Den kvinnelige skjede består av de øvre og nedre områder. Den øvre reproduktive område inkluderer Fallopian rør og livmoren i livmorhalsen. Den lavere skjede inkluderer ectocervix og vagina. Hos mennesker brukes utbrøt sæd på fremre veggen av skjeden, tilstøtende til ectocervix1. I mus, men er semen feiet inn i livmoren minutter etter mating2.
Inneholder sæd seminal gel som er styrket innen sekunder etter utløsning, forårsaker sæd å bli fanget og immobilisert3. LNG utgivelser immobilisert sæd ved å endre sæd fra en gel-lignende til en vannaktig konsistens. Denne prosessen er avgjørende for Spermiemotilitet og pattedyr reproduksjon. Kunnskap om sæd LNG er hovedsakelig basert på i vitro studier der sæd blir flytende i en kultur parabol. Hos mennesker er den enzymatiske aktiviteten av prostata spesifikke antigen eller PSA (også kjent som kallikrein-relaterte peptidase 3 eller KLK3) den viktigste bidragsyteren for LNG gjennom hydrolyse av semenogelins (gel-forming proteiner i sæd)4 ,5,6. Nedbrytning av semenogelins utvikles sperm fra den banebrytende coagulum, øke sæd mobilitet. Frigjort sperm svømme mot Fallopian tube å befrukte egget. LNG (eller sæd viskositet) testene er en av de standard første screenings for semen analyse i fruktbarhet klinikker å vurdere mannlig fertilitet7.
En nylig publisert artikkel viser at analyse av seminal væske fra kvinnelige musen skjede etter mating kan brukes til å illustrere en mangel på LNG som kan deretter føre fruktbarhet defekter8. Defekt LNG som sæd hyperviscosity bidrar til 11,8-32,3% av ufruktbar tilfeller i menn9, antyder at normal LNG er uunnværlig for pattedyr reproduksjon. Men har studier og behandling av LNG defekter fokusert utelukkende på mannlige7. Muligheten for at den kvinnelige skjede spiller en rolle i sæd LNG har ikke blitt utforsket. Derfor vil metodene for semen samling og måling LNG tid gi forskerne en roman Diagnoseverktøy for å fastslå om LNG kan være en av årsakene til ufruktbarhet i modellen organismen rundt.

I mus, settes utbrøt sæd inn i livmoren. Etter utløsning, sæd endres konsistens fra gel-lignende til vannaktig, en prosess som kalles LNG. I denne studien viser vi hvordan du samler etter utbrøt sæd fra den kvinnelige skjede i en musemodell. Første, voksen kvinnelig mus i estrus scenen ble plassert i en mannlig bur over natten. Neste morgen, bekrefter copulation tilstedeværelsen av copulatory plug ved skjedeåpningen. Sæd fra CTRL uteri tok 2.06 ±0.12 min (mean±S.E.M, n = 5) å fylle kapillarrør (figur 1). Imidlertid sæd fra KO uteri tok mer enn 60 min (60.0 ±0.0, mean±S.E.M, n = 5) eksperimentelle tid.
