Tests de microdilution peuvent analyser l’activité antifongique potentielle d’un composé à l’aide de petites quantités de ce composé de la cible et en même temps le tester dans une gamme de concentrations. En conséquence, le présent protocole est recommandé dans un premier temps dans le dépistage de potentiels nouveaux composés antifongiques. Le protocole présenté ici est basé sur le protocole M27-A3, initialement conçu pour aider dans le choix du traitement antifongique dans les cliniques et peut s?...

Les infections fongiques sont devenus une préoccupation médicale importante ces dernières décennies, ayant considérablement augmenté principalement en raison d’une augmentation du nombre des individus immunodéprimés tels que ceux qui subissent le traitement du cancer et ceux qui vivent avec le VIH/sida ou transplanter des organes1,2. Cependant, un éventail très limité de médicaments antifongiques disponibles et le nombre croissant de rapports sur la résistance fongique à eux contribuent aux problèmes majeurs concernant la thérapeutique des mycoses systémiques3.
Une source potentielle de nouveaux composés antifongiques sont des peptides antimicrobiens (ampères), petits peptides cationiques produites par nombreux organismes dans le cadre de leur réponse immunitaire innée à l’infection4. Néanmoins, la méthode de dépistage pour tester ces composés contre les pathogènes fongiques n’est pas standardisée. Procédures différentes ont été utilisées pour évaluer l’activité antifongique des amplis, parfois pour le même modèle micro-organisme5,6,7. Ces différences et le manque de précision dans certains protocoles compliquent les comparaisons entre reproductibilité de composés et de paniers.
Normaliser les tests de nouveaux candidats-médicaments consiste à suivre les lignes directrices permettant de définir la sensibilité aux antifongique en milieu clinique, tels que la clinique et Laboratory Standards Institute (CLSI) M27-A3. Toutefois, ces tests de sensibilité antifongiques sont trop restrictives et ne prennent pas en variation de la contrepartie dans le métabolisme de toutes les espèces, telles qu’elles ont été établies uniquement pour quelques agents sélectionnés. Par exemple, ils ne tiennent pas compte des besoins métaboliques des levures fermentant le non.
Ce protocole permet l’évaluation de l’activité des éventuels composés antifongiques et est mis en place ici pour la recherche de peptides antifongiques. Il repose sur le test de sensibilité de microdilution bouillon des lignes directrices du CLSI M27-A3, avec les adaptations qui permettent d’optimiser le dépistage des nouveaux composés8,9. Ces modifications permettent l’utilisation de faibles quantités de composés, les variations de température ou initiale d’inoculum et différents médias pour une croissance optimale pré-test, tout en standardisant les résultats avec l’utilisation d’antifongiques de référence sous forme de contrôles. Cette méthode, avec l’utilisation de plaques multipuits culture, rend possible un grand nombre de composés de l’écran rapidement et sûrement.
En raison de sa souplesse inhérente, ce protocole peut être utilisé avec différentes classes chimiques des composés et contre d’autres micro-organismes, avec quelques adaptations.

Les infections fongiques sont devenues une condition médicale importante dans les dernières décennies, mais le nombre de médicaments antifongiques disponibles est limité. Dans ce scénario, la recherche de nouveaux médicaments antifongiques est nécessaire. Le protocole rapporté ici en détail une méthode aux peptides de l’écran pour leurs propriétés antifongiques. Il repose sur le test de sensibilité de microdilution bouillon des lignes directrices du Clinical and Laboratory Standards Institute (CLSI) M27-A3, avec des modifications pour répondre à la recherche de peptides antimicrobiens comme de potentiels nouveaux antifongiques. Ce protocole décrit une analyse fonctionnelle afin d’évaluer l’activité des composés antifongiques et peut être facilement modifié pour convenir à une quelconque classe de molécules incriminés. Comme les tests sont effectués en plaques 96 puits à l’aide de petits volumes, un dépistage à grande échelle peut être complété en un court laps de temps, surtout si réalisée dans un cadre de l’automatisation. Cette procédure illustre comment un protocole clinique standardisé et réglable peut aider à la poursuite de banc-travaux de nouvelles molécules pour améliorer le traitement des maladies cryptogamiques.

Le MIC est définie comme la plus faible concentration de composés antimicrobiens qui inhibe complètement la croissance fongique visible à la fin de la période d’incubation.
