För att upptäcka och stadieindelning av cancer finns olika avbildningstekniker. Några av de utbredda avbildningsmetoder är magnetisk resonanstomografi (MRT), röntgen datortomografi (CT), röntgen, ultraljud (oss), positronemissionstomografi (PET), fluorescens imaging, etc.1,2, 3 , 4. men, några av de befintliga imaging teknikerna är antingen invasiv, har skadlig strålning eller är långsamma, dyra, skrymmande eller ovänliga till patienter. Således finns det ett konstant behov av att utveckla nya, snabba och kostnadseffektiva avbildningstekniker för diagnostik och terapi5.
Photoacoustic imaging (PAI) är en framväxande imaging teknik, som kombinerar rik optiska kontrast ultraljud med hög upplösning på en djupare imaging djup5,6,7,8, 9. I PAI används en kort laserpuls för vävnad bestrålning. Ljuset blir absorberas av vävnaden vilket leder till en liten temperaturstegring. Följd av termoelastisk expansion genereras tryckvågorna (i form av Akustiskt vinkar) inom vävnaden. Det genererade akustiska vinkar (även känd som photoacoustic (PA) vågor) förvärvas med en wideband ultraljud givare (UST) utanför gränsen som vävnad. Dessa förvärvade PA signaler kan användas för att rekonstruera PA bilder, avslöjar den strukturella och funktionella informationen inuti vävnaden. PAI har ett brett spektrum av applikationer, inklusive: blodkärl imaging, sentinel lymfkörtel imaging, vaskulatur hjärnavbildning, tumör imaging, molekylär imaging, etc.10,11,12, 13,14,15 PAI har många tillämpningar på grund av dess fördelar, nämligen: djupare genomträngningsdjupet, bra rumslig upplösning och hög mjukvävnad kontrast. Kontrasten i PAI kan vara endogena från blod, melanin, etc. När endogena kontrasten inte är tillräckligt stark, som exogena kontrastmedel organiska färgämnen, nanopartiklar, kvantprickar, etc.16,17,18,19, 20 , 21 kan användas för att förbättra kontrasten.
Även om PAI har många fördelar i förhållande till andra avbildningstekniker, är kliniska översättning fortfarande en mycket stor utmaning. De viktigaste begränsningarna är skrymmande beskaffenhet lasrar som används, de flesta av de låter som används för datainsamling är inte kompatibla med klinisk US system och icke-tillgängligheten av kommersiellt tillgängliga kliniska US bildsystem som ger tillgång till raw channel data. I detta arbete vill vi demonstrera genomförbarheten av PAI med en handhållen set-up med en klinisk US-plattform. Vi syftar till att visa detta genom att visa icke-invasiv avbildning av sentinel lymfkörtlar (SLNs) i en liten djurmodell.
Invasiv bröstcancer tumörer är en av de ledande orsakerna till dödsfall i cancer bland kvinnor. Diagnostisera och Förproduktion bröstcancer tidigt är avgörande för att bestämma behandlingsstrategier, som spelar en viktig roll i prognosen för patienten. För bröstcancer används cancer mellanlagringsplatsen sentinel lymfkörteln biopsier (SLNB) är oftast23,24. SLN är den primära lymfkörteln där möjligheten att hitta cancerceller är högst på grund av metastaser. Negativa SLNB visar att sannolikheten för metastaser är försumbar26. SLNB har många komplikationer i samband med det som arm domningar, lymfödem, etc.27 att eliminera SLNB associerade komplikationer, en icke-invasiv bildteknik behövs.
För SLN mappning i små djur och människor, har PA imaging undersökts i stor utsträckning med hjälp av olika kontrast agenter15,28,29,30,31 , 32. men de system som används för närvarande kan inte användas i kliniska scenario som påpekade tidigare. Ett annat bekymmer tas upp är det kirurgiska ingreppet som är inblandade i SLNB28. Anpassa minimalt invasiva ingrepp för fin nål aspiration biopsi behövdes (FNAB) för att minska återhämtningstiden och biverkningar av patienterna. I detta arbete, en klinisk US-system användes för kombinerade USA och PA imaging användes. För enkel användning i kliniska inställningar, en anpassad gjort handhållna hållare för bostäder optisk fiber och UST utformades. Metylenblått (MB) användes för att identifiera och kartlägga SLNs. Dessutom för att eliminera komplikationer i samband med SLNB operationen, icke-invasiv realtid nål spårning demonstreras också.

hand-held clinical photoacoustic imaging system for real-time non-invasive small animal imaging

Översättning av photoacoustic imaging till kliniken är en stor utmaning. Handhållen realtid kliniska photoacoustic bildsystem är mycket sällsynta. Vi rapporterar här, en kombinerad photoacoustic och kliniska ultraljud imaging system genom att integrera en ultraljudssond med ljus leverans för små djur avbildning. Vi visar detta genom visar sentinel lymfkörtel imaging i små djur tillsammans med minimalinvasiv realtid nål vägledning. En klinisk ultraljud plattform med tillgång till raw channel data tillåter integration av photoacoustic imaging leder till en handhållen realtid kliniska photoacoustic imaging system. Metylenblått användes för sentinel lymfkörtel imaging 675 nm våglängd. Dessutom visades nål vägledning med dubbla modala ultraljud och photoacoustic imaging använder imaging system. Djup avbildning av upp till 1,5 cm visades med 10 Hz laser på en photoacoustic imaging bildfrekvens på upp till 5 bilder per sekund.
