Bien que cBDL est la technique la plus courante expérimentale pour cholestase obstructive, il peut présenter des défis multiples chez les rongeurs. Selon le nombre de jours postopératoires requis pour atteindre un point de terminaison, les animaux peuvent éprouver une cause de morbidité et de mortalité avec le temps. Les modèles chirurgicaux d’atteinte hépatique aiguë et de réparation, par exemple une hépatectomie partielle ou la ligature du cholédoque (LCCC), ont servi pendant des décennies chez les rongeurs pour étudier la régénération du foie et biopathologie1,2. Dans cBDL, le canal cholédoque (qui draine toutes les bile produite dans le foie) est ligaturé, entraînant une cholestase obstructive, l’inflammation et la fibrose dans les 2 ou 3 premières semaines suivant la chirurgie, avec éventuelle évolution vers la cirrhose3. Prolifération des canaux cholédoques, transdifférenciation et l’induction de cellules progénitrices du foie résident sont également caractéristiques de la LCCC4,5. Bien que la LCCC élucide des mécanismes spécifiques de cholangiopathie obstructive, qui sont bien caractérisés avec des résultats reproductibles chez les souris de type sauvage et chez ces souches avec foie normal6,7,8, le procédure peut être techniquement difficile dans certains modèles de souris transgéniques de maladies du foie, nécessitant plus d’animaux que prévu en raison de la morbidité plus élevée et la mortalité d’obstruction des voies biliaires au fil du temps. Cette technique est également bénéfique pour l’étude des maladies des souris transgéniques avec les sous-jacents du foie, qui pourrait tolérer pas sinon le stress d’expériences de cours temps prolongé après la LCCC.
pBDL peut fournir une solution de rechange raisonnable pour aider à surmonter certains de ces défis. PBDL initiale des études chez la souris sauvage visant à distinguer localement par intérim et systémiques des médiateurs de l’inflammation9. Dans pBDL, confluent biliaire principal des canaux biliaires hépatiques droite et gauche au-dessus de la voie biliaire principale est visualisé avec précaution à l’hile et seulement le canal biliaire hépatique gauche est ligaturé pour générer une cholestase obstructive localisée. pBDL offre plusieurs avantages à la LCCC. Dans pBDL, le lobe droit unligated sert de témoin interne identique apparié pour le lobe gauche ligaturé, soumis aux mêmes effets systémiques, tels que l’état nutritionnel ou de facteurs de la circulation. Bien que des médiateurs pro-inflammatoires systémiques étaient présents, Osawa et al. observé moins la fibrose et nécrose dans les lobes unligated, mais augmenté des cellules inflammatoires et l’expression de β-facteur de croissance transformant dans le lobe ligaturé9.
Plus récemment, pBDL a été redéfini pour étudier les effets parenchymateux hépatiques de bile retenu10. En utilisant le même animal contrôle interne (unligated « trompe-l'œil ») et de groupes expérimentaux, pBDL efficacement diminue de moitié le nombre d’animaux nécessaires par l’expérience, qui à son tour est plus rentable. Les souris tolèrent les effets de pBDL beaucoup mieux, afin que le moment idéal cours expérimenter (≥ 2 semaines) peuvent être réalisés dans le lobe ligaturé. Mais surtout, pBDL peut distinguer des effets directs intrahépatiques de cholestase obstructive et conservé des composants de la bile dans le lobe ligaturé, par rapport au lobe unligated contrôle. Dans l’ensemble, pBDL est une méthode intéressante pour étudier les effets de lobe spécifique de la cholestase localisée dans le foie.

Chez les rongeurs, ligature complète du cholédoque (LCCC) de la voie biliaire principale est une technique chirurgicale établie pour l’étude de prolifération de cholestase et canal biliaire obstructive. Cependant, des expériences à long terme peuvent conduire à la mortalité et une morbidité accrue. Contrairement à la LCCC, un groupe de contrôle opérés distinct n’est pas nécessaire. pBDL est très utile de comparer directement les modèles localisés par rapport aux effets systémiques de la cholestase et d’autres composants de la bile retenue. pBDL peut également être réutilisé comme nouvelle méthode pour étudier les mécanismes associés aux médicaments et la migration cellulaire.

Pour cette expérience typique, pBDL a été réalisée chez les souris transgéniques PiZ en triple, qui récapitulent la maladie du foie humaine trouvée en alpha-1 antitrypsine (A1AT).
