De vorming van MGCs begeleidt een aantal pathologische toestanden in het menselijk lichaam gekenmerkt door chronische ontsteking1. Ondanks het akkoord dat macrofagen van mononucleated fuse aan formulier MGCs2, verrassend weinig studies hebben aangetoond fusion in verband met levende specimens3,4. Dit is omdat de macrofaag fusion wanneer geïnduceerd door inflammatoire cytokines5zijn niet bevorderlijk voor schone glazen oppervlakken die vereist voor de meeste beeldvormingstechnieken zijn. Inderdaad, als schoon glas wordt gebruikt als substraat voor macrophage fusion, vervolgens laag tot vergroting van de tussentijdse doelstellingen (dat wil zeggen, 10-20 X) en meer dan 15 uur continue imaging vaak moeten observeren van een enkele fusion-gebeurtenis.
Op de andere hand, fusogenic kunststof oppervlakken (bijvoorbeeld, permanox) of bacteriologische kwaliteit kunststof fusion2te bevorderen. Beeldvorming door middel van kunststof is echter problematisch omdat het substraat dik is en licht verstrooit. Dit compliceert imaging sinds lang werkende afstand (LWD) doelstellingen nodig zijn. LWD doelstellingen hebben echter meestal weinig licht het verzamelen van de capaciteit in vergelijking met hun tegenhanger dekglaasje aan-gecorrigeerd. Verder zijn technieken die gebruik maken van de veranderingen in de polariteit van het passeren van het model zoals differentiële interferentie contrast licht niet onmogelijk omdat kunststof birefringent. De belemmeringen die zijn gekoppeld aan het gebruik van kunststof zijn verder onderstreept door het feit dat het is onmogelijk om te voorspellen waar de macrofaag fusion/MGC vorming zal plaatsvinden op het oppervlak. Samen, deze beperkingen beperken de visualisatie van macrofaag fusie aan fase contrast optica, totale imaging duur verlengd (> 15 ononderbroken uur), en lage resolutie.
We onlangs geïdentificeerd een zeer fusogenic glazen oppervlak terwijl het uitvoeren van single-molecuul super resolutie microscopie met vaste macrofagen/MGCs4. Deze observatie was verrassend omdat schoon glas oppervlakken bevorderen fusion zeer laag debiet van ~ 5% na 24 uur in aanwezigheid van Interleukine-4 (IL-4), zoals bepaald door de fusie index4. We vonden dat de capaciteit ter bevordering van kernfusie te wijten aan oleamide besmetting was. Adsorptie van oleamide of andere verbindingen die ook lange-keten koolwaterstoffen maakte de glazen fusogenic. De meeste fusogenic samengestelde (paraffine) was micropatterned, en het een hoge mate van Spatio beheersing van de macrofaag fusion en een 2-fold stijging van het aantal fusion gebeurtenissen in dezelfde hoeveelheid tijd in vergelijking met permanox waargenomen bijgebracht. Deze optische kwaliteit oppervlakken verschaft de eerste glimp in de morfologische kenmerken en kinetiek die gelden voor de vorming van MGCs in levende specimens.
In dit protocol beschrijven we de fabricage van een verscheidenheid van glazen oppervlakken die kan worden gebruikt voor het controleren van de vorming van MGCs van levende specimens. Bovendien laten we zien dat deze oppervlakken vatbaar voor ver-veld super resolutie technieken zijn. Oppervlakte fabricage is afhankelijk van het doel van het experiment, en elk oppervlak met verwante voorbeelden in de tekst van de procedure wordt beschreven.

fabricating optical-quality glass surfaces to study macrophage fusion

Visualiseren van de vorming van multinucleaire reus cellen (MGCs) van levende specimens heeft zijn uitdagend wijten aan het feit dat meest levende imaging technieken voortplanting van het licht door glas vereisen, maar op glas macrofaag fusion een zeldzame gebeurtenis is. Dit protocol stelt de fabricage van verschillende optische kwaliteit glazen oppervlakken waar adsorptie van verbindingen met lange-keten koolwaterstoffen glas transformeert in een fusogenic oppervlak. Voor het eerst, voorbereiding van schone glazen oppervlakken als grondstof voor oppervlakte modificatie is beschreven. Ten tweede, een methode wordt alleen ondersteund voor de adsorptie van verbindingen met lange-keten koolwaterstoffen om te converteren van niet-fusogenic glas naar een fusogenic ondergrond.
