De kritiske hendelsene i HIV-1 patogenesen skje i vev. Infeksjoner i genital mucosa utgjør flertallet av alle HIV-1 overføring begivenheter verdensomspennende4. Histocultures kan studere intercellulære interaksjoner i menneskelig vev, og de kan benyttes til modell vert-patogen interaksjoner under kontrollerte laboratorieforhold. Denne metoden kan overvåke HIV-1 replikering og patogenesen ved hjelp av flere teknikker, inkludert immunanalyser, qPCR og flowcytometri. Plasser platene i inkubatoren, sett til 37 grader Celsius, 5% CO2, og større enn eller lik 90% fuktighet til vevet explants er klar til å lastes på svampene.Overfør mandlene fra transportbeholderen til en 100 millimeter petriskål som inneholder 10 til 20 milliliter CMT ved hjelp av sterile tang. Overfør explants til en ny 100 millimeter petriskål som inneholder 10 til 20 milliliter CMT for å unngå vevsutsendering mens du fortsetter med disseksjonen.På slutten av disseksjonen, virvle platen for å randomisere explant fordelingen. Legg ni vev explants på toppen av hver gelatin svamp i en 6-brønns plate ved hjelp av tang, slik at rommet mellom dem. Sett platen tilbake til inkubatoren.Samle en prøve av kultur medium med en pipette, og overføre den til sterile 1,5 eller to milliliter skrue cap rør for lagring på minus 80 grader Celsius. Høst vevet explants med sterile tang, og overføre explants til sterile 1,5 eller to milliliter skrue cap rør. Behandle eller lagre vevsprøvene etter behov av eksperimentet.Aspirer restkulturmediet i brønnen med en pipette, og kast det i en beholder med desinfeksjonsmiddelløsning. Vipp platen og skyv forsiktig gelatinsvampene til den øvre delen av brønnen for å la mediet samle seg nederst, og aspirer og kast den. Legg nå til tre til fire milliliter kulturmedium til hver brønn.Bruk sterile tang, returner eventuelle vevsutvinninger som falt ned i mediet til gelatinsvampen før platen returneres til inkubatoren. På slutten av kulturen, kast alt biologisk avfall i apposite beholdere i henhold til instituttets forskrifter om håndtering av farlige biologiske stoffer samt den spesifikke risikovurderingen. Inter-donor variabilitet kan påvirke HIV-1 replikering målt ved konsentrasjonen av HIV-kjerneproteinet p24 gag i vev explant kultur supernatant, som demonstrert for livmorhalsvev fra tre forskjellige donorer smittet med samme virusinokulum.For den første donoren indikerer akkumuleringen av p24 gag i kulturmedium av eksploner infisert med HIV-1 BaL alene tydelig en produktinfeksjon. Det antiretrovirale stoffet 3TC som fullstendig opphever HIV-replikering ble brukt som en negativ kontroll. Mengden p24 gag målt i kulturmedium av 3TC-behandlede explants avslører fraksjonen av virusinokulum aspecifically absorbert og utgitt av vev explants under kultur.I vevseksplanter fra den andre donoren fører HIV-1 tarminfeksjon til en jevn reduksjon i konsentrasjonen av p24 gag over tid, noe som indikerer en infeksjonsavbrudd eller umulig å oppdage ved p24 gag kvantifisering. Dette bekreftes av 3TC-behandlede kontrollpuber som viser en lignende HIV-replikering kinetisk. Infeksjon av vev explants fra den tredje donor gir liten mengde virus.I dette tilfellet er inkludering av 3TC-behandlingen som en negativ kontroll nødvendig for å fastslå tilstedeværelsen av infeksjon med de novo virusproduksjon. Ex vivo menneskelige vev kulturer kan brukes til å studere patogenese av mikrober enn HIV, som vaccinia virus eller herpes virus.
