No presente trabalho, descrevemos um método novo para produzir derivados de medula óssea murino NK células in vitro. Progressão de câncer é largamente dependente do tumor microambiente1,2, incluindo o anfitrião-derivado de immunocytes, por exemplo, as células NK. Vários estudos demonstraram que as células NK intratumoral são negativamente correlacionadas com a progressão de tumor3,4. Além disso, estudos clínicos mostraram que a terapia com células NK adotiva é uma estratégia possível para câncer5,6,7,8,9. Apesar de interrupção de TGF-β sinalização no microambiente do tumor pode ser um caminho possível para eliminar cânceres, bloquear completamente a sinalização do TGF-β causará doenças autoimunes, devido à sua função de anti-inflamatórios, como evidenciado pela evolução da efeitos colaterais adversos incluindo inflamação sistêmica, defeitos cardiovasculares e auto-imunidade mouse modelos19. Assim, entender o mecanismo de trabalho de TGF-β-mediada imunossupressão conduzirá à identificação de um alvo terapêutico acessível para tratamento de câncer.
Para elucidar os eventos moleculares necessários para o desenvolvimento de células NK, Williams et al estabeleceu um sistema in vitro para diferenciar as células-tronco hematopoiéticas murino de medula óssea em NK células20. Este sistema facilita em grande parte o estudo mecanicista do desenvolvimento de célula NK, incluindo a identificação dos progenitores romance de NK células21. No entanto, os progenitores da medula óssea devem ser cultivados no sistema com células estromais OP9 de apoio como um alimentador camada20,21, e esta população celular heterogênea em grande parte limita a aplicação adicional de desregulação gene ferramentas (p. ex., silenciamento de genes mediada por siRNA) especificamente aplicada às células NK diferenciadoras.
Aqui, descrevemos um sistema alimentador-livre que se desenvolveu ainda mais, modificando o sistema in vitro de Williams et al.20. Natural killer (NK) células pertencem ao sistema imunitário inato e são uma primeira linha de defesa imunológica contra o cancro; no entanto, eles são suprimidos no microambiente do tumor e o mecanismo subjacente ainda é desconhecido. A falta de uma fonte confiável e consistente de células NK limita o progresso da investigação da imunidade de célula NK.
