Actualmente el costo de la detección, diagnóstico y tratamiento del cáncer es muy alto. Existen diferentes modalidades que se utilizan para la detección del cáncer y el diagnóstico de la proyección de imagen. Sin embargo, muchas de estas técnicas de imagen tienen limitaciones incluyendo máquina voluminoso tamaño, diagnóstico invasivo, hostilidad a requisito demasiado caro, pacientes, de las radiaciones ionizantes, o el uso de agentes de contraste radiactivo. Por lo tanto, se necesita mucho una imagen eficiente...

Para la detección y estadificación del cáncer, diferente técnicas de imagen están disponibles. Algunas de las modalidades de imagen utilizadas son la proyección de imagen de resonancia magnética (MRI), rayos x tomografía computada (CT), rayos x, ultrasonido (US), tomografía por emisión de positrones (PET), fluorescencia de imagen, etc.1,2, 3 , 4. pero, algunas de las técnicas de imagen existentes son invasoras, tienen radiaciones nocivas, o son lentas, costosas, voluminosas o antipático a los pacientes. Así, hay una necesidad constante para desarrollar nuevas técnicas de imagen rápidas y rentables para el diagnóstico y la terapia5.
Fotoacústica imaging (PAI) es una técnica emergente, que combina el rico contraste óptico con ultrasonido de alta resolución en una más profunda imagen profundidad5,6,7,8, 9. En PAI, un pulso de láser corta se utiliza para la irradiación del tejido. La luz es absorbida por el tejido que lleva a un pequeño aumento de temperatura. Debido a la expansión termoelástica, se generan ondas de presión (en forma de ondas acústicas) dentro del tejido. Las ondas acústicas generadas (también conocido como ondas de fotoacústica (PA)) se adquieren con un transductor de ultrasonido de banda ancha (UST) fuera de los límites del tejido. Estas señales PA adquiridas pueden utilizarse para reconstruir imágenes de PA, revelando la información estructural y funcional dentro del tejido. PAI tiene una gran variedad de aplicaciones, incluyendo: la proyección de imagen vascular, la proyección de imagen de ganglio linfático centinela, imagen de la vasculatura cerebral, proyección de imagen de tumor, la proyección de imagen molecular, etc.10,11,12, 13,14,15 PAI tiene numerosas aplicaciones debido a sus ventajas, a saber: profundidad de la penetración más profunda, buena resolución espacial y contraste de tejidos blandos alta. El contraste en PAI puede ser endógeno de sangre, melanina, etcetera. Cuando el contraste endógeno no es bastante fuerte, agentes de contraste exógeno como colorantes orgánicos, nanopartículas, puntos cuánticos, etc.16,17,18,19, 20 , 21 puede utilizarse para mejorar el contraste.
Aunque el PAI tiene numerosos beneficios en comparación con otras técnicas de imagen, traducción clínica es todavía un desafío muy grande. Las principales limitaciones son la naturaleza voluminosa de los lasers se utilizan, la mayoría de los subterráneos utilizados para adquisición de datos no es compatible con sistemas clínicos de Estados Unidos y la no disponibilidad de sistemas comercialmente disponibles clínicos de Estados Unidos imágenes que dan acceso al canal de crudo datos. Sólo recientemente, comercial clínicas máquinas de Estados Unidos con acceso a los datos crudos se han convertido en disponibles22. En este trabajo, nuestro objetivo es demostrar la viabilidad de PAI con una mano puesta a punto utilizando una plataforma clínica de Estados Unidos. Nuestro objetivo es demostrar esto mostrando la proyección de imagen no invasiva de centinela los nodos de linfa (SLNs) en un modelo animal pequeño.
Tumores de mama invasivos son una de las principales causas de muerte por cáncer entre las mujeres. Diagnóstico y escenificando el cáncer de mama temprano es vital para decidir estrategias de tratamiento, que juegan un papel importante en el pronóstico del paciente. Para mama cáncer estadificación biopsias de ganglio linfático centinela (SLNB) suelen utiliza23,24. SLN es el principal nodo de linfa donde la posibilidad de encontrar células de cáncer es el más alto debido a la metástasis. SLNBs impliquen inyectar un colorante o un trazador radioactivo, seguido por corte abierto en la zona con una pequeña incisión y luego ubicar el SLN visualmente en caso de tintes o con la ayuda de un contador Geiger, en caso de un trazador radioactivo. Después de la identificación, se quitan unos SLN para estudios histopatológicos24,25. SLNB positivo indica que el tumor ha hecho metástasis a los ganglios linfáticos cercanos y tal vez a otros órganos. SLNB negativo indica que la probabilidad de metástasis es insignificante26. SLNB tiene numerosas complicaciones asociadas con ella como entumecimiento de brazo, linfedema, etc.27 eliminar las complicaciones SLNB asociado, se necesita una técnica de imagen no invasivas.
Para la asignación de SLN en pequeños animales y los seres humanos, PA la proyección de imagen se ha explorado extensivamente con la ayuda de contraste diferentes agentes15,28,29,30,31 , 32. sin embargo, los sistemas utilizados actualmente no pueden utilizarse en un escenario clínico como señalado anteriormente. Otra preocupación a abordarse es el procedimiento quirúrgico en SLNB28. Adaptación de procedimientos mínimamente invasivos para la biopsia aspiración con aguja fina (FNAB) era necesario para reducir el tiempo de recuperación y los efectos secundarios de los pacientes. En este trabajo, se utilizó un sistema clínico de Estados Unidos para proyección de imagen de Estados Unidos y PA combinado fue utilizado. Para facilidad de uso en configuración clínica, un encargo de la mano de cubierta de fibra óptica y UST fue diseñado. Azul de metileno (MB) fue utilizado para la identificación y mapeo de SLNs. Además, para eliminar las complicaciones asociadas con la cirugía SLNB, aguja en tiempo real no invasivo de seguimiento también se demuestra.

hand-held clinical photoacoustic imaging system for real-time non-invasive small animal imaging

Traducción de la proyección de imagen fotoacústica en la clínica es un gran reto. Sistemas de imagen fotoacústica clínica en tiempo real mano son muy raros. Aquí, Divulgamos un combinado fotoacústica y clínico ultrasonido sistema de imagen mediante la integración de una sonda de ultrasonido con luz entrega para la proyección de imagen de animal pequeño.
