, y que ahora se considera un LPD causada por la expansión clonal de EBV-infectadas T o NK células4,5,6, 7, pero sin la aparente inmunodeficiencia presentes en mononucleosis infecciosa (IM)-como síntomas incluyendo fiebre, hepatoesplenomegalia, linfadenopatía y disfunción del hígado persistente o recurrente, así como alta DNA del VEB de la carga en el sangre periférica de8,9. Los pacientes con CAEBV tienen un pronóstico pobre10,11y su patogénesis y desconoce el papel de la EBV. Por lo tanto, la célula líneas derivadas de las enfermedades lymphoproliferative EBV-asociado NK/T y los linfomas son muy útiles como modelos celulares para clarificar el mecanismo de la EBV indujo proliferación NK o células T y su relación con alta incidencia de leucemia o linfoma.
Hasta la fecha, se han establecido varias líneas celulares con diferentes técnicas12,13,14,15,16. Una línea humana de la célula NK, NK-YS, fue establecida de leucemia/linfoma de células NK, por co-cultivo con una línea de células estromales de ratón como alimentador y en presencia de rhIL-2 en una concentración de 20U por mL15. KAI3 fue otra línea de células NK establecido de pacientes con alergia severa de mosquito o SCAEBV con línea de células limfoblastoides autólogas (LCL), células B transformadas por EBV, como células de alimentador y adición de rhIL-2 100U/mL16. SNK6 y SNT8 se derivan de los tejidos tumorales de pacientes de linfoma de la célula NK/T nasales mediante la adición de altas dosis de rhIL-2 (700U/mL)12. Con técnica similar, la célula de SNK-1 era de pacientes CAEBV, cultivados de PBMC quitando las células de T y adición de 700U/mL de rhIL-213,17. SNT13 y SNT15 fueron establecidos mediante la eliminación de CD4 + y CD8 + células18. Hasta ahora, otras líneas celulares de células T y NK de pacientes LPD EBV-NK/T fueron todos desarrollados con este método19.
Las desventajas de los métodos existentes antes mencionados incluyen el empleo de células de alimentador, el requisito de una alta dosis de IL-2, la utilización de tanto como 10 mL de sangre entera o la purificación de las células NK/T, que es muy difícil en la clínica debido a la necesidad de verificar que tipos de celular latente EBV infecta antes de empezar a la cultura. Como CAEBV ocurre principalmente en niños en Asia, 10 mL de sangre no es fácil de adquirir en todas las regiones. En este estudio, hemos desarrollado un nuevo método simple con un alto índice de éxito para establecer líneas NK y célula de T por cultivo PBMC de pacientes CAEBV usando una dosis baja de rhIL2 y un volumen de 2 mL de sangre sin células de alimentador. Los resultados de este método han demostrado su alta eficiencia y ahorro de tiempo.

Un número de métodos se ha descrito para establecer líneas de células NK/T de pacientes con linfoma o linfoproliferativo síndrome. Estos métodos emplean células de alimentador, células NK o T purificadas con tanto como 10 mL de sangre o una dosis alta de IL-2. Este estudio presenta un nuevo método con una estrategia potente y simple para establecer líneas de células T y NK por cultivo periférico de la sangre las células mononucleares (PBMC) con la adición de IL-2 humana recombinante (rhIL-2) y utiliza tan poco como 2 mL de sangre entera. Las células pueden proliferar rápidamente en dos semanas y se mantuvo por más de 3 meses. Con este método, se han establecido 7 líneas NK o células T con una alta tasa de éxito. Este método es simple, confiable y aplicable para el establecimiento de líneas celulares de más casos de linfoma CAEBV o NK/T.
