De methode die hier gepresenteerd demonstreert een unieke neurale stimulatie paradigma, waarin retinale neuronen chemisch worden gestimuleerd door het injecteren van inheemse neurotransmitter chemische stoffen in de ondergrond van het netvlies in vitro. Fotoreceptor degeneratieve ziekten, zoals retinitis pigmentosa en leeftijdsgebonden maculadegeneratie, zijn belangrijkste overneembare oorzaken van verlies van de visie en zijn momenteel ongeneeslijke1,2. Hoewel deze ziekten ontstaan uit een aantal specifieke genetische mutaties, worden fotoreceptor degeneratieve ziekten gekenmerkt als een groep door het progressieve verlies van de fotoreceptor cellen in het netvlies, waardoor uiteindelijk blindheid. Het verlies van researchdieren triggers wijdverspreid in het netvlies het remodelleren maar overleven retinale neuronen, met inbegrip van de bipolaire cellen en RGCs, blijven intact en relatief functioneel zelfs in gevorderde stadia van fotoreceptor degeneratie3 ,4,5,6,7.
De mechanismen en de pathologie van deze ziekten zijn goed gekarakteriseerd3,4,5,6,7 , maar een effectieve behandeling blijft ongrijpbaar. In de afgelopen drie decennia, hebben de onderzoekers wereldwijd een scala aan therapeutische behandelingen voor het herstel van de visie aan de betrokkenen met fotoreceptor degeneratieve ziekten met inbegrip van gene therapie8, stamcel behandeling9, onderzocht netvlies transplantatie10, en kunstmatige stimulatie11,12 van de overlevende retinale neuronen. Van deze zijn de meest klinisch beschikbare netvlies prothesen, die kunstmatige neurostimulatie-apparaten die een matrix van elektroden elektrisch bipolaire cellen of de RGCs stimuleren in specifieke patronen met het doel van oudsher hebben gebruikt het creëren van kunstmatige visuele waarnemingen in patiënten11. Huidige generatie elektrische prothesen, zoals de Argus II13 en Alpha-IMS14 apparaten, klinische goedkeuring hebben bereikt en voorbereidende studies hebben aangegeven dat ze verbeteren van de levenskwaliteit voor patiënten door het herstel van kunnen een maatregel van visie met behulp van zowel Epiretinaal (voorkant van de retina) en subretinal (achterkant van het netvlies) geïmplanteerd apparaten15,16. Onderzoeksgroepen over de hele wereld zijn bezig met het bevorderen van de retinale prothesen buiten de successen van deze eerste generatie apparaten17,18,19,20 , maar hebben te kampen moeilijkheden ontwerpen een elektrische prothese kunnen herstellen van hoge gezichtsscherpte visie onder het niveau van juridische blindheid aan patiënten. Recente studies hebben aangetoond dat het bereiken van hogere ruimtelijke resolutie dan de huidige generatie gegevensgestuurd elektrische gebaseerde prothesen is uitdagend vanwege deze beperking tot gratis injectie, die noodzaakt het gebruik van grote elektroden veilig stimuleren netvlies neuronen ten koste van ruimtelijke resolutie, dwz gezichtsscherpte11,21. Bovendien elektrische stimulatie is verder beperkt omdat het meestal alle nabijgelegen cellen stimuleert en daarom onnatuurlijk en verwarrend waarnemingen bij patiënten lokt, vooral omdat er een inherent onnatuurlijke stimulatie paradigma21. De vroege successen van elektrische stimulatie hebben echter aangetoond dat kunstmatige neurostimulatie kunnen een effectieve behandeling voor fotoreceptor degeneratieve ziekten. Dit doet veronderstellen dat een nog doeltreffender behandeling haalbaar wellicht door het stimuleren van het netvlies met neurotransmitter chemicaliën, de natuurlijke agentia communicatiemiddel op chemische synapsen. Het doel van de methode die in dit document gepresenteerd is aan de therapeutische haalbaarheid onderzoeken van chemische stimulatie, die tot doel heeft na te bootsen de natuurlijke systeem van synaptic communicatie tussen retinale neuronen, als een alternatief voor elektrische stimulatie van biomimetische voor een retinale prothese.
