Mange kliniske skrider frem i levertransplantation kan henføres til dyreforsøg. Rotte OLT er en udbredt og godt accepterede model i forskning af orgel bevarelse, transplantation immunologi, fysiologi og patologi. Den rotte model af orthotopic lever transplantation (OLT) er afgørende for transplantation forskning1,2. Den første rotte OLT blev beskrevet af Lee et al. i 19733. I denne model, blev alle fartøjer rekonstrueret af hånd-Sutur teknik. Hånd-Sutur teknik kræver avancerede microsurgical færdigheder, som væsentligt begrænser dets udnyttelse. Siden da, er blevet rapporteret forskellige ændringer til den originale protokol af rotte OLT. Blandt dem, manchet teknik til anastomose af Vena (PV) og infrahepatic vena cava (IHVC) rapporteret af Kamada et al. i 1979 er betragtes som en væsentlig forbedring til denne model, da det væsentligt forenklet genopbygning procedurer4 . Men på grund af den korte forreste væg af modtagernes suprahepatic vena cava (SHVC), manchet teknik er meget vanskeligt at bruge til genopbygning af SHVC. De fleste forskere på dette område stadig bruge hånd-Sutur teknik for SHVC genopbygning, hvilket gør det flaskehals skridt i rotte OLT5,6,7.
Den magnetiske anastomose teknik (dvs, magnamosis) er en metode til at forbinde to skibe eller andre rørformede strukturer ved hjælp af den tiltrækkende kraft mellem to magneter8,9,10,11. Den magnetiske kraft gradvist komprimerer og bygger om vævet ind i en stærk, sutureless anastomose12,13. Denne komprimering anastomose har vist sig for at være effektive i mennesker14,15. Vi har designet et par magnetiske ringe specielt til anastomose af SHVC i rotter. Vores seneste undersøgelse har vist, at den magnetiske anastomose teknik er overlegen i forhold til hånd-Sutur teknik for SHVC genopbygning i rotte OLT16. Formålet med denne artikel er at give en detaljeret, trinvis protokol for SHVC genopbygning i rotter brug af roman magnetiske anastomose teknik.

Den rotte model af orthotopic lever transplantation (OLT) er afgørende for transplantation forskning. Det er en meget sofistikeret dyremodel og kræver en stejl indlæringskurve. I denne model, blev genopbygningen af PV og IHVC udført af standard manchet teknikken, mens genopbygningen af galdegang (BD) blev udført af en stent teknik. Den hepatiske re arterialization blev ikke udført. Den magnetiske anastomose teknik lettere meget SHVC genopbygning og væsentligt forkortet anphepatic fasen. Efter en rimelig indlæringskurve, kan selv forskere uden avanceret microsurgical færdigheder producere pålidelige og reproducerbare resultater ved hjælp af denne rotte model af OLT.

Efter ca. 10 forsøg, blev magnetiske anastomose teknik for SHVC genopbygning med held beherskes af en forsker, der ikke havde nogen forudgående microsurgical uddannelse. Genopbygningen af SHVC tog mindre end 2 min. Anhepatic fasen for de modtagende rotter var ca 10 min. Ingen blodprop, blødning eller angiostegnosis blev observeret på webstedet SHVC Anastomotiske 1, 5 og 30 dage efter reperfusion.
