Her demonstrere vi brugen af CRISPR/Cas9 gen redigering til at studere den biologiske funktion af CALR mutationer i hæmatopoietisk celler. CRISPR/Cas9 teknologi har for nylig revolutionerede målrettede genom-redigering i levende celler og organismer. Det er blevet et meget kraftfuldt værktøj til biomedicinsk forskning og er i øjeblikket ved at blive udnyttet som en potentiel avenue til behandling af genetiske sygdomme1. Grundlaget for alle genom redigeringsværktøjer bygger på oprettelsen af en nukleasen-induceret DNA dobbelt strandede pause (DSB) på den genomisk locus skal ændres. DSBs kan repareres af ikke-homologe ende-sammenføjning (NHEJ) eller homologi-instrueret reparation (HDR)2,3. Fordelen ved Cas9 nukleasen over andre genom engineering nukleaser, såsom zink finger nukleaser (ZFNs) og transkription aktivere-lignende effektor nukleaser (TALENs) er dens afhængighed af RNA for målretning nukleasen til en ønskede DNA sekvens, sammenlignet at de protein-DNA interaktioner findes i ZFNs og TALENs2,3.
Efter opdagelsen af CRISPR/Cas9 nukleasen pathway som en adaptive immunsystem i Prokaryote celler4,5, er meget indsats gået ind i tilpasning pathway til brug i pattedyr cellelinjer og model organismer2, 3. som et værktøj til redigering af genet, CRISPR/Cas9 pathway anvender to hovedkomponenter: Streptococcus pyogenes (Sp) afledte Cas9 nukleasen og sgRNAs rettet mod gen interesse2,3. SgRNA består af 20 nukleotider, direkte Cas9 til et bestemt websted på genom RNA-DNA basepar komplementaritet2,3. Mål-webstedet af sgRNA skal ligge ved siden af en protospacer tilstødende motiv (PAM) site i form af 5' NGG, som er anerkendt af SpCas9 nukleasen. Med disse værktøjer, kan Cas9 rettes til enhver DNA sekvens ved at designe sgRNAs der er rettet mod det pågældende område. Derudover at Sp afledt Cas9, er der yderligere varianter for Cas9 med forskellige funktioner afhængigt af den konkrete anvendelse. I denne protokol kombinerer vi CRISPR/Cas9 metode med Ba/F3 cellulær transformation analysen til at forstå den biologiske funktion af CALR mutationer. BA/F3 celler er en murine IL-3 afhængige hæmatopoietisk cellelinje, der kan gengives IL-3 uafhængige ved udtryk for visse onkogener såsom BCR-ABL12. For at forstå om mutant calreticulin kan omdanne Ba/F3 celler til cytokin uafhængige vækst, vi målrettet exon 9 af den endogene Calr locus ved hjælp af CRISPR/Cas9 for at indføre indel mutationer og derefter trak IL-3 fra cellerne til at anvende en positiv udvælgelse pres, med mål at recapitulating gevinst af funktion CALR mutationer findes i MPN patienter. Protokollen omfatter design, kloning og levering af sgRNAs, udvikling af stabile Cas9 udtrykker celler og screening for CRISPR på-target gen redigering. Grupperet regelmæssigt interspaced korte palindromiske gentagelser (CRISPR) er en adaptive immunitet system i prokaryoter, der har været repurposed af forskere til at generere RNA-styrede nukleaser, såsom CRISPR-forbundet (Cas) 9 for lokationsspecifikke eukaryote genom redigering. Genom engineering af Cas9 bruges effektivt, nemt og håndfast ændre endogene gener i mange biomedically relevante pattedyr cellelinjer og organismer.
