Skeletspieren kunt aanpassen om inspanninstraining door veranderingen in de Bioenergetica en de fysieke structuur1. Een van de grote veranderingen teweeggebracht door duurtraining is mitochondriale biogenese, die kan worden geëvalueerd door een stijging van de uitdrukking van mitochondriale onderdelen (bijvoorbeeld cytochroom c oxidase [COX] subeenheden), alsmede de uitdrukking van de transcriptionele coactivator, PGC-1α2. Een groeiend aantal studies hebben aangegeven dat tal van andere factoren, met inbegrip van mitochondriale omzet en mitophagy, ook belangrijk voor de aanpassingen van de spier zijn. Nochtans, de mechanismen door welke acute of chronische oefening regelen deze processen in skeletspieren zijn nog onduidelijk.
Om de trajecten die oefening geïnduceerde spier aanpassingen regelen af te bakenen, hebben diverse modellen van de oefening zijn gebruikte in knaagdier studies, met inbegrip van de loopband, hardlopen wiel, en zwemmen van oefening. Deze protocollen hebben echter enkele beperkingen in dat ~ 4-12 weken nodig zijn om te observeren deze fenotypische wijzigingen3,4,5. Als een alternatieve experimentele methode, lagefrequentie-stimulatie-geïnduceerde chronische contractiele activiteit (CCA) heeft zijn effectief gebruikt, kan dit leiden tot aanpassingen van de spier in een aanzienlijk kortere termijn (dat wil zeggen, tot 7 dagen) en de gevolgen ervan lijken te zijn vergelijkbaar met, of zelfs groter dan andere oefening protocollen. Bovendien, de aanwezigheid van hormonale6, temperatuur7en8 van de neurologische effecten kan maken het moeilijk om te begrijpen van de spier-specifieke reacties op chronische oefening. Bijvoorbeeld, schildklier hormoon9,10 en insuline-achtige groei factor (IGF) -111 zijn geïdentificeerd om te bemiddelen opleiding-geïnduceerde muscle aanpassingen, die ook andere signaalroutes in skelet kunnen regelen spier. Met name, zijn CCA-geïnduceerde effecten minimaal geregeld systemische factoren, waardoor de nadruk worden gelegd op de directe reactie van de skeletspieren contractiele activiteit.
De externe eenheid voor CCA is geïntroduceerd door Tyler en Wright12, en is ontwikkeld met wijzigingen12. Kortom, de eenheid bestaat uit drie hoofdonderdelen: een Infrarood detector die kan worden omgezet in- en uitschakelen door blootstelling aan infrarood licht, een puls generator en een pulse-indicator (Figuur 1). Het ontwerp van de gedetailleerde circuit van de eenheid van de stimulator heeft geweest eerder13beschreven. De gedetailleerde en specifieke kenmerken van CCA kunnen worden gevonden meer diepte in een aantal review artikelen14,15,16,17. Kortom, het stimulatie-protocol is ontworpen om te activeren van de gemeenschappelijke peroneale zenuw op lage frequentie (dat wil zeggen, 10 Hz), en de bezenuwde spieren (tibialis anterior [TA] en extensor digitorum longus [EDL] spier) worden gedwongen om contract voor een vooraf bepaalde lengte van tijd (bijvoorbeeld 3-6 h). Na verloop van tijd dit hiermee de bovengenoemde spieren verschuift naar een meer aërobe fenotype, aangetoond door een toename van de capillaire dichtheid18 zowel mitochondriale inhoud19,20,21. Dus, deze methode is een bewezen model na te bootsen van sommige van de grote uithoudingsvermogen training aanpassingen binnen de skeletspieren van ratten.
Dit document stelt een gedetailleerde procedure voor de elektrode implantatie chirurgie voor het opwekken van CCA zodat de onderzoekers dit model in hun studie oefening kunnen toepassen. CCA is een uitstekend model voor het bestuderen van het tijdsverloop van muscle aanpassingen, waardoor een doeltreffend instrument voor het onderzoek van diverse gebeurtenissen op het gebied van moleculaire en signalering op beide punten van de vroege en latere tijd na het begin van de opleiding van de oefening.

Skeletspieren is een hoogst aanpasbaar weefsel, zoals de biochemische en fysiologische eigenschappen worden sterk gewijzigd in reactie op de chronische oefening. Om te onderzoeken van de onderliggende mechanismen die tot verschillende aanpassingen van de spier leiden, zijn een aantal oefening protocollen zoals loopband, wiel hardlopen en zwemmen oefening gebruikt in de dierproeven. Echter, deze uitoefenen modellen vereisen een lange periode van tijd om aanpassingen van de spier, die ook kunnen worden geregeld, door humorale of neurologische factoren, dus de beperking van hun toepassingen in het bestuderen van de spier-specifieke contractie-geïnduceerde aanpassingen. Indirecte lagefrequentie stimulatie (10 Hz) voor het opwekken van chronische contractiele activiteit (CCA) is gebruikt als een alternatief model voor de uitoefening opleiding, als het met succes tot spier mitochondriale aanpassingen binnen 7 dagen, onafhankelijk van systemische factoren leiden kan.
