Westelijke bevlekken is een gemeenschappelijke techniek voor het opsporen van specifieke proteïnen en de activering van signaalroutes in weefsels of cellen. Hematopoietische stamcellen (HSCs) zijn zichzelf vernieuwende cellen die aanleiding tot alle bloed geslachten geven kunnen. Ze zijn relatief zeldzaam cellen in het beenmerg, waardoor biochemische analysen moeilijk. Benaderingen die geschikt zijn voor het analyseren van zeldzame cellen, zoals de stroom cytometry, zijn uiterst nuttig voor het kwantificeren van de relatieve hoeveelheden van cel oppervlakte markeringen en intracellulaire eiwitten. Echter, de analyse van intracellulaire eiwitten vereist het gebruik van cel permeabilization procedures antilichaam-toegang inschakelen, en niet alle cel oppervlakte epitopes overleven deze procedures1,2. Bovendien antilichamen die onderscheid tussen verschillende isoforms of decolleté eiwitproducten maken zijn vaak niet beschikbaar voor stroom cytometry en daarom onderzoekers nog steeds rekenen op Western blots voor bepaalde soorten analyses.
Westelijke vlekkenanalyse van cel lysates is een routineprocedure in de meeste laboratoria. Cellen onder inheemse voorwaarden die de epitopes van cel-oppervlakte molecules bewaren kunnen worden gezuiverd, en cel lysates kan vervolgens worden voorbereid en geanalyseerd. De analyse van eiwitten in zeldzame primaire cel populaties door Western blot kunt echter verplicht stellen grote aantallen dieren om voldoende cellen euthanizing. Door kleine aanpassingen aan de verschillende stappen, was een conventionele westelijke bevlekkende protocol kundig voor speurder eiwitten in relatief kleine aantallen HSCs (500-15.000, afhankelijk van de proteïne van belang). De aanpassingen omvatten nauwkeurig tellen de cellen, zorgvuldig hanteren de cel pellet, vermindering van de overdracht van cellen tussen buizen tot een minimum beperken van het verlies van de cel, en een bepaald aantal cellen met een geconcentreerde laden lysing buffer met proteasoom en phosphatase inhibitors. Vele gepubliceerde rapporten omvatten Western blots verkregen met 20.000 of meer HSCs3,4,5,6,7; deze eenvoudige procedure zal het verminderen van het aantal cellen en proefdieren nodig voor de productie van gelijkwaardige gegevens door tussen 4 en 40-voudige. Het protocol is ontworpen voor het normaliseren van resultaten op basis van de per cel, in plaats van aan een interne controle. Hierdoor kan de detectie van algemene verlagingen eiwitniveaus die worden over het hoofd gezien kunnen als gegevens worden genormaliseerd naar een interne controle. Het belang van normalisatie op basis van de per cel werd beschreven voor de analyse van gen expressie gegevens8, en hetzelfde principe geldt voor het kwantificeren van de eiwitten door westelijke vlek. Dit geoptimaliseerd protocol moet zinvol zijn voor iedereen die voor het analyseren van kleine aantallen cellen.

Hematopoietische stamcellen (HSCs) zijn zeldzame cellen, met de muis het beenmerg met slechts 25.000 ~ fenotypische lange termijn repopulatie HSCs. Een westelijke bevlekkende protocol werd geoptimaliseerd en geschikt voor de analyse van kleine aantallen HSCs (500-15.000 cellen). Fenotypische HSCs werden gezuiverd, nauwkeurig geteld en direct lysed in Laemmli monster buffer. Lysates met gelijke aantallen cellen werden geanalyseerd door elektroforese natrium dodecyl sulfaat polyacrylamidegel (SDS-pagina), en de vlek was bereide en verwerkte na standaard Westelijke Bevlekkende Protocollen. Met behulp van dit protocol, 2.000-5.000 HSCs kan routinematig worden geanalyseerd, en in sommige gevallen kunnen gegevens worden verkregen uit zo weinig als 500 cellen, in vergelijking met de cellen van de 20.000 tot 40.000 gemeld in de meeste publicaties. Dit protocol gelden over het algemeen andere hematopoietische cellen en maakt de routinematige analyse van kleine aantallen cellen met behulp van standaard laboratorium procedures.

Representatieve resultaten uit 500-2000 zuivere HSCs en HPs worden weergegeven in Figuur 1 en Figuur 2. Het signaal van de β-actine in Figuur 1 uit zo weinig als 500 HSCs kan worden herkend en HPs gezuiverd uit het beenmerg van een muis.
