Le modifiche e la risoluzione dei problemi:
L'obiettivo principale di questo studio era di descrivere un protocollo per l'utilizzo di un fixator esterno di MRI compatibili per osteotomia di femore nel topo con la possibilità di monitorare lo sviluppo del tessuto di callo longitudinalmente durante il processo di guarigione di frattura endochondral. La progettazione di un dispositivo di montaggio su misura per l'inserimento del fissatore esterno assicurato una posizione standar...

Guarigione della frattura secondaria è la forma più comune di guarigione ossea. È un processo complesso che imita gli aspetti specifici di ontogenic endochondral ossificazione1,2,3. L'ematoma di frattura precoce consiste principalmente di cellule immunitarie, granulazione e tessuto fibroso. Tensione dell'ossigeno basso e alti biomeccanici ceppi ostacolano la differenziazione degli osteoblasti allo spacco frattura, ma promuovono la differenziazione delle cellule progenitrici nei chondrocytes4,5,6. Queste cellule iniziano a proliferare il sito della lesione per formare una matrice cartilaginosa fornendo stabilità iniziale dell'osso fratturato. Durante la maturazione del callo, condrociti diventati ipertrofici, subiscono gli apoptosi, o trans-differenziarsi in osteoblasti. Neovascolarizzazione presso la zona di transizione della cartilagine-osso fornisce i livelli elevati dell'ossigeno, consentendo la formazione di tessuto osseo7. Dopo ossuta colmare il divario di frattura, è aumentata la stabilità biomeccanica e osteoclastica rimodellamento del callo di frattura esterna si verifica per aumento osseo fisiologico contorno e struttura3. Di conseguenza, la quantità di tessuto fibroso, cartilagineo e osseo nel callo di frattura forniscono informazioni importanti sul processo di guarigione dell'osso. Disturbato o ritardata guarigione diventa visibile da alterazioni dello sviluppo del tessuto di callo sia in esseri umani e topi8,9,10,11. Tecniche disponibili in vivo longitudinalmente monitorare lo sviluppo del tessuto di callo in preclinico frattura che guarisce gli studi utilizzando piccoli animali includono radiografia digitale e µCT12,13di imaging. Tuttavia, entrambe le tecniche sono solo in grado di discriminare tra tessuto mineralizzato e non mineralizzato. Al contrario, MRI fornisce tessuti molli eccellente contrasto e potrebbe quindi essere in grado di identificare il tessuto molle e cartilagine nel callo di frattura.
Il lavoro precedente ha mostrato risultati promettenti per post mortem MRI in topi con fratture articolari MRI14 ed in vivo in topi durante la guarigione del difetto osseo intramembranous15. Tuttavia, entrambi gli studi anche dichiarato contrasto di tessuto e risoluzione spaziale limitato. Precedentemente abbiamo dimostrato la fattibilità di MRI ad alta definizione in vivo per la valutazione longitudinale della formazione del callo molle durante la frattura endochondral murino guarigione16.
