In de afgelopen jaren heeft licht ontpopt als een uitstekend hulpmiddel voor spatiotemporally gecontroleerde activering van cellulaire processen. Verschillende methoden zijn ontwikkeld, elk met de bijbehorende voordelen en nadelen. Het systeem hier beschreven, die is gebaseerd op de decaging van geïmmobiliseerdet, extracellulaire ligand, is bij uitstek geschikt voor de analyse van snelle, subcellular, gepolariseerde signalering reacties. Deze aanpak is vereffend te onderzoeken IS vorming in T cellen zoals hierboven besc...

T-lymfocyten (T-cellen) spelen een centrale rol in de cellulaire immuniteit door de erkenning van de antigene peptiden in het kader van het celoppervlak MHC weergegeven. Antigeen erkenning, dat is bemiddeld door de TCR, drijft de differentiatie van naïeve T cellen en bevordert de levering van de cytolytische en communicatieve antwoorden van effector populaties. TCR betrokkenheid induceert ook dramatische veranderingen in cellulaire architectuur. Binnen enkele minuten de T-cellen gloms op de kant van de antigeen-presenteren cel (APC), vormen een gepolariseerde interface genoemd de immunologische synapse (IS)1,2. Het IS potentiates T cel effector reacties doordat de directionele release van cytokines of, in het geval van cytotoxische T-lymfocyten (CTL's), lytische eiwitten die de APC vernietigen.
TCR betrokkenheid door pMHC induceert de snelle fosforylatie van meerdere stroomafwaartse adapter moleculen, met inbegrip van de Linker voor de activering van T-cellen (LAT), die uiteindelijk bevordert robuuste remodelleren van de synaptische cytoskelet2. Corticale filamenteuze actine (F-actine) T cel verspreiden over het oppervlak van de APC-stations, en vervolgens moet omzetten naar een ringvormige structuur gekenmerkt door ophoping van de F-actine onderaan de IS periferie en de uitputting van het centrum. F-actine ring vorming is het strak gekoppeld aan de heroriëntering van het organiserende centrum van microtubuli (MTOC, ook wel de centrosoom in T-cellen) naar een positie net onder het midden van de interface. Beide gebeurtenissen zich voordoen binnen enkele minuten van de eerste antigeen erkenning en stellen de architecturale context waarin de activering van de daaropvolgende gebeurtenissen en effector reacties optreden.
Studeren IS vorming, hebben verschillende labs benaderingen waarbij de APC is vervangen door een glazen oppervlak, dat geïmmobiliseerdet TCR liganden bevat of ondersteunt een lipide dubbelgelaagde dat zelf de liganden3,4 bevatontwikkeld. T-cellen vormen IS-achtige contacten op deze oppervlakken die image kunnen worden gemaakt door de totale interne reflectie fluorescentie Microscoop (TIRF) of confocale microscopie, waardoor hoge resolutie studies van vroege T-cel activatie en IS vorming.
Hoewel deze benaderingen hebben toegestaan voor uitstekende visualisatie van het volledig geassembleerde IS, plaatsvindt veel van de signalering volgende TCR:pMHC afbinding binnen seconden, complicerende inspanningen om te bepalen van de volgorde van de gebeurtenissen na de activering van de TCR nauwkeurig . Om dit probleem te omzeilen, een photoactivation aanpak ontwikkeld, waarin photoactivatable pMHC wordt gebruikt om spatio controle over TCR activering5,6,7. In dit systeem, zijn T-cellen gekoppeld aan glazen oppervlakken met photoactivatable pMHC thats niet-stimulerend voor de TCR totdat bestraald met ultraviolet (UV) licht. UV bestraling van een regio van de micron grootte van het oppervlak onder de T cel verwijdert het creëren van een stimulerend zone die kan worden herkend door de T-cel photocage. Daaropvolgende signalering gebeurtenissen en cytoskeletal remodeling worden vervolgens gecontroleerd met behulp van genetisch gecodeerde fluorescerende verslaggevers. Twee versies van de photoactivatable van antigene peptides, nachtvlinder cytochroom c88-103 (MCC) en ovoalbumine257-264 (OVA), die worden gepresenteerd in het kader van de klasse II MHC-Ek en de klasse ik MHC H2-Kb, respectievelijk, zijn geweest ontwikkeld (Figuur 1). Dit maakt de analyse van beide CD4+ T cellen specifiek voor MCC-ik-Ek (uiting van de 5 C. C7, 2B4, of en TCRs) en CD8+ T cellen specifiek voor eicellen-H2-Kb (uiten de OT1 TCR).

T-lymfocyten zich bezighouden met het snelle, gepolariseerde signalering, die zich voordoen binnen enkele minuten na activering van de TCR.
