Beaucoup progresse clinique en transplantation hépatique peut être attribuée aux études chez l’animal. Rat BTA est un modèle largement utilisé et reconnu dans la recherche de préservation d’organe, transplantation immunologie, physiologie et pathologie. Toutefois, il est également une procédure très complexe et requiert des compétences de microchirurgie avancée. Le modèle de rat de transplantation hépatique orthotopique (BTA) est essentiel pour la transplantation recherche1,2. Le premier rat BTA a été décrit par Lee et al. , en 1973,3. Dans ce modèle, tous les navires ont été reconstruites par la technique de la main-suture. La technique de la main-suture nécessite des compétences microchirurgies avancées, qui limite considérablement son utilisation. Depuis lors, diverses modifications au protocole original de rat BTA ont été signalées. Parmi eux, la technique de Brassard pour l’abouchement de la veine porte (PV) et infrahepatic vena cava (IHVC) rapporté par Kamada et coll. en 1979 constitue une amélioration majeure de ce modèle, comme il a considérablement simplifié les procédures de reconstruction4 . Toutefois, en raison de la courte paroi antérieure de la veine cave sus-hépatique bénéficiaires (SHVC), la technique du brassard est très difficile à utiliser pour la reconstruction de la SHVC. La plupart des chercheurs dans ce domaine toujours utilisent la technique de suture-main pour la reconstruction de SHVC, qui en fait l’étape de goulot d’étranglement rat OLT5,6,7.
La technique d’anastomose magnétique (par exemple, magnamosis) est une variante de raccordement de deux navires ou autres structures tubulaires à l’aide de la force d’attraction entre deux aimants8,9,10,11. La force magnétique progressivement compresse et remodèle le tissu dans une anastomose fort, valvule12,13. Cette anastomose de compression s’est avérée pour être efficace pour les humains14,15. Nous avons conçu une paire d’anneaux magnétiques spécifiquement pour l’anastomose de la SHVC chez le rat. Notre étude récente a montré que la technique d’anastomose magnétique est supérieure à la technique de suture-main pour la reconstruction de SHVC au rat BTA16. Le but de cet article est de fournir un protocole détaillé, étape par étape pour la reconstruction de SHVC chez le rat en utilisant la technique d’anastomose magnétiques roman.

Le modèle de rat de transplantation hépatique orthotopique (BTA) est essentiel pour la recherche de la transplantation. C’est un modèle animal très sophistiqué et nécessite une courbe d’apprentissage abrupte. L’introduction de la technique de Brassard pour l’abouchement de la veine porte (PV) et infrahepatic vena cava (IHVC) a considérablement simplifié la procédure de transplantation chez les rats. Toutefois, en raison de la courte paroi antérieure de la veine cave sus-hépatique bénéficiaires (SHVC), la technique du brassard est très difficile à utiliser pour la reconstruction de la SHVC. La plupart des chercheurs dans ce domaine toujours utilisent la technique de suture-main pour la reconstruction de SHVC, qui en fait l’étape de goulot d’étranglement en transplantation hépatique orthotopique de rat. La technique d’anastomose magnétique (c.-à-d., magnamosis) est une variante de raccordement de deux navires à l’aide de la force d’attraction entre deux aimants. Notre étude récente a montré que la technique d’anastomose magnétique est supérieure à la technique de suture-main pour la reconstruction de SHVC chez le rat. Dans cet article, nous montrons un protocole étape par étape pour la reconstruction de SHVC chez le rat en utilisant la technique d’anastomose magnétiques roman. Dans ce modèle, la reconstruction de la PV et IHVC a été réalisée par la technique de Brassard standard, tandis que la reconstruction de la voie biliaire principale (BD) a été réalisée par une technique de stent. re-L’arterialization hépatique n’a pas été effectuée. La technique d’anastomose magnétique reconstruction SHVC grandement facilitée et réduit considérablement la phase d’anphepatic. Après une courbe d’apprentissage raisonnable, même les chercheurs sans compétences avancées de microchirurgie peuvent produire des résultats fiables et reproductibles à l’aide de ce modèle de rat d’OLT.

Après environ 10 tentatives, la technique d’anastomose magnétique pour la reconstruction de SHVC a été avec succès maîtrisée par un chercheur qui n’avait aucune formation préalable de microchirurgie. La reconstruction de la SHVC a pris moins de 2 min. La phase anhépatique pour les rats bénéficiaires était environ 10 min.
