De aanpasbare microfluidic biofilm assay hier beschreven staat voor de visualisatie van biofilm vorming in real time op het niveau van een eencellige wanneer blootgesteld aan een vaste rente laminaire flow en constante temperatuur. Candida albicans is een commensale lid van de menselijke microbiota, maar het is ook een opportunistisch pathogeen, kan veroorzaken oppervlakkig en ernstige schimmelinfecties1,2. Een kenmerk van de grote virulentie van C. albicans is haar vermogen om vorm veerkrachtig en resistente biofilms, Gemeenschappen van cellen vastgehouden aan een oppervlak en omsloten door een extracellulaire matrix materiële1,3. C. albicans biofilms zijn zeer gestructureerd, met verschillende lagen van meerdere celtypen (ronde ontluikende gist-formulier cellen, ovale pseudohyphal cellen en tubulaire hyphal cellen)4. C. albicans biofilm ontwikkeling begint met de hechting van de cellen van de ronde gist-vorm op een oppervlak (zaaien de biofilm), gevolgd door de verspreiding van deze cellen op het oppervlak, en vervolgens de rijping van de onrijpe biofilm structureert in een volledig gevormde biofilm die is omgeven door extracellulaire matrix materiële4. De volwassen biofilm is voornamelijk samengesteld uit langgerekte hyphal cellen die dichte en onderling verbonden netwerken vormen, die de architecturale stabiliteit met de biofilm4. Gedurende de hele levenscyclus van biofilm, ronde ontluikende gist cellen verspreiden van de volwassen biofilm, en kunnen reizen naar andere regio's van het lichaam veroorzaken gedissemineerde infecties of zaad nieuwe biofilms op andere sites4,5. C. albicans kan biofilms vormen op biotische oppervlakken, zoals mucosal oppervlakken en in de gehele weefsel van de gastheer, en op abiotische oppervlakken, zoals katheters, pacemakers, kunstgebitten en prothetische gewrichten. Als gevolg van de weerspannige eigenschappen van biofilms, ze zijn uiterst moeilijk uit te roeien, en in veel gevallen is de enige effectieve behandeling strategie verwijderen van de geïnfecteerde apparaat4. Het is dus van cruciaal belang voor het onderzoeken van biofilm vorming onder omstandigheden die vergelijkbaar zijn met die waargenomen in klinische instellingen.
Er zijn verschillende kritische in vivo dierlijke modellen gebruikt bij het bestuderen van C. albicans biofilm vorming6,7,8; echter, deze studies kunnen worden duur, tijdrovend, en worden beperkt door het aantal stammen en antimicrobiële stoffen die kunnen worden getest op een bepaald moment. In vitro biofilm testen, aan de andere kant, toestaan voor de beoordeling van het snelle, hoge-doorvoer van antischimmel verbindingen en gemuteerde stammen, en zijn veel meer kosteneffectief en ethische dan biofilm testen uitgevoerd out in dierlijke modellen9, 10,11,12,13,14. Hier beschrijven we een in vitro assay die we ontwikkeld en geoptimaliseerd voor het observeren van biofilm vorming stoffelijk onder laminaire stroom met behulp van een aanpasbare microfluidic apparaat14,15. De bepaling voorziet in de visualisatie van elke fase van de vorming van de biofilm, met inbegrip van de naleving van de eerste stap, celproliferatie biofilm rijping en spreiding van de cel. De bepaling is ook handig om te visualiseren cel morfologie wijzigingen in de gehele ontwikkeling van een biofilm.
Microtiterplaat platen, die meestal worden gebruikt voor in vitro biofilm testen, terwijl de hoge doorvoer, toegestaan niet voor gecontroleerde stromingscondities. Traditionele laminaire flow cel systemen voor de continue beoordeling van biofilm vorming in gecontroleerde stromingscondities toestaan, maar deze zijn vaak tijdrovend te zetten en hebben de neiging te hebben beperkt dynamisch bereik controle en doorvoer. Het microfluidic-apparaat gebruikt hier overwint deze beperkingen door een combinatie van hoge-doorvoer platen (met 48 putten) met een ingebouwde laminaire flow kamer en zeer reproduceerbaar, veelzijdig en aanpasbaar.
Hier beschrijven we een protocol voor het gebruik van een commercieel beschikbare geautomatiseerde microfluidic apparaat te beoordelen van biofilm vorming van een wild-type C. albicans stam, de gevolgen van een bekende antimycoticum voor de ontwikkeling van een biofilm, en de biofilm vorming in twee gemuteerde stammen (bcr1Δ/Δ en efg1 Δ/Δ), die eerder werden gemeld dat biofilm gebreken in vitro en in vivo16,17,18. De beschreven protocol kan worden gebruikt voor het testen van de werkzaamheid van antimicrobiële stoffen in de remming van biofilm vorming in de gehele ontwikkeling van een biofilm, en om genen nodig zijn voor normale biofilm ontwikkeling door screening mutant bibliotheken te identificeren.

Candida albicans is de meest voorkomende schimmel pathogeen van de mens, veroorzaakt ongeveer 15% van de gevallen van de ziekenhuizen opgelopen sepsis. Een kenmerk van de grote virulentie van C. albicans is haar vermogen om vorm biofilms, gestructureerde gemeenschappen van cellen die zijn gekoppeld aan de biotische en abiotische oppervlakken. C. albicans biofilms kunnen vormen op host weefsels, zoals mucosale lagen, en op medische apparaten, zoals katheters, pacemakers, kunstgebitten en gewrichtsprotheses. Biofilms vormen belangrijke klinische uitdagingen omdat ze zeer resistent tegen fysische en chemische verstoringen zijn, en fungeren kunnen als reservoirs op zaad dat wordt verspreid infecties. Verschillende in vitro testen hebben gebruikt om te bestuderen van de vorming van het C. albicans biofilm, zoals microtiterplaat plaat testen, metingen van het droge gewicht, cel levensvatbaarheid testen en confocale scanning laser microscopie. Al deze testen zijn één eindpunt testen, waar de biofilm vorming is beoordeeld op een specifiek tijdstip. Hier beschrijven we een protocol om te bestuderen van biofilm vorming in real-time met behulp van een geautomatiseerde microfluidic apparaat onder laminaire stromingscondities. Deze methode zorgt voor de waarneming van biofilm vorming zoals de biofilm na verloop van tijd ontwikkelt, met behulp van aanpasbare voorwaarden die die van de host, zoals die ondervonden in vasculaire katheters nabootsen. Dit protocol kan worden gebruikt ter beoordeling van de tekortkomingen van de biofilm van genetische mutanten alsmede de remmende effecten van antimicrobiële agentia over biofilm ontwikkeling in real-time.

Wij uitgevoerd de microfluidic biofilm assay beschreven hier met behulp van een wild-type C. albicans stam onder twee media voorwaarden (RPMI-1640 en Spider media), de stam van de wild-type in het bijzijn van de bekende antischimmel drug amfotericine B (16 µg/mL) in RPMI, en twee gemuteerde stammen eerder gemeld dat gebreken in de formatie van de biofilm (bcr1Δ/Δ en efg1 Δ/Δ) in Spider media.
