In tegenstelling tot de procedure tijdrovend genetische manipulatie, wat gewoonlijk weken duurt, hier tonen we eenvoudige twee stappen en one-step chemische vervoeging procedures, die kunnen worden voltooid binnen 3 dagen, van het toevoegen van de kanker targeting ligand en/of fluorescentie detectie kleurstof de Cys/Lys sites van HEVNPs. De techniek kan worden gebruikt aan het scherm voor de beste ligand-doelstelling van een pool van kandidaten, en dus maakt gebruik van de beschikbare peptide/kleine molecuul synthese die...

De ontwikkeling van de nano-sized vectoren in therapeutische en diagnostische levering, bekend als nanotheranostics, is veel van de biomedische veld uit de buurt van gegeneraliseerde behandelingen naar gerichte levering1verschoven. Gerichte nanotheranostic levering integreert nano-sized vectoren (nanodeeltjes) met theranostic moleculen om stabiel theranostic moleculen naar een specifieke zieke weefsel of biochemische traject2,3,4 . Nanogeneeskunde is gekomen op de voorgrond van gerichte levering omdat optimaal formaat nanodeeltjes de capaciteit hebben om verkeer van theranostic moleculen stabiliseren en selectief target cel oppervlakte moleculen gepresenteerd op zieke weefsels. Nog steeds lijden vele nanotheranostic platformen passieve cel opname, pre volwassen afbraak en toxiciteit onvoldoende vereniging met theranostic moleculen. Experimentele overwinnen veel van deze belemmeringen in gerichte levering. Ze zijn gebruikt als nanocarriers om buitenlandse epitopes weergeven en/of leveren van kleine moleculen: een regime dat kan worden gebruikt ter bestrijding van vele ziekten1. Deze toepassing steunt voornamelijk op de eigenschap van zelf-assemblage en het gemak van genetische modificaties, om te voldoen aan de ontworpen toepassing voor de gegeven VLP. Vergeleken met genetische manipulatie, chemische vervoeging van buitenlandse peptides te VLP wordt weergegeven, een aanzienlijk voordeel omdat daardoor een groot aantal entiteiten, zoals peptiden of oligosachariden, aan zijn geconjugeerd aan de oppervlakte van experimentele in een gemoduleerde en flexibel op te treden zonder wijziging van VLP vergadering.
HEVNPs, afgeleid van de recombinante HEV Eiwitmantel eiwit, 2nd open frame (ORF2) lezen, zijn niet-infectieuze, zelf monteren capsids staat van cel-bindende en post. Omdat HEV geëvolueerd voor mucosal transmissie, is de geassembleerde Eiwitmantel eiwit ook stabiel in Proteolytische en zure mucosal voorwaarden5. HEVNPs vormen een hol, T = 1 icosahedrale Eiwitmantel, samengesteld uit 60 identieke eenheden6,7 ORF2, waardoor zij zeer stabiele zowel in opslag Barre fysiologische omstandigheden. Bij gebrek aan een virale genetische elementen, wordt de productie van efficiënte, hoge opbrengst bereikt door baculovirus expressie systeem in insect cellen. Vanwege hun Proteolytische stabiliteit, zijn zelf geassembleerde HEVNPs gewonnen en gezuiverd van cel supernatant, aanzienlijk verminderen nodig zuivering stappen. Bovendien bezitten HEVNPs een oppervlak blootgestelde uitsteeksel domein (P domein) verbonden door middel van een flexibele scharnier aan een stabiele icosahedrale base. Het domein P vormt spikes oppervlak-blootgesteld bovenop de icosahedrale base, terwijl het flexibele scharnier het mogelijk maakt om het domein P aanzienlijk te wijzigen zonder afbreuk te doen aan de basis van de icosahedrale structuur. Met 60 herhaalde eenheden, enkele specifieke wijziging resulteert in 60 symmetrische sites voor chemische modulatie. Onlangs, hebben wij voorgesteld een nano-platform met behulp van HEVNP die chemisch liganden of kleine molecules voor theranostic toepassingen conjugaat kan. Dit werd bereikt door het te vervangen een één aminozuur cysteïne op het domein van de uitsteeksel van HEV-VLP als een reactie site met maleimide-linked peptiden of moleculen. Op basis van eerdere structuuranalyse van HEV-VLP en goed bestudeerde immunogene epitopes8,9, de volgende vijf HEV-VLP-aminozuren werden vervangen door cysteïne als mogelijke kandidaat-lidstaten: Y485C, T489C, S533C, N573C en T586C ( Figuur 1). Na expressie en reiniging van insect cellen, hun VLP-formaties werden bevestigd door Transmissie Electronenmicroscopie (TEM) observatie (Figuur 2), en de blootgestelde cysteïne sites werden geanalyseerd door Western blot na maleimide-linked Biotine vervoeging (Figuur 2). Onder de vijf mutanten, HEVNP - 573C weergegeven van het sterkste signaal van maleimide-Biotine geconjugeerde (Figuur 2) en werd gebruikt voor de follow-up van de demonstratie als de nanocarrier voor borst kankercel gericht op4 (Figuur 3).
Dit protocol beschrijft chemische vervoeging methoden om tumor-targeting moleculen aan te HEVNPs via oppervlakte cysteïne Woordherkomst en-opbouw. We detail de vervoeging van tumor targeting en detectie moleculen voor de levering van de tumor met recombinant HEVNPs met een cysteïne bij N573 (HEVNP - 573C). We gericht op een proces van twee stappen Klik chemie vervoeging te binden een borst kanker tumor gericht op peptide, LXY3010 te HEVNPs aan formulier LXY30-HEVNP (Figuur 4). Vervolgens, N-hydroxysuccimide (NHS)-Cy5.5 waren geconjugeerd met de afzonderlijke Lys-site op HEVNPs te bouwen LXY30-HEVNP-Cy5.5 voor fluorescerende detectie beide in vitro (Figuur 5) en in vivo4.

