Mens fokus for fleste av disse studiene var identifisere vert faktorer som påvirker noen aspekter av viruset replikering og ikke identifisere biotherapeutic mål for forbedring kan viruset terapi, kan verdifulle data være minelagt fra disse datasett i denne forbindelse. En mengde kan virus plattformer blir nå testet i prekliniske og kliniske studier inkludert herpes simplex virus, reovirus og vaccinia virus, som har hatt påviselig suksess i sent kliniske forsøk. For fleste av disse plattformene, har innsatsen vært rettet mot endre virus genomer å forbedre terapi. Vi rapporterte den første studie demonstrere proof-of-concept for bruk av høy gjennomstrømming genetisk screening for å identifisere vert mål som kan manipuleres til å forbedre kan viruset terapi. For å oppdage vert gener som modulerer Maraba virus oncolysis, en kan rhabdovirus testes i fasen jeg og II prøvelser, vi gjennomført genomet hele RNAi skjermer over tre forskjellige svulst cellelinjer i duplikat med en siRNA (kort konflikt RNA) bibliotek målretting 18,120 gener15. Sti analyse av treff i skjermene avdekket anriking av medlemmer av de ER (endoplasmatiske retikulum) stress respons. In vitro og i vivo testing ved bruk av et lite molekyl inhibitor av IRE1α resulterte i dramatisk forbedret kan effekt.

genome-wide rnai screening to identify host factors that modulate oncolytic virus therapy

Høy gjennomstrømming genomet hele RNAi (RNA-interferens) screening teknologi har vært mye brukt for å oppdage vert faktorer som påvirker virus replikering. Her presenterer vi bruk av denne teknologien for å avdekke vert mål som spesielt modulerer replikering av Maraba virus, kan rhabdovirus, og vaccinia virus med mål om å forbedre terapi.
