Aunque cBDL es la técnica experimental más común de colestasis obstructiva, puede presentar múltiples desafíos en roedores. Los modelos quirúrgicos de reparación, como la hepatectomía parcial o ligadura del conducto biliar común (cBDL) y lesión del higado aguda se han utilizado durante décadas en roedores para estudiar la regeneración del hígado y pathobiology1,2. En cBDL, es ligado el conducto colédoco (que drena toda la bilis producida en el hígado), dando por resultado colestasis obstructiva, la inflamación y la fibrosis en las primeras 2-3 semanas después de la cirugía, con eventual progresión a la cirrosis3. Proliferación de conductos biliares, transdiferenciación e inducción de células progenitoras residentes de hígado son también características de cBDL4,5. Aunque cBDL elucida mecanismos específicos de cholangiopathy obstructiva, que están bien caracterizados con resultados reproducibles en ratones de tipo salvaje y en esas tensiones con hígados normales6,7,8, la procedimiento puede ser técnicamente exigente en algunos modelos de ratón transgénico de enfermedades del hígado, que requieren más animales de lo previsto debido a la mayor morbilidad y mortalidad de la obstrucción biliar en el tiempo. Esta técnica también es beneficiosa para el estudio de la enfermedad de ratones transgénicos con base de hígado, que de lo contrario no podría tolerar el estrés de los experimentos de curso prolongado tras cBDL.
pBDL puede proporcionar una alternativa razonable para ayudar a superar algunos de estos retos. PBDL inicial estudios en ratones de tipo salvaje pretenden distinguir localmente actuando y sistémicos mediadores de la inflamación9. En pBDL, cuidadosamente, la confluencia biliar principal de los conductos biliares hepáticos derecho e izquierdos por encima del conducto biliar común se visualiza en el hilum, y sólo el conducto biliar hepático izquierdo se unen para generar una colestasis obstructiva localizada. pBDL ofrece varias ventajas a cBDL. Aunque estaban presentes mediadores pro-inflamatorios sistémicos, Osawa et al. había observado menos fibrosis y necrosis en los lóbulos unligated, pero aumento de células inflamatorias y expresión de factor de crecimiento β transformador en el lóbulo ligados9.
Más recientemente, se ha redefinido pBDL para estudiar efectos parenquimales hepáticas de bilis retenida10. Utilizando el mismo animal para los grupos experimentales y control interno (unligated "sham"), pBDL mitades con eficacia el número de animales necesarios por experimento, que a su vez es más rentable. En roedores, ligadura del conducto biliar completa (cBDL) del conducto biliar común es una técnica quirúrgica establecida para el estudio de colestasis obstructiva, proliferación de conducto biliar. A diferencia de cBDL, un grupo de control independiente operado simulada no es necesario. pBDL es muy útil para comparar directamente localizada versus efectos sistémicos de la colestasis y otros componentes de la bilis retenida. también puede reutilizar pBDL como un nuevo método para investigar mecanismos relacionados con medicamentos y migración celular.
