De meest kritische stap van dit protocol is het lijmen van het elektrode-pakket op het oppervlak van bot. In geval van onjuiste afdichting, een kloof ontstaat tussen de elektroden en het bot, en secundaire littekenweefsel kan groeien in deze leemte, die de kwaliteit van stimulatie vermindert. Het oppervlak van bot moet volledig droog zijn tijdens de stappen van de steken op de verpakking, en in het geval van ervaren instabiliteit van de elektroden, het moet worden verwijderd en vervangen door een nieuw pakket te krijgen ...

Transcraniële elektrische stimulatie (TES) is een waardevolle methodologische benadering hersenactiviteit beïnvloeden in een stoffelijk beperkte manier. Afhankelijk van de grootte en plaatsing van de elektroden van de stimulatie, TES kan invloed hebben op de volumes van de grote hersenen en later neuronale populaties diffuus1,2,3. Transcraniële gelijkstroom stimulatie is al medisch goedgekeurd voor de behandeling van depressieve stoornis4,5, en de vele studies focus op vertoon van de cognitieve effecten van Transcraniële stimulatie in mens6 , 7. Bovendien, veelbelovende resultaten gerapporteerd met betrekking tot het potentieel van TES beheersing van epileptische aanvallen8,9.
Ondanks de intensieve onderzoek zijn er nog veel open vragen met betrekking tot het gedetailleerde mechanisme van actie, mogelijke bijwerkingen en de lange termijn uitkomst van de toepassing van deze methode10,11,12. Daarom is het uitermate belangrijk om een robuuste, reproduceerbare protocol bij het onderzoeken van de gevolgen van TES in diermodellen. Gezien het feit dat vele aandoeningen (bijvoorbeeld, depressie, epilepsie en schizofrenie) kunnen alleen bij wakker dieren uitgebreid worden onderzocht, en de aard van deze medische aandoeningen vereisen meestal langdurige behandeling, bieden we een protocol voor chronische implantatie van Transcraniële elektroden bij ratten. De methode die hier gepresenteerd kan worden gebruikt voor behavioral studies of kan worden gecombineerd met implantatie van elektroden opname (d.w.z.draden, siliconen sondes, juxtacellular-elektroden) of met chronische craniale windows voor elektrofysiologische experimenten en beeldvorming studies, respectievelijk. Afhankelijk van de proefopzet kunnen de timing van de prikkels willekeurige of gebeurtenis naar specifieke gedragsmatige signalen of naar elektrofysiologische kenmerken van de bijzondere hersenen staten (vangsten, theta oscillaties)8, 11 , 13.
Het is belangrijk te vermelden dat in tegenstelling tot de momenteel gebruikte menselijke aanpak, die gebruik maakt van een belichaming van elektroden op de huid geplaatst, hier laten we zien een methode die gebruikmaakt van onderhuidse implantatie recht over het oppervlak van de temporale-bone wordt sinds ratten nauwelijks tolereren iets geplaatst op hun huid die gemakkelijk toegankelijk is via hun poten.
In overeenstemming met de beginselen van vervanging, vermindering en verfijning, het chronische karakter van implantatie helpt deze methode bij het verminderen van het aantal dieren, aangezien elk dier kan worden aangeworven in verschillende experimentele omstandigheden voor maanden, waardoor het gebruik van minder dieren om verschillende hypothesen te toetsen.
In de huidige studie, wij bieden een gedetailleerde, stapsgewijze protocol van Transcraniële stimulatie elektrode productie (figuur 1A-B) en de chronische inplanting van deze elektroden tonen over de temporale botten van een zes-maand-oude mannelijke Long-Evans rat.

Elektrische stimulatie transcranial (TES) is een krachtige en relatief eenvoudige benadering te diffuus hersenactiviteit beïnvloeden willekeurig of in een kringloopsysteem gebeurtenis manier. Hoewel veel studies zijn gericht op de mogelijke voordelen en bijwerkingen van TES in gezonde en pathologische hersenen, zijn er nog vele fundamentele open vragen met betrekking tot het werkingsmechanisme van de stimulatie. Daarom is er een duidelijke behoefte aan een robuuste en reproduceerbare methode voor het testen van de acute en de chronische effecten van TES bij knaagdieren. TES kan worden gecombineerd met regelmatige gedrags, elektrofysiologische en beeldvormende technieken te onderzoeken van de neuronale netwerken in vivo. De inplanting van Transcraniële stimulatie elektroden legt geen extra beperkingen op de proefopzet terwijl het biedt een veelzijdige, flexibele tool om te manipuleren van de hersenactiviteit. Hier voorzien wij een gedetailleerde, stapsgewijze protocol implantaat transcraniale stimulatie elektroden om hersenactiviteit beïnvloeden in een stoffelijk beperkte wijze maandenlang te fabriceren.

De inplanting van chronische stimulering elektroden (figuur 1B) kan worden gecombineerd met extra implantatie van elektroden (Figuur 1 c-D) opnemen. Dergelijke instellingen zijn geschikt om op aanvraag opname en stimulatie systemen te bemoeien met de activiteiten van de specifieke hersenen te vormen.
