Erken çalışmalar Reaktif oksijen türleri (ROS) kritik rolü sisplatin ve aminoglikozitler tarafından üretilen ototoksisite arabuluculuk göstermiştir. Sonraki çalışmalarda NADPH oksidaz NOX3 izoformu ROS birincil kaynak koklea içinde ve sisplatin6,7tarafından aktif gösterdi.
