Sterilisatie van oppervlakken met behulp van lage-temperatuur, lagedruk plasma is een veelbelovend alternatief aan vrij conventioneel sterilisatie procedures zoals behandeling met ioniserende straling, chemische stoffen (zoals gassen als H2O2 of ethyleenoxide) of droge en vochtige warmte23. Gewone sterilisatie methoden bieden meestal een doeltreffende sterilisatie, maar ze zijn bekend om te beïnvloeden het behandelde materiaal en vertegenwoordigen een potentieel ris...

Een belangrijk doel van de verkenning van de ruimte is de zoektocht naar handtekeningen van levensvormen en biomoleculen op andere planetaire lichamen en manen in ons zonnestelsel. De overdracht van micro-organismen of biomoleculen van terrestrische oorsprong naar kritieke gebieden van exploratie is bijzonder risico bestaat om de impact van de ontwikkeling en de integriteit van leven-detectie missies op planetaire lichamen zoals Mars en Europa1. De internationale richtlijnen voor de bescherming van de planetaire, vastgesteld door het Comité voor ruimte onderzoek (COSPAR) in 1967, strikte regels op bemande en gerobotiseerde missies naar andere planeten, hun manen, asteroïden en andere hemellichamen opleggen en regelen de reiniging en sterilisatie van een ruimtevaartuig en essentiële hardwarecomponenten vooraf te lanceren om te elimineren besmetten terrestrische micro-organismen en het voorkomen van kruisbesmetting van hemellichamen2. In het afgelopen decennium, heeft de toepassing van niet-thermisch plasma's opgedaan brede aandacht in biomedische en nutritionele onderzoek, evenals in de bemande ruimtevaart toepassingen3,4,5. Plasma sterilisatie is een veelbelovend alternatief aan conventionele sterilisatie-methodes, want het biedt snelle en efficiënte microbiële inactivatie6, terwijl het zacht om gevoelige en hitte-labiele materialen. Plasma lozingen bevatten een mengsel van reactieve agenten zoals vrije radicalen, geladen deeltjes, fotonen in het ultraviolette (UV) en vacuüm ultraviolet (VUV) spectrum die tot snelle microbiële inactivatie3 leidenneutrale/opgewonden atomen. In deze studie, we gebruiken lagedruk plasma gegenereerd door dubbele inductief gekoppeld lagedruk plasma (DICP) bron-7,8 te inactivering van Bacillus subtilis endosporen verdeeld over test glasoppervlak.
Gram-positieve bacteriën van de familie Bacillaceae zijn op grote schaal verspreid in natuurlijke habitats van bodem, sedimenten en lucht evenals in ongewone omgevingen zoals schone Kamerfaciliteiten en de International Space Station9,10 ,11. De meest duidelijke functie van het geslacht Bacillus is de mogelijkheid om zeer resistent slapende endosporen (hierna "sporen" genoemd) om te overleven van ongunstige omstandigheden, zoals de uitputting van de voedende12vormen. Sporen zijn over het algemeen veel bestendiger dan hun tegenhangers van de vegetatieve cel naar een groot aantal behandelingen en milieu benadrukt, met inbegrip van warmte, UV, gamma bestraling, uitdroging, verstoring van de mechanische en giftige chemische stoffen, zoals sterke oxidatiemiddelen of pH-veranderende agenten (herzien in verwijzingen13,14) en zijn daarom ideaal objecten voor het testen van de efficiëntie van de microbiële inactivatie methoden. Aangezien genomic DNA een belangrijke doelstelling van de plasma behandeling van bacteriën15,16, de reparatie van plasma-geïnduceerde DNA laesies is (bijvoorbeeld dubbele bundel van DNA breekt) op spore opleving is van cruciaal belang voor het voortbestaan van de bacteriën13, 17.
Dus, we bestuderen de kiemkracht van de sporen en de rol van DNA-herstel tijdens spore ontkieming en uitgroei na behandeling van de sporen met lage druk argon plasma door volgende afzonderlijke sporen en hun uitdrukking van fluorescentie-gelabelde DNA herstellen eiwit RecA met time-resolved confocal fluorescentie microscopie. We geven een stap voor stap instructie van de voorbereiding van B. subtilis sporen in monolayers om reproduceerbare testresultaten, de behandeling van spore monolayers met lage druk plasma voor sterilisatie, de voorbereiding van plasma behandeld sporen voor Ultrastructurele evaluatie met behulp van scanning elektronen microscopie (SEM) en levende cel microscopie analyse op het niveau van individuele sporen in concert met het toezicht op de actieve DNA-herstelprocessen die zich voordoen binnen de cel op reactie op plasma behandeling.

investigating the detrimental effects of low pressure plasma sterilization on the survival of bacillus subtilis spores using live cell microscopy

Plasma sterilisatie is een veelbelovend alternatief voor conventionele sterilisatie methoden voor industriële, klinisch, en ruimtevaart doeleinden. Lagedruk plasma (LPP) lozingen bevatten een breed spectrum van actieve soorten, die tot snelle microbiële inactivatie leiden. Studeren op de efficiëntie en de mechanismen van sterilisatie door LPP, gebruiken we sporen van het testorganisme Bacillus subtilis vanwege hun buitengewone weerstand tegen conventionele sterilisatie procedures. We beschrijven de productie van B. subtilis spore monolayers, het sterilisatieproces door lagedruk plasma in een dubbele inductief gekoppeld plasma reactor, de karakterisatie van spore morfologie met behulp van scanning elektronen microscopie (SEM), en de analyse van kieming en uitgroei van sporen door levende cel microscopie. Een belangrijk doelwit van plasma soorten is genomische materiaal (DNA) en reparatie van plasma-geïnduceerde DNA laesies op spore opleving is van cruciaal belang voor het voortbestaan van het organisme. Hier, bestuderen we de capaciteit van de kieming van sporen en de rol van DNA herstellen tijdens spore ontkieming en uitgroei na behandeling met LPP door het bijhouden van fluorescently-gelabelde DNA reparatie eiwitten (RecA) met time-resolved confocal fluorescentie microscopie. Behandeld en onbehandeld spore monolayers zijn geactiveerd voor kieming en gevisualiseerd met een omgekeerde confocal levende cel Microscoop na verloop van tijd te volgen van de reactie van afzonderlijke sporen. Onze waarnemingen blijkt dat de Fractie van ontkiemen en ontgroei sporen afhankelijk van de duur van LPP-behandeling minimaal bereiken is nadat 120 s. RecA-YFP (gele fluorescentie eiwit) fluorescentie werd alleen in enkele sporen ontdekt en ontwikkeld in alle ontgroei cellen met een lichte verhoging in sporen LPP-behandeld. Bovendien, sommige van de vegetatieve bacteriën afgeleid van LPP-behandelde sporen sprake van een toename in cytoplasma en neiging lyse. De beschreven methoden voor de analyse van afzonderlijke sporen zou exemplarisch zijn voor de studie van andere aspecten van spore kieming en uitgroei.
