Bidrag af en neuron netværk aktivitet er i vid udstrækning bestemt af den dynamiske natur af synaptic indgange det modtager. Traditionelt har påberåbt den fremherskende metode til kendetegner synaptic aktivitet i neuroner somatiske hele-celle patch-clamp optagelser af postsynaptiske strømninger fremkaldt af elektrisk stimulation af axoner af passage. Dog er kun aktiviteten af proksimalt beliggende synapser sandfærdigt rapporteret i denne sag1. Derudover for at vurdere synapse-specifikke mekanismer, har optagelser fra par af neuroner og fra dendritter på en bestemt placering været anvendt til at målrette synapser af interesse og mekanismer for dendritiske integration, henholdsvis. Et stort gennembrud inden for synaptic fysiologi blev opnået gennem et ægteskab af optiske og elektrofysiologiske teknikker. To-foton excitation laser scanning mikroskopi (2PLSM) i kombination med Ca2 + imaging og optogenetic værktøjer kan afsløre enorme detaljer af den dynamisk organisation af synaptic aktivitet på bestemte neuronal forbindelser i hjerne skiver i vitro og i vivo.
Flere vigtige fordele lavet 2PLSM skiller sig ud fra de konventionelle, one-foton excitation mikroskopi2: (1) som følge af en ikke-lineære karakter af to-foton excitation, fluorescens genereres kun i den fokale volumen, og alle udsendte fotoner repræsenterer nyttige signaler (ikke behov for pinhole); (2) længere bølgelængder, bruges i 2PLSM, trænge spredning væv mere effektivt; Derudover er spredte fotoner også fortyndes til at producere to-foton excitation og baggrunden fluorescens; (3) solskader og fototoksicitet er også begrænset til brændplanet. Derfor, trods den høje pris på 2-foton systemer i forhold til konventionelle Konfokal mikroskoper, 2PLSM stadig en metode til høj opløsning undersøgelse af neuronal struktur og funktion i tyk levende væv.
Den første anvendelse af 2PLSM i spredning væv var. struktur og funktion af dendritiske pigge3billedet 2PLSM i kombination med Ca2 + imaging har afsløret at pigge fungerer som isolerede biokemiske rum. Da der er en en til en-korrespondance mellem pigge og individuelle synapser4i mange neuronal typer blev to-foton Ca2 + imaging snart en nyttig værktøj rapportering aktivitet af individuelle synapser i intakt væv5, 6,7,8. Desuden blev 2PLSM-baserede Ca2 + imaging med succes brugt til at overvåge aktiviteten på enkelt calcium kanaler og ikke-lineære samspillet mellem de iboende og synaptic conductances, samt at vurdere stat - og aktivitet-afhængige regulering af Ca2 + signalering i neuronal dendritter5,6,7,8,9,10,11.
Calcium er en allestedsnærværende intracellulære anden budbringer, og dens subcellulært spatiotemporelle organisation bestemmer retningen af fysiologiske reaktioner fra ændringer i synaptisk styrke på reguleringen af ion-kanaler, dendrit og rygsøjlen vækst, som samt celledød og overlevelse. Dendritiske Ca2 + stigninger opstår via aktivering af flere veje. Handling potentialer (APs), backpropagating til dendritter, åbne spænding-gated calcium kanaler12 og producerer relativt globale Ca2 + transienter (katte) i dendritter og pigge13. Synaptisk transmission er forbundet med aktivering af postsynaptiske Ca2 +-gennemtrængelige receptorer (NMDA, Ca2 +-gennemtrængelige AMPA og kainate), udløse synaptic katte6,14,15. Endelig kan supralinear Ca2 + begivenheder genereres i dendritter under visse betingelser11,12,13,16.
To-foton Ca2 + imaging i kombination med patch-clamp elektrofysiologiske optagelser beskæftiger syntetiske Ca2 +-følsomme fluorescerende indikatorer, der typisk leveres gennem patch elektrode under hele-celle optagelser . En standard metode til kvantificering af Ca2 + dynamics er baseret på dual indikator metode17,18. Det bruger to fluorophores med godt adskilt emission spektre (f.eks, en kombination af en rød Ca2 +-ufølsom farvestof med grønne Ca2 + indikatorer, såsom Oregon Green BAPTA-1 eller Fluo-4) og har flere fordele sammenlignet med den enkelt indikator metode. Første, en Ca2 +-ufølsom farvestof bruges til at finde små strukturer af interesse (dendritiske grene og pigge) hvor Ca2 + billedbehandling vil blive udført. For det andet beregnes forholdet mellem ændringen i grønne og røde fluorescens (ΔG/R) som en foranstaltning af [Ca2 +], som er i vid udstrækning ufølsom over for ændringer i baseline fluorescens på grund af udsving i [Ca2 +]017 , 18. Endvidere fluorescens ændringer kan kalibreres med hensyn til absolutte Ca2 + koncentrationer19.
En generel bekymring, når du kører to-foton Ca2 + imaging eksperimenter i akut skiver er celle sundhed og stabilitet af image erhvervelse på grund af den høje laser power typisk bruges. Derudover i Ca2 + imaging eksperimenter er der bekymring om undertrykkelse af netbårne og betydelig overvurdering af subcellulært Ca2 + dynamics at Ca2 + indikatorer fungere som meget mobile eksogene Ca2 + buffere. Således valget af Ca2 + indikator og dets koncentration afhænger af typen neuronal, den forventede amplitude af katte, og den eksperimentelle spørgsmål.
Vi tilpasset to-foton Ca2 + billedbehandling metode til undersøgelse af Ca2 + udsving i dendritter GABAergic interneurons9,10,11,20,21 , 22. mens hovedparten af tidlige Ca2 + billeddiagnostiske undersøgelser blev udført i vigtigste neuroner, hæmmende interneurons fremvise en lang række funktionelle Ca2 + mekanismer, der adskiller sig fra dem i pyramideformet celler20 , 23 , 24. disse interneuron-specifikke mekanismer (fx, Ca2 + gennemtrængelig AMPA-receptorer) kan spille bestemte roller i regulerer celleaktivitet. Mens dendritiske Ca2 + signalering i interneurons er et fristende mål for yderligere undersøgelse, giver to-foton Ca2 + imaging i dendritter af disse celler yderligere udfordringer, fra en tyndere diameter af dendritter og mangel på pigge til en særlig høj endogene Ca2 + bindende kapacitet. Som vores forskningsinteresser fokuserer på studiet af hippocampus interneurons, blev følgende protokol, mens gælder for forskellige neuronal populationer, tilpasset til at håndtere disse udfordringer.
Denne protokol blev udført med en kommerciel Konfokal to-foton mikroskop, som var udstyret med to eksterne, ikke-descanned detektorer (NDDs), elektro-optiske modulator (EOM) og et Dodt scanning gradient kontrast (SGC), og installeret på en optisk tabel. Mikroskopet blev kombineret med en Ti:Sapphire multiphoton laser mode-låst på 800 nm (> 3 W, 140 fs pulser, 80 Hz gentagelseshyppighed). Den imaging system var udstyret med en standard Elektrofysiologi rig, herunder en perfusion kammer med temperaturkontrol, en oversætte platform med to micromanipulators, en computer-styrede mikroelektrode forstærker, en digitizer, en stimulation enheden, og data erhvervelse software.

