Selv om disse metodene har tillatt for utmerket visualisering av ferdigmonterte er, oppstår mye av den signalnettverk følgende TCR:pMHC ligation innen sekunder, kompliserer innsats som avgjør rekkefølgen av hendelser etter TCR aktivisering nøyaktig . For å omgå dette problemet, er en photoactivation tilnærming utviklet, der photoactivatable pMHC brukes til å oppnå spatiotemporal kontroll av TCR aktivisering5,6,7.
