T-lymfocyter (T-celler) spelar en central roll i cellulär immunitet genom att erkänna antigen peptider visas i samband med cellytan MHC. Antigen erkännande, som medieras av TCR, driver differentiering av naiva T-celler och främjar leverans av cytolytisk och kommunikativa svar av effektor populationer. TCR interaktion inducerar också dramatiska förändringar i cellulära arkitektur. Inom minuter gloms T-cellerna på sidan av den antigen-presenterande cellen (APC), bildar en polariserad gränssnitt som kallas immunologisk synaps (IS)1,2. IS förstärker T cell effector Svaren genom att aktivera riktad frisläppandet av cytokiner eller, när det gäller cytotoxiska T-lymfocyter (CTL), lytisk proteiner som förstör APC.
TCR interaktion av pMHC inducerar den snabba fosforyleringen av flera nedströms adapter molekyler, inklusive Linker för aktivering av T-cellerna (LAT), som i slutändan främjar robust remodeling av synaptic cytoskelettet2. Kortikala fintrådiga aktin (F-aktin) driver T cell spridning över APC ytan, och sedan löser in en ringformig struktur som kännetecknas av F-aktin ackumulering vid IS periferi och utarmning från centrum. F-aktin ring bildandet är tätt kopplade till reorientationen av mikrotubuli organisera center (MTOC, även kallad centrosome i T-celler) till en position strax under mitten av gränssnittet. Båda händelserna inträffar inom minuter om inledande antigen erkännande och upprätta det arkitektoniska sammanhang i vilka efterföljande aktivering händelser och effektor Svaren används.
För att studera är bildning, har olika labs utvecklat metoder som APC ska ersättas med en glasyta som antingen innehåller immobiliserade TCR ligander eller stöder en lipid lipidens att själv innehåller ligander3,4. T-celler bildar IS-liknande kontakter på dessa ytor som kan avbildas av totalreflexion fluorescens Mikroskop (Frida) eller konfokalmikroskopi, aktivera högupplösta studier av tidig T cellaktivering och IS bildas.
Även om dessa metoder har tillåtit för utmärkt visualisering av färdigmonterad är, sker mycket av den signalering följande TCR:pMHC ligation inom sekunder, komplicerande ansträngningar för att fastställa händelseförloppet efter TCR aktiveringen exakt . För att kringgå problemet, har en ljusaktivering strategi utvecklats, där photoactivatable pMHC används för att uppnå spatiotemporal kontroll av TCR aktiveringen5,6,7. I detta system, är T-celler kopplade till glasytor som innehåller photoactivatable pMHC som är icke-stimulerande till TCR tills bestrålas med ultraviolett (UV) ljus. UV-bestrålning av en micron storlek region av ytan under den T-cellen tar bort den photocage att skapa en stimulerande zon som kan kännas igen av T-cellen. Två photoactivatable versioner av antigena peptider, moth cytokrom c88-103 (MCC) och äggalbumin257-264 (ägg), som presenteras i samband med klassen II MHC-Ek och klassen jag MHC H2-Kb, respektive, har varit utvecklade (figur 1). Detta gör analysen av både CD4+ T-celler specifika för MCC-jag-EKarlsson (som uttrycker den 5 C. C7, 2B4, eller och TCRs) och CD8+ T-celler specifika för ägg-H2-Kb (som uttrycker OT1 TCR).
