. בדקנו את הכדאיות של התומכים ב- WGS הקרנה של מוטציות הקשורות תרופתיים לסוגים שונים של סוכני פטריות. אנו מציגים מתודולוגיה ליישום WGS של משתמש הקצה ופרספקטיבות מעבדה מיקולוגיה אבחון. אנו כלולים בניתוח זה לבודד שלושה זוגות תרבותי של שלושה מקרים קליניים נפרדות אשר במבחנה ההתנגדות echinocandins ו- 5-flucytosine שפותחו לאורך זמן בעקבות טיפול נגד פטריות.

whole genome sequencing of candida glabrata for detection of markers of antifungal drug resistance

קנדידה glabrata יכול לרכוש במהירות מוטציות שגורמות עמידות לתרופות, במיוחד אל azoles ואל echinocandins. זיהוי של מוטציות גנטיות חיוני, כמו ההתנגדות זוהה במבחנה ניתן לעיתים קרובות בקורלציה עם כשלון קליני. בדקנו את הכדאיות של שימוש רצף הגנום כולו (WGS) לניתוח ברמת הגנום של עמידות לתרופות נגד פטריות ב- glabrata ג. המטרה הייתה torecognize נחותות ומחסומים ביישום WGS ומדידת יעילותו. מאמר זה מתאר את בקרת איכות מפתח מחסומים ואת מרכיבים חיוניים של מתודולוגיה WGS לחקור גנטיים המשויכים הרגישות מופחתת סוכני פטריות. זה גם מעריך את הדיוק של ניתוח נתונים, סיבוב סביב הזמן של בדיקות.

לסיכום, WGS של glabrata ג היה ריאלי בגילוי משמעות קלינית מוטציות מעורב ההתנגדות סמים פטריות שונות שיעורים ללא צורך מרובים PCR/DNA רצף תגובות. זה מייצג צעד חיובי לקראת הקמת WGS יכולת במעבדה קלינית לגילוי סימולטני של החלפות ומעניקה עמידות נגד פטריות.
