In den letzten Jahren hat Licht als ein ausgezeichnetes Werkzeug für raumzeitlich kontrollierte Aktivierung von zellulären Prozessen entstanden. Verschiedene Methoden wurden entwickelt, jede mit damit verbundenen vor- und Nachteile. Das System hier beschrieben wird, die auf die decaging von immobilisierten, extrazellulären Liganden, eignet sich besonders für die Analyse von rapid, subzelluläre, polarisierte Signalisierung Antworten. T-Lymphozyten (T-Zellen) spielen eine zentrale Rolle in der zellulären Immunität durch das erkennen Antigene Peptide, die im Rahmen der Zelloberfläche MHC angezeigt. Antigen-Anerkennung, die durch die TCR vermittelt wird, treibt die Differenzierung der naiven T-Zellen und fördert die Lieferung der Zytolyse und kommunikative Reaktionen der Effektor Populationen. TCR Engagement induziert auch dramatische Veränderungen in der zellulären Architektur. Innerhalb von Minuten die T-Zellen gloms auf der Seite der Antigen-präsentierenden Zelle (APC), bilden eine polarisierte Schnittstelle bekannt als der immunologischen Synapse (IS)1,2. Der IS potenziert T-Zell-Effektor-Reaktionen durch die Aktivierung der direktionalen Freisetzung von Zytokinen oder, im Falle von zytotoxischen T-Lymphozyten (CTLs), lytische Proteine, die die APC zerstören.
TCR Engagement von pMHC induziert die schnelle Phosphorylierung mehrere nachgeschaltete Adapter Moleküle, einschließlich Linker für die Aktivierung von T-Zellen (LAT), die letztlich fördert robuste Umgestaltung der synaptischen cytoskelett2. Kortikale filamentösen Aktin (F-Aktin) treibt T Zelle auf der APC-Oberfläche verteilt und löst sich dann in eine ringförmige Struktur zeichnet sich durch F-Aktin-Ansammlung an der Peripherie IS und Erschöpfung von der Mitte. F-Aktin Ringbildung ist eng an die Neuausrichtung der Mikrotubuli organisierenden Zentrum (MTOC, auch genannt die Centrosome in T-Zellen) an eine Position unterhalb der Mitte der Schnittstelle gekoppelt. Beide Veranstaltungen auftreten innerhalb von Minuten der erste Antigen-Anerkennung und den architektonischen Kontext, in dem nachfolgenden Aktivierung Veranstaltungen, den und Effektor Reaktionen auftreten, zu etablieren.
Um IS Bildung zu untersuchen, haben verschiedene Labors Ansätze entwickelt, in denen die APC durch eine Glasoberfläche, die ersetzt wird entweder immobilisierte Liganden TCR enthält oder ein Lipid Bilayer unterstützt, dass selbst die Liganden3,4enthält. T-Zellen bilden IS-ähnliche Kontakte auf diesen Flächen, die durch Totalreflexion Fluoreszenzmikroskop (TIRF) oder konfokalen Mikroskopie, hochauflösende Studien der frühen T-Zellaktivierung und IS Bildung abgebildet werden können.
Obwohl diese Ansätze für ausgezeichnete Visualisierung des fertig montierten IS erlaubt haben, tritt ein Großteil der Signalisierung folgende TCR:pMHC Ligatur innerhalb von Sekunden, erschweren die Bemühungen um die Abfolge der Ereignisse nach TCR Aktivierung genau bestimmen . Um dieses Problem zu umgehen, wurde ein photoaktivierungen Konzept entwickelt, in denen verwendet wird Photoactivatable pMHC, räumlich-zeitliche Kontrolle der TCR Aktivierung5,6,7zu erreichen. In diesem System sind T-Zellen Glasflächen mit Photoactivatable pMHC, die nicht-stimulierende, TCR bis Bestrahlung mit ultraviolettem (UV) Licht ist beigefügt. UV-Bestrahlung ein Mikrometer großen Bereich der Oberfläche unter der T-Zelle entfernt die Photocage schaffen eine anregende Zone, die von der T-Zellen erkannt werden kann. Ereignisse nach dem Bilanzstichtag Signalisierung und Zytoskeletts Umbau werden dann durch genetisch codierte fluoreszierende Reporter überwacht. Zwei Photoactivatable Versionen von Antigenen Peptide, Motte Cytochrom C88-103 (MCC) und Ovalbumin257-264 (OVA), die im Zusammenhang mit der Klasse II MHC-Ek und die Klasse ich MHC H2-Kb, bzw. vorgestellt werden, wurden entwickelt (Abbildung 1). Dies ermöglicht die Analyse der beiden CD4+ T-Zellen spezifisch für MCC-I-E-k (mit dem Ausdruck der 5 C. C7, 2B4, oder AND TCR) und CD8+ T-Zellen spezifisch für OVA-H2-Kb (mit dem Ausdruck der OT1 TCR).
Im letzten Jahrzehnt ist der TCR photoaktivierungen und Imaging-Ansatz, die präzise Kinetik der frühen TCR Signalisierung Schritte zu etablieren und zu identifizieren, die molekulare Wege polarisiert Zellskelett Umbau5, EZB verwendet worden 6 , 7 , 8 , 9 , 10. beispielsweise der Test war maßgeblich bei der Bestimmung, dass Centrosome Neuorientierung in Richtung der APC wird durch eine lokalisierte Steigung von Lipid zweite Bote Diacylglycerol zentriert auf den IS vermittelt. Es wird erwartet, dass diese Methodik wird weiterhin wertvoll für Anwendungen, die hochauflösenden bildgebenden Analyse des T-Zellfunktion zu fordern.

T-Lymphozyten engagieren sich in schnellen, polarisierte Signale, tritt innerhalb von Minuten nach der Aktivierung des TCR. Dies induziert Bildung der immunologischen Synapse, eine stereotype Zell-Zell-Kreuzung, die T-Zell-Aktivierung reguliert und direktional richtet sich an Effektor Antworten. Um diese Prozesse effektiv zu studieren, ist ein bildgebender Ansatz, der zugeschnitten ist zur Erfassung schneller, polarisierter Antworten notwendig. Dieses Protokoll beschreibt ein solches System, basiert auf einer Photoactivatable Peptid-Major Histocompatibility complex (pMHC), das nicht-stimulierende bis UV-Licht ausgesetzt ist. Gezielte decaging von diesem Reagens während Videomicroscopy Experimente ermöglicht präzise räumlich-zeitliche Steuerung der TCR-Aktivierung und hochauflösende Überwachung der anschließende zelluläre Reaktionen der Totalreflexion (TIRF) imaging. Dieser Ansatz ist auch kompatibel mit genetischen und pharmakologischen Störung Strategien. Dies ermöglicht die Montage von klar definierten Molekulare Signalwege, die TCR-Signalisierung link zur Bildung der polarisierten Zellskelett Strukturen, die die immunologische Synapse zugrunde liegen.

Der photoaktivierungen und Imaging Ansatz ermöglicht für Beobachtung und einfache Quantifizierung von rapid, polarisierte Signalisierung Antworten. Um seine Fähigkeiten, reproduziert zu illustrieren hier ist ein Experiment, die räumlich-zeitliche Korrelation zwischen TCR-induzierte DAG Akkumulation und Centrosome Neuorientierung zu prüfen. 5 C.
