Dit protocol wordt een methode voor het leveren van nauwkeurige mechanische stimulatie aan de huid van een rondworm gevangen in een microfluidic chip gedemonstreerd. Het is bedoeld ter vergemakkelijking van de integratie van fysieke stimuli voor het beantwoorden van de vragen van de biologische en wil stroomlijnen mechanobiology onderzoek in biologische laboratoria. Het gevoel van aanraking biedt dieren met cruciale informatie over hun omgeving. Afhankelijk van de toegepaste kracht, wordt aanraking waargenomen als onschuldig, plezierig of pijnlijk. De vervorming van de weefsel tijdens aanraking wordt gedetecteerd door gespecialiseerde mechanoreceptor cellen ingebed in de huid die uitdrukken van receptor eiwitten, meestal ionenkanalen. De stappen die kracht perceptie koppelen tot activering van het kanaal van de ion tijdens touch en pijn zijn niet volledig begrepen. Nog minder is bekend over hoe het huidweefsel filters mechanische vervorming en of mechanoreceptors opsporing van wijzigingen in spanning of stress1,2,3. Deze kloof in begrip ontstaat, gedeeltelijk uit een gebrek aan geschikte instrumenten nauwkeurige mechanische stimulaties worden toegepast op het oppervlak van de huid van een levend dier tijdens het observeren van de reacties op cellulair niveau. Overwegende dat de atomaire kracht microscopie is uitvoerig gebruikt toe te passen en meten van krachten in geïsoleerde cellen4,5 , en ook om te activeren Piezo1 receptoren in levende cellen6, soortgelijke experimenten met behulp van levende dieren, met name C. elegans, zijn notoir uitdagend vanwege de intrinsieke mobiliteit van het onderwerp. Deze uitdaging is traditioneel omzeild door het gebruik van veterinaire - of chirurgische grade Cyanoacrylaat lijm te immobiliseren van individuele dieren op agar pads1,7,8,9. Deze aanpak is productief, maar heeft beperkingen met betrekking tot de vaardigheid die nodig is voor immobilisatie door lijmen en de oppervlakte van de zachte agar op mechanische naleving. Een microfluidics strategie is een gratis alternatief, dat enkele van de complicaties die verband houden vermijdt met het lijmen.
De nematode C. elegans is een genetisch modelorganisme met een volledig toegewezen zenuwstelsel dat, vanwege de grootte van het dier, een goed geschikt voor microfluidics technologie is. Microfluidics-gebaseerde apparaten bieden het voordeel dat het anders zeer mobiele dieren kunnen worden gefixeerd tijdens het uitvoeren van high-resolution beeldvorming en levering van relevante neuro-f stimuli. Met de hulp van microfluidic kunnen technologieën, leven dieren zonder schade10,11, waardoor controle van gedrags bedrijvigheid gedurende de gehele levensduur12,13 en met hoge resolutie worden geïmmobiliseerd Imaging van neuronale activiteit14,15,16,17. Verder veel mechanoreceptor neuronen die nodig is voor het gevoel van aanraking en pijn kan gekarakteriseerd worden op hun fysiologische1,8, mechanische4,18,19, en moleculaire niveau20,21,22.
C. elegans zintuigen zachte mechanische prikkels aan de muur van haar lichaam met behulp van zes TRNs, waarvan er drie innervate van het dier anterior (ALML/R en AVM) en drie van die innervate van het dier posterior (PLML/R en PVM). De ion kanaal moleculen die nodig zijn voor het transducing een toegepaste kracht in een biochemische signaal zijn uitvoerig bestudeerd in haar TRNs8. Dit artikel presenteert een microfluidic platform23 waarmee onderzoekers toe te passen nauwkeurige mechanische krachten op de huid van een geïmmobiliseerdet C. elegans rondworm, terwijl het uitlezen van de vervorming van zijn interne weefsels door optische beeldvorming. Naast de presentatie van welomschreven mechanische stimuli, kunnen calcium transiënten worden opgenomen in de mechanoreceptor neuronen met subcellular resolutie en gecorreleerd met morfologische en anatomische kenmerken. Het apparaat bestaat uit een centrale overlapping-kanaal dat een enkel dier houdt en presenteert haar huid naast Pneumatische bediening kanalen (Figuur 1 en Figuur 2). De zes kanalen staan langs het kanaal van de overlapping voor mechanische prikkels naar elk van de zes TRNs van de worm. Deze kanalen zijn gescheiden van de kamer van de overlapping door dunne PDMS pessarium, die kunnen worden aangedreven door een externe lucht druk bron (Figuur 1). Wij gekalibreerd de uitwijking ten opzichte van de druk en de metingen in dit artikel bieden. Elke actuator kan worden individueel gericht en gebruikt ter stimulering van een mechanoreceptor van keuze. De druk wordt geleverd met een piëzo-gedreven drukpomp maar alternatieve apparaat kan worden gebruikt. We laten zien dat de druk-protocol kan worden gebruikt om te activeren TRNs in vivo en demonstreren van huishoudelijke apparaten geschikt voor het leveren van mechanische stimuli aan volwassen C. elegans, laden van volwassen dieren in apparaten, uitvoeren van calcium imaging experimenten, en analyseren van de resultaten. Apparaat fabricage bestaat uit twee hoofdstappen: 1) fotolithografie te maken van een mal van SU-8; en 2) molding PDMS om een apparaat te maken. Ter wille van de beknoptheid en duidelijkheid, worden lezers verwezen naar eerder gepubliceerde artikelen en protocollen24,25 voor instructies over hoe de mallen en de apparaten te produceren.

Een centrale doelstelling van mechanobiology is te begrijpen van de wederzijdse invloed van mechanische belasting op eiwitten en cellen. Ondanks het belang ervan, is de invloed van mechanische belasting op cellulaire functie nog slecht begrepen. Gedeeltelijk bestaat deze kenniskloof omdat enkele hulpmiddelen gelijktijdige vervorming van weefsels en cellen, beeldvorming van cellulaire activiteit in levende dieren en efficiënte beperking van de beweeglijkheid in anders zeer mobiele modelorganismen, zoals de nematode kunt Caenorhabditis elegans. De kleine grootte van C. elegans maakt ze een uitstekende match op microfluidics gebaseerde onderzoek apparaten, en oplossingen voor immobilisatie zijn aangebracht met behulp van microfluidic-apparaten. Hoewel deze apparaten toestaan voor hoge resolutie beeldvorming, is het dier volledig ingekapseld in Polydimethylsiloxaan (PDMS) en glas, het beperken van fysieke toegang voor de levering van de mechanische kracht of elektrofysiologische opnames. Onlangs, we hebben een apparaat dat integreert pneumatische aandrijvingen met een overlapping design die compatibel is met hoge-resolutie fluorescentie microscopie. Het kanaal van de bediening wordt gescheiden van de worm-overlapping kanaal door een dunne PDMS diafragma. Dit membraan wordt afgebogen in de zijkant van een worm door druk uit te oefenen vanuit een externe bron. Het apparaat kan richten op individuele mechanosensitive neuronen. De activering van deze neuronen is beeld op hoge resolutie met genetisch-gecodeerde calcium indicatoren. Dit artikel presenteert de algemene methode met behulp van C. elegans spanningen uiten calcium-gevoelige activiteitsindicator (GCaMP6s) in hun aanraking receptor neuronen (TRNs). De methode is niet beperkt tot TRNs noch tot calcium sensoren als een sonde, maar kan worden uitgebreid tot andere mechanisch-gevoelige cellen of sensoren.
