Terapia de gene do ouvido interno é um campo de rápido desenvolvimento de investigação. Aplicação em vários modelos animais para combate ototoxicidade, trauma de ruído e de perda de audição hereditária1. Vários estudos recentes têm mostrado recuperação funcional da audição e funções de equilíbrio em camundongos mutantes após ouvido interno gene terapia entrega2,3,4,5,6, 7. um dos principais fatores na determinação do sucesso da terapia do gene de orelha interna é a abordagem cirúrgica utilizada para acessar o ouvido interno. Idealmente, a abordagem cirúrgica seria fácil de executar, os marcos anatômicos seria consistente e fácil de identificar e a transdução resultante de tipos de células alvo seria alta.
Em um estudo recente, mostramos que, quando a terapia do gene viral foi injectada através do canal semicircular posterior do rato mutante whirler (um modelo de perda auditiva e disfunção vestibular), transdução eficiente de células sensoriais do cabelo foi vista nos vestibulares órgãos, assim como a cóclea5. A alta eficiência de transdução sensorial cabelo célula resultou em melhora das funções auditivas e vestibulares nestes ratos mutantes.
Neste artigo, descrevemos em detalhe a abordagem de canal semicircular posterior para acessar o ouvido interno do rato neonatal.

Terapia de gene do ouvido interno uma grande promessa como um potencial tratamento para perda de audição e tontura. Um dos determinantes essenciais do sucesso da terapia de gene do ouvido interno é encontrar um método de entrega que resulta em eficiência de transdução consistente dos tipos de célula alvo, minimizando a perda de audição. Neste estudo, descrevemos a abordagem de canal semicircular posterior como um método viável para a entrega de gene do ouvido interno em camundongos neonatais. Mostramos que a entrega do gene através do canal semicircular posterior é capaz de perfundir toda orelha interna. A fácil identificação anatômica do canal semicircular posterior, bem como, manipulação mínima do osso temporal exigido, fazer neste cirúrgico aproximar-se uma opção atraente para entrega de gene do ouvido interno.
