Canaux ioniques membranaires ont été vérifiées comme les principales cibles thérapeutiques de plus de 13 % des médicaments connus pour le traitement d’une variété de maladies humaines, y compris les troubles cardiovasculaires et neurologiques,18. Patch-clamp de l’enregistrement, la méthode de référence pour mesurer le fonctionnement des canaux ioniques à petites molécules, a été utilisée pour les ligands de canal d’ion de dépistage. Ciblant les protéines de canal cellulaire ion surface accessible avec petites molécules offre un énorme potentiel pour le dépistage de ligand et découverte de médicaments biologiques1,2,3. Ainsi, un outil approprié est nécessaire pour l’étude de l’interaction entre les canaux ioniques et de petites molécules et de leur fonction correspondante. L’enregistrement de patch-clamp a été démontrée pour être une technique unique et irremplaçable dans test fonctionnel de canal ionique. Toutefois, déterminer si les petites molécules ciblent directement les canaux ioniques nécessitent des autres technologies. Traditionnellement, l’essai de liaison du ligand radioactif a été utilisé pour observer les cinétiques de liaison entre les petites molécules et de sa protéine cible de canal ionique. Cependant, l’utilisation de cette technique est limitée en raison de son exigence de marquage radioactif et la détection. Par ailleurs, l’étape préalable d’étiqueter le petit ligand dans l’étude empêche son utilisation dans de nombreux types de canaux ioniques sans ligand spécifique connu. Certaines techniques d’étiquette comme NMR spectroscopy, diffraction des rayons x, a petite Echelle thermophorèse (MST)4 et résonance plasmonique de surface (SPR) ont été utilisés pour mesurer les interactions de protéine-petite molécule. Mais ces types d’analyses habituellement ne peut pas fournir l’information requise en raison de la difficulté à obtenir la protéine pleine longueur, la faible résolution des dynamique, faible débit et le coût élevé de5. Contrairement à ces techniques, bio-couche interférométrie (BLI) s’impose comme une roman méthodologie exempte d’étiquette pour surmonter ces inconvénients pour la détection des interactions de protéine-petite molécule par un échantillon de protéines en petites quantités sur les surfaces de l’immobilisation Biocapteur et mesurer le changement d’optique des signaux6,7. Comme une plateforme de biocapteurs prometteur, technique de BLI est déjà effectuée pour observer l’interaction de petites molécules avec de l’eau naturelle des protéines solubles comme un anticorps monoclonal humain CR80208 et la procédure de test détaillée a été rapportée chez un précédent article9. Bien que le rôle clé de protéine canal ionique pour une nouvelle découverte de cibles thérapeutiques a été reconnu, le dosage d’interaction de protéine-petite molécule canal d’ion basée sur BLI n’a pas été décrite.
L’homme l’éther à Go-Go canaux (hEAG1) est exprimés dans divers types de cellules cancéreuses et du système nerveux central qui en fait la canal cible thérapeutique potentielle a de nombreux cancers et troubles neuronaux10,11, 12,13,14. L’étude électrophysiologique dans notre laboratoire a confirmé l’effet inhibiteur du phosphatidylinositol 4, 5-bisphosphate (PIP2) le hEAG1 canal15. Selon nos résultats, essais PIP2 directement l’interaction avec le hEAG1 en utilisant BLI technique peut être un modèle pour d’autres types d’interaction composé de molécule de protéine de petit canal ionique spécialement pour ces chaînes manquent des ligands spécifiques. Conformément aux instructions de dosage BLI, nous avons préparé biotinylé protéines hEAG1 et eux immobilisé sur la surface de streptavidine (SA) conseils de biocapteurs ont suivis par interaction aux solutions2 PIP pour observer leur liaison directe entre le les protéines et les lipides. Après que la fixation de PIP2 sur la surface recouverte de hEAG1 protéine, l’épaisseur de la couche sur la surface augmente, qui met en corrélation le décalage spectral directement et peut être mesurée en temps réel16. La cinétique de liaison peut être déterminée en raison d’un changement positif dans l’étape de l’association et un déplacement négatif à l’étape de dissociation. Selon ce principe, nous purifiée de la protéine de canal ionique hEAG1 fonctionnelle du système d’expression stable HEK-239T en utilisant la méthode de purification d’affinité pour maintenir l’État fonctionnelle in vitro , puis mesuré les cinétiques de liaison de différente concentration PIP2et donné une semblable données cinétiques tel qu’observé dans des mesures électrophysiologiques,15. L’étroite correspondance entre les résultats de la BLI et mesures électrophysiologiques démontrent pour la première fois la pertinence du BLI comme un outil d’analyse approprié pour l’interaction protéine-petite molécule ion canal membranaire.

L’essai de l’interférométrie (BLI) bio-couche est un outil précieux pour la mesure des protéines et des interactions entre protéines à petites molécules. Nous décrivons ici tout d’abord l’application de cette nouvelle technique exempte d’étiquette pour étudier l’interaction des humains EAG1 protéines de canal (hEAG1) avec la petite molécule PIP2. canal de hEAG1 a été reconnu comme cible thérapeutique potentielle en raison de son aberrante surexpression dans les cancers et des mutations de gain de fonction un peu impliqués dans certains types de maladies neurologiques. Nous avons purifié hEAG1 protéines de canal d’un système d’expression stable chez les mammifères et mesuré l’interaction avec PIP2 par BLI. La mesure efficace de la cinétique de la liaison entre la protéine hEAG1 et PIP2 montre que le dosage BLI est une approche de haut débit potentielle utilisée pour le dépistage de nouvelles petites molécules ligand en pharmacologie des canaux ioniques.