two-photon calcium imaging in neuronal dendrites in brain slices

Calcium (Ca2 +) imaging er et kraftfuldt værktøj til at undersøge spatiotemporelle dynamikken i intracellulære Ca2 + signaler i neuronal dendritter. Ca2 + udsving kan opstå gennem en række membran og intracellulære mekanismer og spiller en afgørende rolle i induktion af synaptisk plasticitet og regulering af dendritiske ophidselse. Derfor, evnen til at registrere forskellige typer af Ca2 + signaler i dendritiske grene er værdifulde for grupper at studere, hvordan dendritter integrere oplysninger. Fremkomsten af to-foton mikroskopi har foretaget sådanne undersøgelser betydeligt lettere ved at løse problemerne forbundet med billedbehandling i levende væv, såsom lysspredning og solskader. Derudover gennem kombinationen af konventionelle elektrofysiologiske teknikker med to-foton Ca2 + imaging, er det muligt at undersøge lokale Ca2 + udsving i neuronal dendritter parallelt med optagelser af synaptic aktivitet i Soma. Vi beskriver her, hvordan du bruger denne metode til at studere dynamikken i lokale Ca2 + transienter (katte) i dendritter af GABAergic hæmmende interneurons. Metoden kan også anvendes til at studere dendritiske Ca2 + signalering i forskellige neuronal typer i akut hjernen skiver.

Ved hjælp af protokollen præsenteres her, opnåede vi katte fremkaldt af somatiske nuværende injektion og elektrisk stimulation i dendritter af oriens/alveus interneurons i området CA1 hippocampus. Efter lappe en neuron, identificeret baseret på dens form og stilling, vi overtog linescans på tværs af en proksimal dendrit på flere punkter på givet afstande fra soma (figur 1A).