Virusachtige deeltjes (experimentele) zijn gebruikt als nanocarriers weergeven van buitenlandse epitopes en/of leveren van kleine moleculen in de opsporing en behandeling van verschillende ziekten. Deze toepassing is afhankelijk van genetische modificatie, zelf-assemblage en cysteïne vervoeging te vervullen van de tumor-targeting toepassing van recombinant experimentele. vergeleken met genetische wijziging alleen, chemische vervoeging van buitenlandse peptiden aan experimentele biedt een belangrijk voordeel omdat het toestaat dat een aantal entiteiten, zoals synthetische peptides of oligosachariden bevatten, te worden op het oppervlak van experimentele geconjugeerd in een gedifferentieerde en flexibele wijze zonder wijziging van de VLP-vergadering.
We laten hier zien hoe met het hepatitis E virus nanoparticle (HEVNP), een modulair theranostic capsule, als een multifunctionele levering "vervoerder". Functies van HEVNPs omvatten weefsel-targeting, imaging en therapeutische levering. Gebaseerd op de reeds lang gevestigde structurele onderzoek van HEVNP, werden de structureel onafhankelijk en oppervlakte-blootgesteld residuen geselecteerd voor de vervanging van cysteïne als conjugatie sites voor maleimide-linked chemische groepen via thiol-selectieve verbanden. Een specifieke cysteïne-gewijzigd HEVNP (Cys ter vervanging van de asparagine op 573 aa HEVNP - 573C) was geconjugeerd met een borst kanker cel-specifieke ligand, LXY30 en voorzien van nabij-infrarood (NIR) fluorescentie kleurstof (Cy5.5), waardoor de tumor-gerichte HEVNPs als doeltreffende diagnostische capsules (LXY30-HEVNP-Cy5.5). Soortgelijke engineering strategieën kunnen worden toegepast met andere macromoleculaire complexen met bekende atomaire structuren te verkennen mogelijke toepassingen in theranostic levering.

Verwant aan HEV-experimentele, alle Cys gemodificeerde HEVNPs gevormd oplosbare icosahedrale capsids en deed niet aggregaat in oplossing tijdens productie of zuivering. Voor en na-voor-stapmodus maleimide-Biotine conjugatie, elk van de Cys bewerkt waren HEVNPs niet te onderscheiden van HEV-experimentele in de negatieve vlek EM (Figuur 2).
