Zika וירוס (ZIKV) האבחנה מבוססת על סימפטומים קליניים הוא מאתגר כי היא חולקת וקטורים תפוצה גאוגרפית, הסימפטומים עם קדחת דנגה, Chikungunya וירוס זיהום1. לאור הסיכון לתוצאות שליליות הריון בנשים נגוע ZIKV במהלך ההריון, חשוב להבחין בין 3 וירוסים. בדיקות האבחון המולקולרי הנוכחי אמנם ספציפיים, הם שימושיים רק דם או רוק בתקופה קצרה יחסית של דלקת חריפה2,3. מבחני סרולוגית חיוניים לצורך אבחון מחוץ תקופה ראשונית של זיהום4.
הפיתוח של ZIKV assay סרולוגית ספציפי הוא מאתגר משתי סיבות: ראשית, אנטיגנים Zika אשר מערכת החיסון האנושית מגיבה לא כיום ידועים; רצפים של חומצות אמיניות flaviviruses שנית, שנשמרת זירוז אשיג נוגדן. המטרה שלנו הייתה לגלות ייחודי ZIKV פפטידים מסוימים כדי לשמש אבחון. גישות שונות פותחו לספריות פפטיד המסך מכסה כל החלבונים כולל phage, חיידקי, ולהציג שמרים משטח5,6,7,8,9, 10. האסטרטגיה שלנו היה להשתמש microarray פפטיד בצפיפות גבוהה המתיר מהיר וזול תפוקה גבוהה סרולוגית הקרנות11,12 , לאחר מכן פפטידים מזוהה יכול לשמש כדי לשפר את זרם סרולוגית מבחני לגילוי של זיהום ZIKV.
פרוטוקול זה מאפשר גילוי Zika וירוס מסוים אבחון פפטידים באמצעות microarray של פפטיד בעל צפיפות גבוהה (איור 1). Microarray פפטיד בצפיפות גבוהה הופק באמצעות הטכנולוגיה להדפסת לייזר פפטיד. הרצף כולו ZIKV חלבון המורכב שאריות חומצה אמינית 3,423 בהתבסס על המתח פולינזיה הצרפתית (GenBank: KJ776791.2), הודפסה על משטח זכוכית רגילה בבלוקים של 15 שאריות ליניארי עם חפיפה בין שאריות חומצה אמינית 14 כפילויות עבור סך נקודות פפטיד 6,846. בנוסף כולו Zika חלבון רצף פפטידים, מנצל microarray שפעת hemagglutinin (HA) פפטידים עבור פקדים פנימיים.
Zika לאמת חיובי דגימת נסיוב שהושג ממרכז Wadsworth (אולבני, ניו יורק), היה משמש לזיהוי ספציפי אימונוגלובולינים ז (IgM), פפטידים תגובתי אימונוגלובולין G (IgG). לאחר המקננת עם הדגימה בן לילה, microarray מוכתם של נוגדן fluorochrome משני מצומדת (האנטי-האדם IgM או אנטי-אנושית IgG), והוא ניתח על סורק microarray. כימות של עוצמות ספוט, ביאור פפטיד בוצעה עם תוכנה ספציפיים המסופקים על ידי אותה חברה שייצרה את microarray.
