Selon l’American Heart Association (2017), un arrêt cardiaque (CA)-mortalité induite se produit toutes les quatre minutes et touche plus de 400 000 personnes par an dans les États-Unis1. Il est bien documenté que CA peut causer des blessures de neuronale cérébrale à la suite de sanguin insuffisant perfusion2,3,4. Lésion cérébrale induite par le CA se produit dans la région sensible à l’ischémie de CA1 de l’hippocampe5,6,7, touchant les neurones qui sont essentielles à l’apprentissage et la mémoire8,9, 10,11,12. En outre, la perte de densité de l’épine dendritique, dans des conditions ischémiques dans l’hippocampe (c.-à-d. les neurones CA1), joue un rôle essentiel dans la mémoire spatiale déficience13,14,15. En raison de ces changements pathologiques après CA, troubles comportementaux tels que : anxiété, dépression, stress post-traumatique et la perte de mémoire sont plus fréquents. Bien qu’il y a eu des progrès dans la technologie médicale (c'est-à-dire un service ambulatoire efficace) qui sont en corrélation avec les taux de survie améliorés CA, la plupart des traitements neuroprotecteurs (à l’exception d’hypothermie) ne parviennent pas à améliorer les résultats fonctionnels après CA16 ,,17. Généralement, les survivants de CA ont une mauvaise qualité de vie et sont accablés avec différentiel médical dépenses16.
Évaluations de la situation cognitive pour ischémie cérébrale par l’intermédiaire de tests comportementaux sont importantes pour déterminer les deux l’efficacité du médicament et en fin de compte développer un essai clinique concluant. Dans les années 1940, Edward Tolman a conçu le premier essai de comportement afin d’étudier la mémoire spatiale axée sur l’hippocampe,18. Par la suite, différents dédales (labyrinthe aquatique de Morris, labyrinthe radial, T - ou labyrinthe en Y et le labyrinthe Barnes) ont été élaborés pour évaluer l’hippocampe apprentissage spatial et la mémoire chez les rats19,20,21,22 ,,23. Un des essais comportementaux plus largement utilisé est le labyrinthe de l’eau de Morris, qui examine l’apprentissage spatial et mémoire chez le rat modèles24. Toutefois, le labyrinthe de l’eau de Morris exige le rat à nager et à exercer le contrôle et la pleine fonction. Pour des expériences telles que le modèle d’un arrêt cardiaque asphyxie (ACA, un modèle de rat de CA), ischémie canulation de l’artère fémorale/veine sont tenus d’obtenir artérielle essentielle, gaz du sang et l’introduction de divers médicaments. Puisque la canulation de l’artère fémorale/veine peut inhiber la mobilité de la jambe rendant de pouvoir le rat nager correctement, le labyrinthe de l’eau de Morris n’est peut-être pas le plus approprié tester des troubles cognitifs en vertu de l’ACA.
Le labyrinthe de Barnes est l’autre test comportemental largement utilisée pour examiner l’apprentissage spatial et la mémoire dans les modèles de rongeurs. Le labyrinthe de Barnes n’exige pas l’effort de motricité complète et le contrôle et donc moins stressant que le labyrinthe de l’eau de Morris. Dans le passé, nous avons effectué des expériences utilisant le labyrinthe Barnes pour déterminer si les différences fonctionnelles d’apprentissage/mémoire se produisent entre contrôle ou imposture que ACA-induite chez les rats. Les données obtenues pour le labyrinthe de Barnes n’avaient pas la résolution pour tester des déficiences cognitives suite légère à modérée ACA due au fait que le labyrinthe de Barnes est largement utilisé pour mesurer la mémoire de référence (long terme)25,26, tandis que Déficits neurocognitifs induite par l’ACA plus étroitement liés à l’utilisation (court terme) mémoire27,28,29,30 suggérant que le labyrinthe de Barnes est moins viable pour évaluer la fonction de mémoire dans notre ACA modèle.
Nous avons donc développé un labyrinthe en t modifié à l’aide spontanée alternance test pour évaluer le travail de mémoire (à court terme) après ACA. Avantage majeur du test modifié du spontanée alternance labyrinthe en t est sa simplicité et la contrainte minimale sur les rats par rapport à d’autres tests comportementaux dû au fait que le labyrinthe en t mis à jour le ne nécessite pas de formation préalable d’animaux, comme bien plus lourd computational analyse ou sous-programmes (c.-à-d. l’imagerie vidéo du rat) comme l’exige le labyrinthe aquatique de Morris et labyrinthe de Barnes. Nous démontrons que le critère d’alternance spontanée en labyrinthe en t modifié est un paradigme du procès comportemental simple et pourtant très efficace qui peut offrir une résolution suffisante pour détecter et évaluer la fonction hippocampe dans les maladies qui causent la perte de mémoire à court terme de manière précise (p. ex. ACA).

utilizing the modified t-maze to assess functional memory outcomes after cardiac arrest

Rappel des faits : Évaluation de légère à modérée troubles cognitifs dans un modèle d’ischémie cérébrale globale (c'est-à-dire un arrêt cardiaque) peut être difficile en raison de la mauvaise locomotion après la chirurgie. Par exemple, les rats qui subissent des interventions chirurgicales et le labyrinthe aquatique de Morris ne subissent peut-être pas capables de nager, annulant ainsi l’expérience.
Nouvelle méthode : Nous avons établi un test du labyrinthe en t mis à jour le comportement alternance spontanée. L’avantage majeur du protocole mis à jour le labyrinthe en t est sa conception relativement simple qui est assez puissante pour évaluer l’apprentissage/mémoire fonctionnelle après ischémie. En outre, l’analyse des données est simple et directe. Nous avons utilisé le labyrinthe en t pour déterminer les déficits d’apprentissage/mémoire rats tant en la présence ou l’absence de légère à modérée (6 min) asphyxie un arrêt cardiaque (ACA). Des rats ont une tendance naturelle à l’exploration et exploreront les bras remplaçant dans le labyrinthe en t, alors que les rats hippocampe-lésés ont tendance à adopter une préférence côté entraînant rapports une diminution spontanée alternance, révélant l’hippocampe-associés apprentissage/mémoire fonctionnelle en présence ou en absence d’ACA.
Résultats : Les groupes ACA ont des taux élevés de préférence de côté et alternances plus faibles par rapport au contrôle.
Comparaison avec les méthodes existantes : l’eau de la Morris et labyrinthe de Barnes sont plus proéminents d’évaluation des fonctions d’apprentissage/mémoire. Toutefois, le labyrinthe de l’eau de Morris est plus stressant qu’autres labyrinthes. Le labyrinthe de Barnes est largement utilisé pour mesurer la mémoire de référence (longue durée), tandis que les déficits neurocognitifs induite par l’ACA sont plus étroitement liés à l’utilisation mémoire (court terme).
Conclusions : Nous avons mis au point une stratégie simple mais efficace pour délimiter le travail de mémoire (court terme) via le labyrinthe en t dans notre modèle d’ischémie cérébrale globale (ACA).