Vertaling van het concept van therapeutische chemische stimulatie tot een chemische retinale prothese is gebaseerd op chemisch activeren doel retinale neuronen met kleine hoeveelheden van inheemse neurotransmitters, zoals glutamaat, uitgebracht door middel van een microfluidic apparaat bestaande uit een groot scala aan microports in reactie op visuele stimulatie. Op deze manier zou een chemische retinale prothese in wezen een biomimetische kunstmatige fotoreceptor laag waarmee fotonen natuurlijk het bereiken van het netvlies naar chemische signalen worden omgezet. Aangezien deze chemische signalen dezelfde neurotransmitters gebruikt gebruiken in normale netvlies signalering en de overlevende retinale neuronen van een ontaarde netvlies door de dezelfde synaptic trajecten gebruikt door normaal gezichtsvermogen trajecten, de resulterende visuele stimuleren perceptie bereikt door middel van een chemische retinale prothese zou meer natuurlijke en begrijpelijke t.o.v. een opgeroepen door middel van een elektrische prothese. Bovendien, aangezien de microports via welke neurotransmitters worden uitgebracht kan worden gemaakt zeer kleine en gekleed in hoge dichtheid, in tegenstelling tot de elektroden, een potentiële chemische prothetische zou kunnen bereiken meer focale stimulatie en hogere ruimtelijke resolutie dan een elektrische prothese. Dus, op basis van deze potentiële voordelen, een chemische retinale prothese biedt een zeer veelbelovend alternatief voor elektrische prothesen.
Chemische stimulatie van het netvlies, echter is relatief weinig onderzocht tot voor kort. Terwijl elektrische stimulatie van het netvlies goed decennia van werk door middel van klinische studies13,2322, in vitroen in vivo23,24 gekenmerkt ,14, studies over chemische stimulatie geweest beperkt uitsluitend tot een paar in vitro werken25,26,27,28. Iezzi en Finlayson26 en Inayat et al. 27 aangetoond Epiretinaal chemische stimulatie van het netvlies in vitro met behulp van een enkele indicatorelektrode en een multielectrode matrix (MEA), respectievelijk, de glutamaat opgeroepen reacties van retinale neuronen opnemen. Meer recentelijk Rountree et al. 28 aangetoond de differentiële stimulatie van de OFF and ON retinale trajecten met behulp van glutamaat uit de subretinal kant en een MEA opnemen van de neuronale reacties van meerdere sites op het netvlies.Hoewel deze werken de haalbaarheid van chemische stimulatie preliminair opgericht hebben, verdere studies zijn essentieel voor de onderzoeken veel aspecten van deze aanpak die verder gaan dan gericht tot nu toe25,26,27 , 28, en fine-tunen van de therapeutische stimulatie parameters in zowel in vitro als in vivo dierlijke modellen voor het vertalen van dit concept naar een chemische retinale prothese zoals hierboven besproken. Echter op dit moment is er geen vastgelegde methode voor het uitvoeren van chemische stimulatie van het netvlies in de literatuur en de methoden die worden gebruikt in de eerdere werken niet zijn beschreven in zo gedetailleerd als essentieel voor Dr. studies zou zijn. De reden voor deze methoden papier daarom een duidelijk afgebakende kader te bieden voor het uitvoeren van in vitro chemische stimulatie van het netvlies voor deze onderzoekers ook geïnteresseerd bij het repliceren van onze vorige studies27, 28 of verder bevorderen dit ontluikende concept van chemische neurostimulatie.
