Er zijn verschillende kritische stappen voor de generatie van dit systeem co cultuur. Het is belangrijk om ervoor te zorgen dat de muis DRG neuronen en peritoneale macrofagen vers worden bereid en gezond. Wij hebben ervaren verminderde neurite activiteit van de uitgroei van de CM de dissectie van alle de DRG toen meer dan 30 min. Een aantal studies hebben getracht te verbeteren CNS axon regeneratie na de verwondingen van het ruggenmerg of de hersenen. Ontstekingsreacties, onvermijdelijk bij de verwondingen in het zenuwstelsel, worden traditioneel verondersteld deel te nemen aan secundaire pathologische processen die leiden tot de schadelijke resultaten1,2. Methylprednisolon waarin kan worden onderdrukt ontstekingsreacties is immers de enige erkende therapie voor acute ruggenmerg letsel3. Echter, meer recente studies hebben aangetoond dat de regeneratie of reparatie van gewonde zenuwstelsel4,5,6door macrofagen, een representatieve inflammatoire celtype, kunnen deelnemen. Bijvoorbeeld Infiltrerend macrofagen na een lens letsel produceren pro-regeneratieve moleculen ter bevordering van de regeneratie van de retinale ganglion neuronen7,8. Daarnaast verhoogd getransplanteerde DRG neuronen axon groei van de regio waar de macrofagen waren geactiveerd door zymosan9. Bovendien, de macrofagen op de site van de laesie kunnen leiden tot een groei-tolerante milieu voor gewonde perifere zenuwen10.
Ons werk ook sterke aanwijzingen dat macrofagen tot de capaciteit van axon regeneratie in aangrenzende neuronen bijdragen kunnen geboden. We hebben aangetoond dat de activering van de macrofagen in de achterwortelganglia, bevatten (DRG) zijn essentieel in de verbeterde regeneratief vermogen van DRG sensorische neuronen na een voorbehandeling perifere zenuwen letsel11. Vergelijkbaar onderzoek werd onafhankelijk gemeld uit een ander laboratorium12. We toonden ook aan dat intraganglionic injectie van dibutyryl cyclisch ADENOSINEMONOFOSFAAT (db-kamp), die een bekende molecule is tot vergroting van de capaciteit van axon regeneratie13, de activering van macrofagen veroorzaakt. Het deactiveren van macrofagen afgeschaft de effecten van db-kamp op neurite uitgroei activiteit. Volgende werken geïdentificeerd letsel-geïnduceerde expressie van CCL2 in de neuronen als een signaal aan het stimuleren van macrofagen met een pro-regeneratieve fenotype14,15.
Op basis van de bovengenoemde experimentele resultaten, hebben we een in vitro model lijkt op de moleculaire gebeurtenissen die zich voordoen in de DRG na een conditioneringscycli letsel model11,14opgericht. In dit model wordt db-cAMP toegepast op het neuron-macrofaag co culturen opwekken intercellulaire signalering dat leidt tot de activering van de macrofagen met een pro-regeneratieve fenotype. Hier beschrijven we gedetailleerde protocollen waarmee we kunnen genereren macrofagen die afscheiden van moleculaire factoren ter bevordering van neurite uitgroei (Figuur 1). Deze experimentele model illustreert een concept dat macrofagen kunnen worden gestimuleerd of geïnduceerde ter ondersteuning van axon regeneratie na de verwondingen aan zenuwstelsel. Ons model zal ook nuttig zijn bij het bestuderen van de mechanismen in de intercellulaire signalering die leidt tot de activering van de pro-regeneratieve macrofagen zijn.

neuron-macrophage co-cultures to activate macrophages secreting molecular factors with neurite outgrowth activity

Er zijn sterke aanwijzingen dat macrofagen het herstel of de reparatie van gewonde zenuwstelsel kunnen deelnemen. Hier beschrijven we een protocol waarin macrofagen zijn aangezet te produceren geconditioneerd medium (CM), dat neurite uitgroei bevordert. Volwassen achterwortelganglia ganglion (DRG) neuronen zijn acuut losgekoppeld en verguld. Nadat de neuronen stabiel vastgemaakt zijn, zijn peritoneale macrofagen mede gekweekte op een cel cultuur invoegen die op hetzelfde goed worden bedekt. Dibutyryl die cyclisch AMP (db-cAMP) wordt toegepast op de co culturen gedurende 24 uur, waarna de celcultuur invoegen waarin de macrofagen wordt verplaatst naar een ander goed voor het verzamelen van CM voor 72 uur. De CM vanaf de co culturen behandeld met db-cAMP, wanneer toegepast op een afzonderlijke volwassen DRG neuron cultuur, vertoont robuuste neurite uitgroei activiteit. De CM verkregen uit de db-kamp-testgroep culturen die bestaat uit één celtype alleen, DRG neuron of peritoneale macrofaag, deed niet neurite uitgroei activiteit vertonen. Dit geeft aan dat de interactie tussen neuronen en macrofagen is onmisbaar voor de activatie van macrofagen afscheidende moleculaire factoren met neurite uitgroei activiteit in CM. Dus zullen onze co cultuur paradigma ook nuttig zijn te onderzoeken intercellulaire signalering in de neuron-macrofaag interactie te stimuleren de macrofagen te worden begiftigd met een pro-regeneratieve fenotype.

We beschrijven een protocol dat macrofagen staat afscheidende moleculaire factoren met neurite uitgroei activiteit kan genereren. De macrofaag CM verkregen uit de co culturen behandeld met db-cAMP resulteerde in robuuste neurite uitgroei wanneer toegepast op een apart DRG neuron cultuur (figuur 2A).
