Sterilisering av flater med lav temperatur, lavt trykk plasma er et lovende alternativ til heller vanlig dampsterilisering som behandling med ioniserende stråling, kjemikalier (f.eks gasser som H2O2 eller etylenoksid) eller tørr og fuktig varme23. Overføring av mikroorganismer eller biomolecules Pǔtōnghuà til kritiske områder av leting er av spesiell risiko å påvirke av utvikling og livet-gjenkjenning oppdrag på planetenes organer som Mars og Europa1. Internasjonale retningslinjene for planetenes beskyttelse, etablert av komité for Space forskning (COSPAR) i 1967, innføre strenge regler på bemannet og robot oppdrag til andre planeter, deres måner, asteroider og andre himmellegemer og regulere den rengjøring og sterilisering et romskip og kritisk maskinvarekomponenter tidligere for å starte for å eliminere forurensende terrestriske mikroorganismer og forhindre kryss-kontaminering av himmellegemene2. Det siste tiåret, har anvendelse av ikke-termisk plasmas fått bred oppmerksomhet i biomedisinsk og ernæringsmessige forskning og romfart programmer3,4,5. Plasmasterilisering er et lovende alternativ til konvensjonelle sterilisering metoder som tilbyr rask og effektiv mikrobiell inaktivering6, samtidig som mild til sensitive og varme labil materialer. Plasma utslipp inneholder en blanding av reaktive stoffer som frie radikaler, ladede partikler, nøytral/glade atomer, fotoner i ultrafiolett (UV) og vakuum ultrafiolett (VUV) spektrum som fører til rask mikrobiell inaktivering3. I denne studien bruker vi lavt trykk plasma generert av dobbel Induktivt kombinert lavt trykk plasma (DICP) kilden7,8 for å deaktivere Bacillus subtilis endospores fordelt på test glassoverflaten.
Gram-positive bakteriene av familien Bacillaceae er svært utbredt i naturlige habitater av jord sedimenter og luften også som uvanlig miljøer som rene rom og den internasjonale romstasjonen9,10 ,11. Funksjonen mest tydelig i slekten Bacillus er evnen til å danne motstandsdyktige sovende endospores (heretter referert til som sporer) for å overleve ugunstige forhold, for eksempel næringsstoffer uttømming12. Sporene er generelt langt mer motstandsdyktig enn sine vegetative celle kolleger til en rekke behandlinger og miljømessige påkjenninger, inkludert varme, UV, gamma bestråling, uttørking, mekanisk avbrudd og giftige kjemikalier, som sterke oksidanter eller pH endring agenter (omtalt i referanser13,14) og er derfor ideelt objekter for å teste effektiviteten av mikrobielle inaktivering metoder. Dermed vi studere spiring kapasitet sporer og rollen til DNA reparasjon spore spiring og resultatet etter behandling spores med lavtrykk argon plasma av følgende personlige spores og deres uttrykk fluorescens-merket DNA reparasjon protein RecA med tid-løst fluorescens AC confocal mikroskopi. Vi gir en trinnvis instruksjon om utarbeidelse av B. subtilis sporer i monolayers for å oppnå reproduserbar testresultater, behandling av spore monolayers med lavtrykk plasma for sterilisering, utarbeidelse av plasma behandlet sporer for ultrastructural evaluering bruker skanning elektronmikroskop (SEM) og levende celle mikroskopi analyse på nivået av individuelle sporer sammen med overvåking aktive DNA reparasjon innenfor cellen i respons på plasma behandling.

investigating the detrimental effects of low pressure plasma sterilization on the survival of bacillus subtilis spores using live cell microscopy

Plasmasterilisering er et lovende alternativ til konvensjonelle sterilisering metodene for industrial, kliniske, og romfart. Lavtrykk plasma (LPP) utslipp inneholder et bredt spekter av aktiv arter, som fører til rask mikrobiell inaktivering. For å studere effektiviteten og mekanismer for sterilisering av LPP, bruker vi sporer av testen organismen Bacillus subtilis på grunn av deres ekstraordinære motstand mot konvensjonelle dampsterilisering. Behandlet og ubehandlet spore monolayers er aktivert for spiring og visualisert med en invertert AC confocal levende celle mikroskop over tid å følge reaksjonen av personlige sporer. Våre observasjoner viser at brøkdel av spirende og outgrowing sporer er avhengig av varigheten av LPP-behandlingen nå minst etter 120 s. RecA-YFP (gul fluorescens protein) fluorescens ble funnet i noen sporer og utviklet i alle outgrowing celler med en liten høyde i LPP-behandlet sporer. Videre noen av vegetative bakterier avledet fra LPP-behandlet sporer viste en økning i cytoplasma og tendens til å lyse. Overlevelse av plasma-behandlet B. subtilis sporer