Hier tonen we een methode voor het uitvoeren van in vitro chemische stimulatie van het netvlies neuronen in wholemount netvlies van wild-type ratten en een fotoreceptor gedegenereerd rat model dat nauw samen van de Nabootsers van de progressie van fotoreceptor degeneratieve aandoeningen bij de mens. De grondgedachte achter deze stimulatie-methode in in vitro modellen ontwikkelen is om te evalueren van de therapeutische bereiken van verschillende parameters van de stimulatie en bestuderen van de neurale reactie kenmerken dat zou onmogelijk of moeilijk te observeren in in vivo modellen, vooral tijdens de eerste studies gericht op een beoordeling van de haalbaarheid van deze aanpak. In deze procedure tonen we zowel één site en gelijktijdige multi-site chemische stimulaties van netvlies door het leveren van kleine hoeveelheden van 1 mM glutamaat in de buurt van de retinale neuronen doel via verkrijgbare één poort glas micropipetten en een op maat micromachined Multi-port microfluidic apparaat, respectievelijk. Terwijl zowel één site en meerdere sites stimulaties de fundamentele doelstelling bereiken van het onderzoek naar de therapeutische haalbaarheid van chemische neuromodulatie, dient elk een specifiek doel met een uniek voordeel. De stimulatie van één site, die kan worden bereikt met verkrijgbare vooraf getrokken glas micropipetten, kan worden gebruikt voor het injecteren van chemicaliën direct in de ondergrond van het netvlies op één site en dient om te onderzoeken of de regering van Cambodja waarneembare spike tarief reacties die vergelijkbaar met visueel evoked lichte reacties zijn kunnen focally worden ontlokte onder de injectieplaats. Aan de andere kant, multi-site stimulatie, waarvoor een speciaal gefabriceerde multipoort microfluidic apparaat, kan worden gebruikt voor het injecteren van chemicaliën ruimtelijk op meerdere locaties over het oppervlak van het netvlies en dient om te onderzoeken hoe goed glutamaat-opgeroepen RCG reactie patronen komen overeen met de glutamaat injectie patronen in patroon stimulatie studies.

Fotoreceptor degeneratieve ziekten veroorzaken onherstelbare blindheid door het progressieve verlies van fotoreceptor cellen in het netvlies. Netvlies prothesen zijn een opkomende behandeling voor fotoreceptor degeneratieve ziekten die herstellen visie willen door kunstmatig de overlevende retinale neuronen in de hoop tot begrijpelijke visuele perceptie bij patiënten te stimuleren. Huidige retinale prothesen hebben bewezen succes in het herstellen van beperkte visie aan patiënten met behulp van een matrix van elektroden aan elektrisch stimuleren van het netvlies, maar geconfronteerd met aanzienlijke fysieke barrières in het herstellen van hoge gezichtsscherpte, natuurlijke visie aan patiënten. Chemische neurostimulatie met behulp van inheemse neurotransmitters is een biomimetische alternatief voor elektrische stimulatie en kon negeren de fundamentele beperkingen retinale prothesen met behulp van elektrische neurostimulatie is gekoppeld. In het bijzonder heeft chemische neurostimulatie het potentieel om te herstellen meer natuurlijke visie met vergelijkbaar of beter visual acuities patiënten door het injecteren van zeer kleine hoeveelheden van neurotransmitters, de dezelfde natuurlijke agenten van communicatie gebruikt door retinale chemische synapses, bij veel fijnere resolutie dan de huidige elektrische prothesen. Echter, als een relatief onontgonnen stimulatie paradigma, er is geen gangbaar protocol voor de verwezenlijking van chemische stimulatie van het netvlies in vitro. Het doel van dit werk is een kader te bieden gedetailleerde voor het uitvoeren van chemische stimulatie van het netvlies voor onderzoekers die willen studeren het potentieel van chemische neuromodulatie van het netvlies of soortgelijke zenuwweefsel in vitro. In dit werk, beschrijven we de experimentele opzet en methodologie voor het opwekken van retinale ganglion cel (regering van Cambodja) spike reacties vergelijkbaar met visuele lichte reacties bij wild-type en wholemount fotoreceptor-verworden rat netvlies door het injecteren van volumes van gecontroleerde de neurotransmitter glutamaat in de subretinal ruimte met behulp van glas micropipetten en een aangepaste multipoort microfluidic apparaat. Deze methodologie en protocol zijn algemeen genoeg om te worden aangepast voor neuromodulatie met behulp van andere neurotransmitters of zelfs andere zenuwweefsel.

Dit protocol kan worden gebruikt voor chemisch stimuleren zowel normale, wild-type netvlies evenals fotoreceptor verworden netvlies, ondanks de aanzienlijke cellulaire remodelleren veroorzaakt door het verlies van de researchdieren.
