Schimmelinfecties zijn geworden een belangrijke medische zorg in de afgelopen decennia aanzienlijk is gestegen voornamelijk te wijten aan een stijging van het aantal immuungecompromitteerde personen zoals die in de behandeling van kanker en degenen die met HIV/AIDS leven of getransplanteerde organen1,2. Echter bijdragen een zeer beperkt scala van beschikbare antischimmel geneesmiddelen en het toenemende aantal verslagen over schimmel weerstand tegen hen aan de grote problemen met betrekking tot de therapeutiek van systemische mycoses3.
Een potentiële bron van nieuwe antischimmel compounds zijn antimicrobiële peptiden (ampère), kleine kationische peptiden geproduceerd door vele organismen als onderdeel van hun aangeboren immune reactie op infectie4. De screeningsmethode voor het testen van deze verbindingen tegen schimmel ziekteverwekkers is echter niet gestandaardiseerd. Verschillende procedures zijn gebruikt om de antischimmel activiteiten beoordelen van versterkers, soms voor de dezelfde model micro-organisme5,6,7. Deze verschillen en het gebrek aan detail in sommige protocollen bemoeilijken vergelijkingen tussen verbindingen en belemmert reproduceerbaarheid.
Unidirectioneel om te standaardiseren het testen van nieuwe drugkandidaten is richtsnoeren gebruikt om te definiëren antischimmel gevoeligheid in klinische instellingen, zoals de klinische en laboratorium Standards Institute (CLSI) M27-A3 richtsnoeren te volgen. Deze antischimmel gevoeligheid tests zijn echter te beperkend, en niet rekening overweging variatie in metabolisme over soorten, als zij slechts voor een paar selecteren agenten waren gevestigd. Bijvoorbeeld, doen ze niet rekening houden met de metabolische behoeften van niet-fermenteren-gisten.
Dit protocol staat de beoordeling van de activiteit van potentiële antischimmel verbindingen, en hier is geïmplementeerd voor het zoeken naar antischimmel peptiden. Het is gebaseerd op de Bouillon microdilution gevoeligheid test van de CLSI M27-A3-richtlijnen met betrekking tot wijzigingen die de screening van nieuwe verbindingen8,9te optimaliseren. Deze wijzigingen toestaan voor het gebruik van kleine hoeveelheden van stof, variaties in temperatuur of eerste entmateriaal en verschillende media voor optimale pre-test groei, terwijl de resultaten met het gebruik van referentie antischimmelmiddelen als besturingselementen te standaardiseren. Deze methode, met het gebruik van multi goed cultuur platen, maakt het mogelijk om snel en betrouwbaar scherm een groot aantal verbindingen.
Als gevolg van de inherente flexibiliteit, kan dit protocol worden gebruikt met verschillende chemische klassen van samenstellingen en tegen andere micro-organismen, met enkele aanpassingen.

broth microdilution in vitro screening: an easy and fast method to detect new antifungal compounds

Schimmelinfecties zijn een belangrijke medische voorwaarde geworden in de laatste decennia, maar het aantal beschikbare antischimmel geneesmiddelen is beperkt. In dit scenario is het zoeken naar nieuwe antischimmel geneesmiddelen nodig. Het protocol gemeld hier gegevens een methode om scherm peptiden voor hun schimmeldodende eigenschappen. Het is gebaseerd op de Bouillon microdilution gevoeligheid test van de Clinical and Laboratory Standards Institute (CLSI) M27-A3 richtsnoeren met wijzigingen aan het onderzoek van antimicrobiële peptiden als potentiële nieuwe antischimmelmiddelen. Dit protocol beschrijft een functionele test om te evalueren van de activiteiten van antischimmel verbindingen en kan eenvoudig worden aangepast om aan te passen van een bepaalde categorie van moleculen onder onderzoek. Aangezien de testen worden uitgevoerd in 96-wells-platen met behulp van kleine volumes, kan een grootschalig onderzoek worden uitgevoerd in een korte hoeveelheid tijd, vooral als uitgevoerd in een omgeving van automatisering. Deze procedure laat zien hoe een gestandaardiseerde en verstelbare klinische protocol het nastreven van de Bank-werk van nieuwe moleculen ter verbetering van de therapie van schimmelziekten kan helpen.

De MIC wordt gedefinieerd als het laagste antimicrobiële samengestelde concentratie dat volledig de groei van de zichtbare zwammen aan het eind van de incubatietijd remt.
