In aanwezigheid van serum, die een rijke bron van Pro-lijm extracellulaire matrix moleculen zoals fibronectin is, zal allantois explantaten gemakkelijk hechten aan verschillende kunststof, glas of9,16van de oppervlakken van de filter. Als het doel van de uitgevoerde test is te specifiek onderzoeken van het effect van een genetische modificatie of iedere andere behandeling op allantois gehechtheid aan geïmmobiliseerd α4β1, is het dus cruciaal voor het blokkeren...

De placenta is onmisbaar voor embryonale groei en ontwikkeling in de baarmoeder omgeving. Het vormt de interface voor zuurstof, metaboliet, en voedende gegevensuitwisseling tussen de foetale en maternale circulatie, en ook functies als endocriene en immunologisch orgaan. Een endogene of exogene belediging dat belemmert de ontwikkeling van de placenta kan leiden tot uteriene groei retardatie, foetale verlies of zwangerschapscomplicaties, zowel in mens1in muizen. Terwijl het aantal gerichte muis mutaties die leiden abnormale placenta fysiologie tot blijft toenemen2, met 219 genotypen momenteel vermeld op de website van de muis genoom informatica (MGI) (http://www.informatics.jax.org/vocab/ mp_ontology / MP:0010038), veel van deze fenotypen zijn niet goed wordt begrepen vanuit een mechanistische oogpunt. Naast genetische veranderingen, klein molecuul drugs, monoklonale antilichamen, toxinen, ziekteverwekkers, overtollige metabolieten of verschillende milieu-agentia kunnen invloed op de ontwikkeling van de placenta en functie3,4,5 , 6 , 7. toch eenvoudig in vitro testen dat model kritische stappen in de ontwikkeling van de placenta schaars zijn.
Lymfkliertest placenta ontwikkeling wordt gestart in de vroege midgestation, wanneer de allantois (de ontwikkelingstoxiciteit voorloper van de navelstreng) komt naar voren uit het posterieure einde van het embryo als een knop die naar de chorion-8 groeit. Chorioadhesive cellen in de buitenste laag van de allantoïs kiem bemiddelen de gehechtheid en de verspreiding van de allantois op het oppervlak van het chorion (chorion "fusion"). De chorion vouwen vervolgens in villi waarin de therapieën van de allantois groeit om te vormen van het labyrint, de placenta binnenlaag waar voedingsstoffen en zuurstof uitgewisseld tussen dicht naast elkaar geplaatste moederlijk bloed sinusoïdes en embryonale vaartuigen9 ,10.
De bevestiging van de allantois aan de chorion is de eerste en essentiële stap in labyrint vorming en gebreken in dit proces behoren tot de meest voorkomende oorzaken van embryonale letaliteit in midgestation1. Hoewel een aantal mutanten muis hebben beschreven waar chorion gehechtheid mislukt optreden van10en de MGI-database bevat momenteel 108 muis mutanten die worden gekenmerkt door abnormale chorion fusion (http:// www.Informatics.Jax.org/vocab/mp_ontology/MP:0002824), de intercellulaire interactie tussen de vasculaire cel adhesie molecuul-1 (VCAM-1, uitgedrukt op het allantoïs mesoderm) en α4β1 integrine (uitgedrukt op de chorionic mesothelium, die is afgeleid van extraembryonic mesoderm) lijkt te zijn onmisbaar voor het chorion gehechtheid en fusion11,12,13. Immunohistochemische kleuring van geheel-mount wild-type embryo's is gebleken dat VCAM-1 wordt uitgedrukt in de distale tweederde van de allantoïs stengel op ongeveer embryonale dag (E) 7,513 (1-4 Somiet fase), en dat haar expressie hoog blijft tijdens het chorion gehechtheid en -fusion11,12. Gebrek allantoïs gehechtheid aan het chorion wordt normaal gesproken gedetecteerd door histologische analyses van baarmoeder toppen. Echter, allantois maten zijn fysiologisch zeer variabel in tussen, en zelfs binnen de nesten van de dezelfde ontwikkelingsstadium14, en de allantois kunt alleen toevoegen aan de chorion wanneer het een voldoende omvang te maken van fysiek contact heeft bereikt. Als gevolg van deze natuurlijke variabiliteit, chorion bijlage vindt plaats ergens tussen ~ E 8.0 en 9.0 E in uteroen een statistisch betrouwbare evaluatie van dit proces door histologie is daarom afhankelijk van de analyse van een groot aantal conceptuses verkrijgen van voldoende exemplaren op de juiste ontwikkelingsstadium.
Hier beschrijven we een methode om te beoordelen allantoïs bijlage ex utero dat is minder afhankelijk van de grootte van de allantois. We tonen de dissectie van muis embryo's en hun allantoides uit de baarmoeders van zwangere muizen ~ 8 dagen post coitum (dpc; dpc 0,5 geeft de detectie van een vaginale plug), en de daaropvolgende cultuur van allantois explants op geïmmobiliseerdet α4β1 Integrins. Deze methode maakt een snelle, functionele evaluatie van de bijlage en het verspreiden van gedrag van meerdere allantois explantaten parallel en op verschillende opeenvolgende tijdstippen. Het protocol kan worden gebruikt aan het scherm voor de gevolgen van gerichte mutaties, drugs of verschillende omgevingsfactoren voor allantois bijlage ex vivo.

De placenta is essentieel voor de groei en ontwikkeling van zoogdieren embryo's. Om deze reden vele genetische veranderingen en waarschijnlijk ook milieu beledigingen die verstoren de ontwikkeling van de placenta of functie kunnen leiden tot vroege zwangerschap verlies bij muizen en mensen. Niettemin, is het eenvoudig in vitro testen om te screenen op de mogelijke gevolgen voor de vorming van de placenta ontbreken. Hier, concentreren we op het modelleren van de eerste en essentiële stap in de vorming van de placenta, die uit de gehechtheid van de allantois aan het chorion bestaat. Beschrijven we een methode om snel beoordelen van de gehechtheid van allantoïs explantaten op geïmmobiliseerdet α4β1 integrine, die als een chorio-mimetische substraat fungeert.
