Terwijl de passieve, niet-elektroforetische extractiemethoden werkzaam verbeterd PACT aanzienlijk de consistentie bereikt met vorige weefsel clearing methoden zoals duidelijkheid2,,3,,4,7 , 8, de techniek draagt nog steeds een aantal tekortkomingen, de meest urgente van die de lengte van de benodigde tijd is voor het bereiken van maximale weefsel duidelijkheid...

Een fundamentele doelstelling van wetenschappelijke en klinische onderzoek omvat het bereiken van een volledig inzicht in de structuur van het orgel en functie; echter dient de buitengewoon complexe aard van zoogdieren organen vaak als een barrière voor het volledig bereiken van deze doelstelling1. DUIDELIJKHEID (duidelijk lipide-uitgewisseld Acrylamide-gekruist rigide Imaging-compatibele Tisssue-hYdrogel)2,3,4, waarbij de opbouw van een hybride acrylamide gebaseerde hydrogel uit intact weefsels, behaalt optische Goedkeuringvande allerlei organen, met inbegrip van de hersenen, lever en milt, met behoud van hun structurele integriteit5. DUIDELIJKHEID heeft dus niet alleen de visualisatie, maar ook de mogelijkheid om complexe cellulaire netwerken en weefsel morphologies zonder de noodzaak voor afdelen fijn te ontleden.
Met het oog op goedkeuring van het weefsel, hanteert duidelijkheid elektroforetische methodes te verwijderen van het vetgehalte van het monster bij de hand. Terwijl duidelijkheid is opgemerkt voor de productie van fysiek stabiele weefsel-hydrogel hybriden, studies hebben aangetoond dat het gebruik van elektroforetische weefsel clearing (ETC) methoden variabele resultaten in termen van kwaliteit van weefsel levert, met inbegrip van bruinen, epitoop schade, en eiwit verlies5,6. Om deze kwesties te behandelen, geweest gewijzigde protocollen zoals PACT (passieve duidelijkheid techniek), waarbij de enz behandeling wordt vervangen door een passieve, Ionische-wasmiddel op basis delipidation-techniek, ontwikkelde7,8,9. Ondanks het bereiken van een grotere samenhang van resultaten, vereist PACT echter meer tijd om te verkrijgen maximale klaring. Bovendien, geen van deze technieken hebben nog zijn toegepast bij het hele CNS-formulier of in grotere modellen, knaagdieren zoals ratten en cavia's.
De huidige studie tracht te stellen deze beperkingen door het voorstellen van nieuwe methodologieën, psPACT (proces-aparte PACT) en mPACT (gemodificeerde PACT), ter vergemakkelijking van de snelle goedkeuring van het hele CNS en inwendige organen in zowel muis en rat modellen10. Specifiek, psPACT verwerkt weefsels in 4% acrylamide en 0,25% VA-044 uitgebracht in twee afzonderlijke stappen tijdens hydrogel vorming; mPACT hoofdzakelijk omvat de dezelfde stappen, maar vormt een aanvulling op de clearing SDS gebaseerde oplossing met 0,5% α-thioglycerol als een belangrijk reagens. Beide technieken benutten de endogene systemische en hersen bloedsomloop systems om aanzienlijk minder tijd nodig voor de productie van optische klaring. Als een bewijs van beginsel tonen wij het gebruik van de confocal microscopie bloedvat patronen in de gewiste weefsels10analyseren.

novel passive clearing methods for the rapid production of optical transparency in whole cns tissue

Sinds de ontwikkeling van duidelijkheid, een bioelectrochemical clearing techniek die het mogelijk maakt voor toewijzing van de driedimensionale fenotype binnen transparante weefsels, een veelheid van nieuwe clearing methodologieën, met inbegrip van CUBIC (de weergave van de duidelijke, unobstructed hersenen cocktails en computationele analyse), schakelaar (systeembrede controle van interactie tijd) en kinetiek van chemische stoffen, kaart (vergrote analyse van het proteoom), PACT (passieve duidelijkheid techniek), zijn opgezet en verder uit te breiden de bestaande toolkit voor de microscopische analyse van biologische weefsels. De huidige studie is gericht op het verbeteren en optimaliseren van de oorspronkelijke PACT procedure voor een matrix van intact knaagdier weefsel, met inbegrip van de hele centrale zenuwstelsel (CNS), de nieren, de milt, en de hele muis embryo's. PsPACT (proces-aparte PACT) en mPACT (gemodificeerde PACT) genoemd, dienen deze nieuwe technieken als zeer doeltreffend middel om mapping cel circuits en visualiseren van subcellular structuren in intact normale en pathologische weefsels. In het volgende protocol bieden wij een gedetailleerde, stapsgewijze overzicht over het bereiken van maximale weefsel goedkeuring met minimale invasie van hun structurele integriteit via psPACT en mPACT.
