Enkele voorzorgsmaatregelen zijn nodig voor de bovenstaande protocollen om ervoor te zorgen consequent en kwantitatieve resultaten. Eerst, mHTT in beide breuken spontaan zal aggregaten vormen na verloop van tijd op meerdere bevriezen dooi cycli, met name bij een hoge concentratie. Het is dus cruciaal voor aliquots van het eiwit preps bevriezen en ontdooien alleen het benodigde volume voordat u de test uitvoert zoals beschreven in het bovenstaande protocol. De verstoring van eiwit kwaliteitscontrole netwerken, zodat het juiste vouwen en aantasting van de cellulaire eiwitten is waarschijnlijk centraal in de pathologie in de ziekte van Alzheimer (AD), de ziekte van Parkinson (PD), de ziekte van Huntington (HD), en andere "eiwit misfolding" stoornissen2,3. Een gedetailleerd inzicht in de proteostasis-netwerkonderdelen en hun bijdragen aan de pathologie zijn daarom cruciaal voor de ontwikkeling van verbeterde therapeutische interventies. HD wordt veroorzaakt door de abnormale groei van een herhaling van het CAG binnen het HD-gen wat resulteert in een uitgebreid stuk van polyglutamines (polyQ) in het eiwit huntingtin (HTT)4. Een consistente pathologische functie van HD pathogenese is de resulterende misfolding, accumulatie en aggregatie van HTT in gebieden van de hersenen en perifere weefsels, waarvan is aangetoond te bemoeien op verschillende manieren met de normale cel homeostase en functie 5 , 6. terwijl HTT overal wordt uitgedrukt, middellange Maxomys neuronen in het striatum zijn selectief kwetsbare en meest openlijk waarin dit probleem optreedt met opmerkelijke corticale atrofie ook geassocieerd met HD pathogenese.
De vorming van aggregaten van HTT in de hersenen van HD patiënten en diermodellen heeft meestal geserveerd als een proxy-marker van progressie van de ziekte en dominant-negatieve winst van functie voor mHTT7. Hoewel de precieze mechanismen door welke proteïne misfolding en aggregatie aan mHTT-geïnduceerde toxiciteit bijdragen kunnen onduidelijk blijven, is de vorming van deze insluitsels in de hersenen van HD patiënten en verschillende diermodellen een invariant en onvermijdelijke functie. Insluitsels lijken te bestaan grotendeels uit geaccumuleerde HTT fragmenten met de N-terminal uitgebreid polyQ, die aangeeft dat proteolyse en verwerking van full-length HTT evenals alternatieve splicing8,9 kan spelen een belangrijke rol in de pathogenese van HD. N-terminale HTT fragmenten kan vormen een pathologische vorm van HTT die kan snel aggregaat, ervoor, en versnellen of propageren de aggregatie proces10.
De aanwezigheid van deze insluitsels doet echter niet noodzakelijkerwijs met HTT-geïnduceerde toxiciteit of cel dood11correleren. HTT wordt voorgesteld te ondergaan het proces van een samenvoeging van een oplosbare monomeer door oplosbare oligomere soorten en β-sheet fibrillen tot onoplosbare aggregaten en insluitsels12. Oplossen van deze verschillende eiwit soorten via standaard biochemische tests is een uitdaging op het gebied als gevolg van hun stabiliteit in lage-wasmiddel lysis-buffermengsel en moeilijkheden in het visualiseren met behulp van standaard biochemische tests. Methodologische overwegingen zijn dus cruciaal voor het opsporen van de mate en het patroon van opgebouwde en geaggregeerde mHTT.
Het hier gepresenteerde protocol biedt een methode om te visualiseren van verschillende tussenproducten van HTT, specifiek de vorming van een onoplosbare HMW HTT-soorten, die lijkt te nauw track met HD pathogenese en ziekte progressie1,13 ,14. Zijnde kundig voor oplossen en het bijhouden van meerdere soorten mHTT biedt onderzoekers een biochemische instrument om te studeren van de pathogenese van de ziekte en het evalueren van potentiële therapeutische interventies door hun modulatie en het effect op de pathogenese van de ziekte.

De accumulatie van misfolded eiwitten staat centraal in de pathologie in de ziekte van Huntington (HD) en vele andere neurodegeneratieve aandoeningen. Met name is een pathologische spil van HD de afwijkende accumulatie van mutant HTT (mHTT) eiwit in ultrahoog moleculair gewicht complexen en intracellulaire opneming organen samengesteld van fragmenten en andere eiwitten. Conventionele methodes voor het meten en begrijpen dat de bijdragen van verschillende vormen van mHTT-bevattende aggregaten zijn fluorescentie microscopie, westelijke vlekkenanalyse en filter overlappen testen.
Meeste van deze methoden zijn echter conformatie specifiek, en daarom kan niet kunt oplossen door de volledige staat van mHTT eiwit flux vanwege de complexe aard van de totale oplosbaarheid en resolutie.
Voor de identificatie van geaggregeerde mHTT verschillende complexen en aangepaste formulieren is scheiding en solubilisatie van de cellulaire aggregaten en fragmenten verplicht. Hier beschrijven we een methode om te isoleren en visualiseren van oplosbare mHTT monomeren, oligomeren, fragmenten en een onoplosbare ultrahoog moleculair gewicht (HMW) geaccumuleerde mHTT soorten. HMW mHTT bijgehouden met de progressie van de ziekte, komt overeen met muis gedrag uitlezingen en heeft geweest beneficiair gemoduleerd door bepaalde therapeutische interventies1. Deze aanpak kan worden gebruikt met muis hersenen en perifere weefsels celkweek maar kan worden aangepast aan andere modelsystemen of ziekte contexten.

Resolutie van oplosbare en onoplosbare cel lysates na fractionering kan worden opgespoord met behulp van westerse analyse en filter retardatie assays (Figuur 2).
