We ontwierpen een protocol met behulp van een diskette met hoge dichtheid peptide microarray met de gehele Zika virus eiwit sequentie (Frans-Polynesische stam). Zika virus (ZIKV) diagnose op basis van klinische symptomen is uitdagend omdat het deelt vectoren, geografische spreiding en symptomen met Dengue en Chikungunya virus infectie1. Gezien het risico voor ongewenste zwangerschap resultaten bij vrouwen geïnfecteerd met ZIKV tijdens de zwangerschap, is het belangrijk om te onderscheiden tussen de 3 virussen. Hoewel de huidige moleculaire diagnostische tests specifiek zijn, zijn ze alleen nuttig in bloed of speeksel in de relatief korte periode van acute infectie2,3. Serologische tests zijn essentieel voor de diagnose buiten deze beginperiode van infectie4.
De ontwikkeling van een specifieke serologische test ZIKV is een uitdaging om twee redenen: ten eerste de Zika-antigenen die het menselijke immuunsysteem reageert op niet op dit moment bekend zijn; en tweede, geconserveerde flavivirussen aminozuur sequenties veroorzaken antilichaam Kruisallergie. Ons doel was om te ontdekken unieke ZIKV specifieke peptiden in diagnostiek worden gebruikt. Verschillende benaderingen zijn ontwikkeld om scherm peptide bibliotheken die betrekking hebben op hele eiwitten met inbegrip van phage, bacteriële, en gist oppervlak weer5,6,7,8,9, 10. Onze strategie was het gebruik van een hoge dichtheid peptide microarray waarmee snelle en goedkope high-throughput serologische screenings11,12 en nadien de geïdentificeerde peptiden kunnen worden gebruikt ter verbetering van de huidige serologische tests voor de opsporing van ZIKV infectie.
Dit protocol maakt de ontdekking van Zika virus specifieke diagnostische peptiden met behulp van een diskette met hoge dichtheid peptide microarray (Figuur 1). De high-density peptide microarray werd geproduceerd met behulp van de peptide laser printing technologie. De gehele ZIKV eiwit reeks bestaande uit 3,423 aminozuurresidu's op basis van de Frans Polynesische stam (GenBank: KJ776791.2), werd gedrukt op een standaard glasplaatje in blokken van 15 lineaire residuen met een overlapping van 14 aminozuurresidu's in tweevoud voor een totaal 6,846 peptide vlekken. Naast de gehele Zika eiwit sequentie peptides, de microarray maakt gebruik van Influenza hemagglutinine (HA) peptiden voor interne controle.
Een Zika gevalideerd positieve serummonster verkregen Wadsworth Center (Albany, NY), werd gebruikt voor het identificeren van specifieke immunoglobuline M (IgM) en immunoglobuline G (IgG) reactieve peptiden. Na incubatie met het monster 's nachts, was de microarray gekleurd met een secundaire fluorescerende geconjugeerd antilichaam (Anti-menselijke IgM of anti-menselijke IgG) en geanalyseerd op een microarray scanner. Kwantificering van plek intensiteiten en peptide aantekening werd uitgevoerd met een specifieke software geboden door hetzelfde bedrijf dat de microarray vervaardigd.

High-density peptide microarrays toestaan screening van meer dan zesduizend peptiden op een uniforme standaard microscopie dia. Deze methode kan worden toegepast voor drugontdekking therapeutische target identificatie en ontwikkeling van diagnostiek. Hier presenteren we een protocol om te ontdekken van specifieke Zika virus (ZIKV) diagnostische peptiden met behulp van een diskette met hoge dichtheid peptide microarray. Een menselijke serummonster gevalideerd voor ZIKV infectie was geïncubeerd met een hoge dichtheid peptide microarray die hele ZIKV eiwit vertaald in 3,423 unieke 15 lineaire aminozuurresidu (aa) 's met een overlapping van 14-aa residuen bevatten afgedrukt in tweevoud. Kleuring met verschillende secundaire antilichamen binnen dezelfde array, we gedetecteerd peptiden die zich aan immunoglobuline M (IgM) en immunoglobuline G (IgG) antilichamen in het serum aanwezig binden. Deze peptiden werden geselecteerd voor verdere validatie experimenten. In dit protocol beschrijven we de strategie gevolgd om te ontwerpen, verwerken en analyseren van een high-density peptide microarray.

De resultaten verkregen met behulp van het protocol beschreven zijn afgebeeld in Figuur 2 en Figuur 3.
