Volgens de American Heart Association (2017), hartstilstand (CA)-geïnduceerde sterfte vindt elke vier minuten plaats, en beïnvloedt meer dan 400.000 mensen per jaar in de Verenigde Staten1. Het is goed gedocumenteerd dat CA neuronale hersenletsel als gevolg van onvoldoende bloed perfusie,2,,3,4kan veroorzaken. CA-geïnduceerde hersenletsel treedt op in de ischemie-gevoelige CA1-regio van de hippocampus5,6,7, op het gebied van neuronen die essentieel voor leren en geheugen8,9zijn, 10,11,12. Bovendien, het verlies van Dendritische spine dichtheid, ischemische omstandigheden in de hippocampus (d.w.z. CA1 neuronen), speelt een cruciale rol in het ruimtelijke geheugen bijzondere waardevermindering13,14,15. Als gevolg van deze pathologische veranderingen na CA, gedragsmatige stoornissen zoals: angst, depressie, posttraumatische stress-stoornis en geheugenverlies zijn vaker. De status van de cognitieve beoordeling voor cerebrale ischemie via gedrags proeven zijn belangrijk voor het bepalen van de werkzaamheid van beide drugs en uiteindelijk het ontwikkelen van een succesvolle klinische proef. In de jaren 1940 ontwierp Edward Tolman de eerste proef van het probleem te bestuderen van de ruimtelijke geheugen hippocampus gebaseerde18. Vervolgens werden verschillende doolhoven (d.w.z. De doolhof Barnes is de andere gebruikte gedrags test ruimtelijke leren en geheugen in knaagdier modellen te onderzoeken. De doolhof Barnes hoeft niet de inspanning van volledige motor functie en controle, en dus minder stress dan de Morris water maze. In het verleden hebben we uitgevoerd experimenten met behulp van de Barnes doolhof om te bepalen als functionele leren/geheugen verschillen tussen optreden controle of sham versus ACA-geïnduceerde ratten. Dus ontwikkelden we een gewijzigde T-maze met behulp van spontane afwisseling test om te beoordelen (korte termijn) geheugen na ACA. utilizing the modified t-maze to assess functional memory outcomes after cardiac arrest

Achtergrond: Evaluatie van milde tot matige cognitief stoornis in een globale cerebrale ischemie (d.w.z. hartstilstand)-model kan worden moeilijk te wijten aan slechte motoriek na de operatie. Bijvoorbeeld, ratten die chirurgische ingrepen ondergaan en zijn onderworpen aan de Morris water maze niet mogelijk om te zwemmen, het experiment dus ongeldig te verklaren.
Nieuwe methode: We vastgesteld een gemodificeerde gedrags spontane afwisseling T-maze test. Het grote voordeel van het gewijzigde T-maze-protocol is het relatief eenvoudig ontwerp dat krachtig genoeg is om te beoordelen van functionele leren/geheugen na ischemie. Bovendien is de data-analyse is eenvoudig en ongecompliceerd. We gebruikten de T-maze om te bepalen van de ratten leren/geheugen tekorten zowel in de aan- of afwezigheid van milde tot matige (6 min) asphyxial hartstilstand (ACA). Ratten hebben een natuurlijke neiging voor exploratie en zal de alternatieve armen in de T-maze, terwijl hippocampal-lesioned ratten de neiging te nemen een zijde-voorkeur wat resulteert in verminderde spontane afwisseling verhoudingen, onthullen de hippocampal-gerelateerde functioneel leren/geheugen in de aanwezigheid of afwezigheid van ACA.
Resultaten: ACA groepen hebben hogere kant-voorkeur-ratio's en lagere afwisseling in vergelijking met controle.
Vergelijking met bestaande methode(n): de Morris water en Barnes doolhof zijn meer prominente voor het beoordelen van leren/geheugenfunctie. De Morris water maze is echter meer belastend dan andere doolhoven. De Barnes-doolhof wordt veel gebruikt voor het meten van de verwijzing (lange termijn) geheugen, terwijl ACA-geïnduceerde Neurocognitieve tekorten meer verwant zijn aan het werken (korte termijn) geheugen.
Conclusies: We hebben een eenvoudige, maar effectieve strategie om het af te bakenen (korte termijn) geheugen via de T-maze werken in ons model voor wereldwijde cerebrale ischemie (ACA) ontwikkeld.

ACA (global cerebrale ischemie) veroorzaakt vooral werken (korte termijn) geheugen tekorten28,29.
