In questo studio, descriviamo un in vitro, non invasivo e privo di etichetta metodo usando QPI per caratterizzare quantitativamente i fenotipi angiogenici e non-angiogeniche delle cellule di osteosarcoma umano. Più parametri cellulari sono stati analizzati simultaneamente da questo metodo integrato, ad alta produttività, tra cui area di cella, spessore della cella, volume delle cellule, il tasso di proliferazione, raddoppiando il tempo, immediatezza di migrazione, velocità di motilità, migrazione e motilità...

Uno dei primi punti di controllo nello sviluppo e nella progressione di un tumore solido è l'acquisizione del fenotipo angiogenico, un marchio di garanzia di cancro. Questa progressione comporta tutta una serie di processi biochimici e molecolari1,2,3. Una sfida tecnica nello studio di questo passaggio chiave nella progressione del tumore è la mancanza di strumenti per continuamente e quantitativamente caratterizzare e distinguere tra fenotipi angiogenici e non-angiogeniche delle cellule tumorali dal vivo in modo imparziale. Le analisi tradizionali utilizzate per indagare i comportamenti cellulari delle cellule angiogenici e non-angiogenici solitamente richiedono strumenti e reagenti costosi, ad esempio, proliferazione e migrazione di cellule dosaggi4,5, 6,7,8,9,10,11,12,13,14 o complementari in vivo valutazioni4,5,6,8,15,16, come pure che richiedono ad alta intensità e significativa esperienza consumo di tempo e manodopera.
Recentemente, la fase quantitativa imaging (QPI) è emerso come una nuova tecnica che consente la valutazione di time-lapse e privo di etichettatura di una varietà di cellula morfologia e comportamento parametri17,18,19, 20 , 21 , 22. a differenza di microscopia ottica convenzionale, QPI quantifica variazioni di pixel per pixel di sfasamento dopo luce passa attraverso un oggetto ottico e ricostruisce un testamento olografo con spessore ottico convertito e volume, consentendo in tal modo la diretta analisi di cellule vive e le seguenti caratteristiche: (1) quantitativa imaging, imaging (2) non-invasivo e time-lapse, imaging (3) privo di etichetta e (4) simultanea multi-parametro imaging. Queste caratteristiche rendono QPI un potente strumento per valutare e comprendere i processi patologici a livello cellulare.
In un recente studio, abbiamo utilizzato QPI per quantitativamente caratterizzare e distinguere tra angiogenici KHOS-A e non-angiogenici KHOS-N fenotipi di cellule di osteosarcoma umano in maniera sistematica e quantitativa, combinando analisi della morfologia delle cellule, proliferazione e la motilità23. Utilizzando software di analisi di immagine, un pannello di cella morfologica e comportamento parametri quantitativamente sono stati confrontati fra le cellule di osteosarcoma umano angiogenici e non-angiogenici e sono state identificate cinque differenze di caratteristiche tra questi due fenotipi. Questo approccio novello fornisce una piattaforma integrata e quantitativa per valutare una gamma di caratteristiche cellulari biologicamente rilevanti.

L'acquisizione del fenotipo angiogenico è una componente essenziale della fuga dalla dormienza del tumore. Anche se parecchi saggi classici in vitro (ad esempio, proliferazione, migrazione e altri) e in vivo modelli sono stati sviluppati per studiare e caratterizzare fenotipi cellulari angiogenici e non-angiogenici, questi metodi sono tempo lavoro intensivo e spesso richiedono costosi reagenti e strumenti, nonché significative esperienze.
