Ionenkanalen van de membraan zijn geverifieerd als de primaire therapeutische doelen van meer dan 13% van de op dit moment bekende medicijnen voor de behandeling van een verscheidenheid van ziekten bij de mens, met inbegrip van cardiovasculaire en neurologische stoornissen18. Patch-clamp opnemen, is de gouden standaard voor het meten van de functionele van ionenkanalen met kleine molecules, wijd verbeid gebruikt voor ion kanaal liganden screening. Gericht op de cel met kleine moleculen aan de oppervlak-toegankelijk ion kanaal eiwitten biedt een enorm potentieel voor de ligand screening en biologische drug discovery1,2,3. Zo nodig een passend instrument is voor de studie van de interactie tussen ionkanaal en kleine moleculen en hun corresponderende functies. De patch-clamp-opname heeft aangetoond een uniek en onvervangbaar techniek in ion kanaal functionele assay. Bepalen of de kleine molecules rechtstreeks richten ionenkanalen echter andere technologieën. Traditioneel, werd de radioactieve ligand bindende bepaling gebruikt voor het observeren van de kinetiek van de binding tussen klein molecuul en haar doel ion kanaal eiwit. Het gebruik van deze techniek is echter beperkt vanwege haar eis in radioactieve labeling en detectie. Bovendien, voorkomt de vereiste stap naar het label van de kleine ligand in de studie zijn gebruikt in vele soorten ionenkanalen zonder bekende specifieke ligand. Sommige label-vrije technieken zoals NMR spectroscopie, X-ray diffractie, microscale thermophoresis (MST)4 en oppervlakte plasmon resonantie (SPR) werden gebruikt voor het meten van de eiwit-kleine molecule interacties. Maar dit soort tests meestal niet voldoende informatie kunt verstrekken vanwege de moeilijkheid om de full-length eiwit, lage resolutie van dynamiek, lage doorvoersnelheid en hoge kosten5. In tegenstelling tot deze technieken, bio-laag Interferometry (BLI) ontpopt zich als een nieuwe label-vrije methodologie te overwinnen deze nadelen voor het opsporen van eiwit-kleine molecule interacties door immobilizing van een kleine hoeveelheid eiwitSteekproef op de oppervlakken van biosensor en het meten van de optische veranderende signalen6,7. Als een veelbelovende biosensor platform, BLI techniek is al uitgevoerd om te zien hoe de interactie van kleine moleculen met natuurlijke water oplosbare eiwitten zoals een menselijke monoklonale antistof CR80208 en de gedetailleerde bepaling procedure heeft gemeld in een vorige artikel9. Hoewel de belangrijke rol van ion kanaal eiwitten voor nieuwe therapeutische doelen ontdekking heeft erkend, is de ion kanaal eiwit-kleine molecule interactie bepaling op basis van BLI niet beschreven.
De mens Ether à go-go kanalen (hEAG1) worden uitgedrukt in verschillende types van kankercellen en centrale zenuwstelsel, waardoor het kanaal a potentiële therapeutische doel van veel kankers en neuronale stoornissen10,11, 12,13,14. Het elektrofysiologische studie in ons lab heeft bevestigd dat de remmende werking van phosphatidylinositol 4, 5-difosfaat (PIP2) op hEAG1 kanaal15. Op basis van onze resultaten, PIP2 testen direct interactie met de hEAG1 met behulp van BLI techniek kan worden als een model voor andere soorten ion kanaal eiwit-kleine molecule samengestelde interactie met name voor die kanalen ontbreken van specifieke liganden. Volgens de instructies van BLI assay, wij bereid biotinyleerd hEAG1 eiwitten en geïmmobiliseerd hen op het oppervlak van daar (SA) biosensor tips gevolgd door interactie hen naar PIP2 oplossingen voor het observeren van hun directe binding tussen de eiwitten en de lipide. Na het aanbrengen van PIP2 op het gecoate oppervlak van hEAG1 eiwit, de dikte van de laag aan de oppervlakte toeneemt, die direct correleert de spectrale verschuiving en kan worden gemeten in real-time16. De bindende kinetiek kan worden bepaald als gevolg van een positieve verschuiving in vereniging stap en een negatieve verschuiving in dissociatie stap. Volgens dit principe, we gezuiverd van het functionele hEAG1 ion kanaal eiwit van HEK-239T stabiele expressie systeem met behulp van affiniteit zuivering methode de functionele toestand in vitro te handhaven, dan de kinetiek van de binding van gemeten verschillende concentratie PIP2, en leverde een semblable kinetische gegevens zoals waargenomen in elektrofysiologische metingen15. De nauwe correspondentie tussen de resultaten van de BLI en elektrofysiologische metingen tonen voor het eerst de geschiktheid van BLI als een passende analytische tool voor ion kanaal membraan eiwit-kleine molecule interactie.

De bio-laag interferometrie (BLI) bepaling is een waardevol instrument voor het meten van de eiwit-eiwit en eiwit-kleine molecule interacties. Hier beschrijven we eerst de toepassing van dit nieuwe etiket-vrije techniek op de studie van de interactie tussen menselijke EAG1 (hEAG1) kanaal eiwitten met de kleine molecuul PIP2. hEAG1 kanaal is erkend als potentieel therapeutisch doel vanwege zijn afwijkende overexpressie in kanker en enkele winst-van-functie mutaties die betrokken zijn bij sommige soorten neurologische ziekten. We gezuiverd hEAG1 kanaal eiwitten uit een zoogdieren stabiele expressie-systeem en de interactie met PIP2 door BLI gemeten. De succesvolle meting van de kinetiek van de binding tussen hEAG1 eiwit en PIP2 toont aan dat de bepaling BLI een potentiële hoge gegevensdoorvoer aanpak gebruikt voor nieuwe kleine-molecuul ligand screening in ion kanaal farmacologie is.

We de vlag hEAG1 kanaal fusieproteïne van HEK-293T cellen stabiel overexpressie hEAG1 gezuiverd.
